苜蓿下胚轴培养特性研究

摘    要:苜蓿无菌苗下胚轴在附加2mg/L 2,4-D和0.1~0.9mg/L KT的SH培养基中诱导出愈伤组织,愈伤组织再在附加1.0mg/L IAA和1.0-2mg/L 6BA的SH培养基中分化;其中诱导愈伤率为100%,阿尔刚金苜蓿分化率在35.4%~43.7%时,随着KT的浓度增加,愈伤组织的出愈率反而延迟;而在分化过程中发现分化时不宜采用浓度过高的激素组合.0.1%Hgcl2消毒时间对苜蓿种子萌发效果的影响较大,在考虑降低污染率和提高存活率时,阿尔刚金种子以消毒10min最佳,紫花苜蓿以20min最佳.

Abstract:Alfalfa no vaccine in hypocotyl additional 2 mg / 1 2,4-D and 0.1 to 0.9 mg/ 1 KT SH medium induced callus, then callus additional 1.0 mg / 1 IAA and 1.0-2 mg/l 6BA SH medium differentiation; which callus induction rate was 100%, al-steel division of the alfalfa in 35.4%- 43.7%, with the increase in the concentration of KT, the callus rate instead of the more delay; In the course of differentiation, differentiation found inappropriate use of excessive concentration of hormone combination. 0.1% Hgcl2 disinfection alfalfa seed germination time on the effect of large, in considering reducing pollution and improving survival rates, the al-gang seeds to disinfect 10 minutes best, Alfalfa to 20 minutes best.
