农杆菌介导法遗传转化巴西橡胶树转化条件的研究

摘    要:用包含GAI基因的EHA105农杆菌,对巴西橡胶树海垦2花药愈伤组织进行侵染,对多项转化条件进行了梯度实验。结果表明选择培养30d的橡胶花药愈伤组织,农杆菌菌液浓度OD600为0.6时侵染3min,20℃共培养2d,Cef浓度为200mg/L进行抑菌培养是比较合理的转化条件。

Study on the Conditions of the Agrobacterium -mediated Genetic Transformation for Hevea Brasiliensis

Abstract:The Agrobacterium containing GAI gene was used to infect the anther callus of Haiken 2 - a variety of Hevea brasiliensis to get a transgenie plant. The gradient experiments of several transformation conditions were done. The results showed that: the anther callus cultured for 30 days, 0.6 OD600 value of the Agrobacterium solution, 3min infection time, co -culture for 2 days at 20℃ and 200 mg/L Cef concentration for Agrobacterium inhibition culture were optimal transformation conditions.
