6-BA和2,4-D对黄独带芽茎段生长发育的影响

摘    要:【目的】探讨6-BA和2,4-D对黄独带芽茎段生长发育的影响,为黄独的产业化生产提供理论依据。【方法】采用植物组织培养的方法,将黄独带芽茎段(0.5~1.5 cm)接种至液体培养基进行单因子试验。【结果】黄独带芽茎段芽增殖的最佳培养基为MS;黄独带芽茎段芽增长的最佳培养基为MS+6-BA 1 mg/L+2,4-D 0.5 mg/L;黄独带芽茎段芽增粗的最佳培养基为MS+6-BA 2 mg/L+2,4-D 0.1 mg/L;黄独带芽茎段生根的最佳培养基为MS;黄独带芽茎段茎长增加的最佳培养基为MS+6-BA 2 mg/L+2,4-D 0.1 mg/L;黄独带芽茎段茎粗增加的最佳培养基为MS。【结论】黄独带芽茎段的芽增殖和生根最好不要使用6-BA与2,4-D组合,但黄独带芽茎段如在其他培养基诱导出新芽后,则可利用6-BA与2,4-D组合来促进芽的增长、增粗以及茎长的增加。

Abstract:[Objective]The effects of 6-BA and 2,4-D on growth and development of Dioscorea bulbifera L.budded stems was studied in order to utilize the technique in large scale production of Dioscorea bulbifera L.plantlets.[Method]Using plant tissue culture methods,D.bulbifera L.budded stems(0.5-1.5 cm) were inoculated into liquid culture media in single factor experiment.[Result]MS medium was best for bud multiplication,rooting,and increase in shoot diameter.The best medium for growth of regenerated buds and increase in bud diameter and shoot length of D.bulbifera L.were identified as MS+6-BA 1 mg/L+2,4-D 0.5 mg/L,MS+6-BA 2 mg/L+2,4-D 0.1 mg/L and MS+6-BA 2 mg/L+2,4-D 0.1 mg/L,respectively.[Conclusion]The hormone combination 6-BA and 2,4-D could be used for growth of regenerated buds and increase in bud diameter and shoot length of D.bulbifera L.
