坛紫菜rbcS及rbcL-rbcS基因间隔区的序列分析

摘    要:Rubisco是光合作用和光呼吸的关键酶,目前已成为高等植物中研究最深入的酶之一,而在低等藻类中关于该酶的相关研究却很少。以来自浙江和福建的6个坛紫菜(Porphyra haitanensis)无性系为研究对象,以江苏条斑紫菜(P.yezoensis)为参照,克隆了其Rubisco小亚基(rbcS)及大小亚基基因间隔区共516 bp的序列。序列分析结果表明,其中4个坛紫菜系(Hza、Hzf、Hpt、Hzz)的基因序列完全相同(以其作为标准),坛紫菜Hzb和Hzc与其均只有1个碱基差异,而条斑紫菜Yn55与其差别碱基为46个。将本实验的7条紫菜序列与GenBank数据库收录的17条紫菜Rubisco序列一起做系统分析,结果表明坛紫菜和条斑紫菜都分别归到了各自的类群中。以上结果说明,紫菜rbcS和rbcL-rbcS基因间隔区序列的同源性较高,可用于种以上水平的系统分析。

Abstract:Rubisco-1,5-bisphosphate carboxylase/oxylase(Rubisco) is the key enzyme in photosynthesis and photorespiration,which has become one of the most extensive studied enzymes in higher plants.However,related investigation of the enzyme in the algae is comparatively very few.In this study,six lines of laver Porphyra haitanensis used as materials,one line of P.yezoensis as control,their sequences of rbcS and rbcL-rbcS intergenic spacer region(516 bp) were cloned.Sequence analyses showed that 4 lines of P.haitanensis(Hza,Hzf,Hpt,Hzz) were identical(used as standard sequence),and 2 lines(Hzb,Hzc) were only one base different from the standard.P.yezoensis of Yn55 was found 46 bp distinct from the standard sequence of P.haitanensis.In the phylogenetic tree built on the total 24 sequences of Porphyra from this study and the GenBank databases,all lines of laver were classified into their respective cluster.The above results showed that rbcS sequence and rbcL-rbcS intergenic spacer region of the laver were highly homologous,so it was more suitable to confirm interspecific relationship rather than intraspecific determination
