柿树离体繁殖研究

摘    要:以成年柿树休眠芽为外植体,研究了影响柿树离体繁殖的有关因素。结果表明:附加ZT1~2mg/L、IAA0.05~0.1mg/L、叶酸1mg/L、D-泛酸钙1mg/L、LH100mg/L、PVP500mg/L的改良MS(KNO3和NH4NO3减半)培养基对柿芽的增殖和生长最为适宜。在继代培养中,适当缩短每代培养时间可加快新梢的增殖速度。IAA诱导生根的效果优于IBA,用生长素溶液浸新梢基部后转入1/2MS中对生根更有利。在较低温度下,以腐殖土作基质,试管苗的移栽成活率可达68.6%。

Abstract:In current paper, taken dormant bud of mature trees of persimmon as the explants,some factors affecting in vitro propagation of persimmon were studied. The results showed that modified MS (1/2 KNO3, and 1/2NH4NO3, ) medium supplemented with ZT1 ~21mg/L, IAA0.05 ~ 0. 101mg/L, folic acid 11mg/L, calcium pantothenatelmg/L, lactoprotein hydrolysate 1001mg/L and PVP5001mg/L were the most suitable to the proliferation and the growth of persimmon buds. In the proceeding of subculture,shortening times of subculture could accelerate the proliferation rate of shot. The inducing rooting activity of IAA was better than that of IBA. Dipping the basis of shoots in the auxins solution before changing it into 1/2MS medium was better than adding auxins into the medium directly for rooting. In the lower temperature condition,Using humus soil as trasplant medium, the survival rateof plantlet transplanted could be 68, 6%.
