山药食用部分石蜡切片的制作方法和鉴定研究

摘    要:通过设置不同处理,对山药地下食用部分进行脱水、透明、浸蜡等主要影响因素进行试验,得到适合的石蜡切片制作方法。结果表明,合适的脱水时间为每阶段2h,透明时间为每阶段8h,浸蜡时间为每阶段10h。对材料显微结构的观察表明其维管束排列方式、维管束内部结构及分化特征均符合单子叶植物茎的一般特征,判定山药地下食用部分其本质是茎而不是根。

Abstract:A suitable method for paraffin section of underground edible parts of yam was obtained by testing diverse factors such as dehydration,transparent,and paraffin in different treatments.Results showed that the proper time for dehydration,transparent,and paraffin in each stage,are 2h,8h,and 10h,respectively.Microscopic observation presented that the vascular bundle arrangement,internal structure and differentiation in the sample accorded with the general characteristics of monocots stem,and further identified that the underground edible part of yam is the stem rather than root.
