新郑灰枣基因组DNA提取方法比较

摘    要:以新郑灰枣苗叶片为材料,采用CTAB方法提取叶片DNA,并对该方法进行改良。比较4种DNA提取方式,对提取结果进行紫外分光光度、电泳、酶切等方法的鉴定,结果表明,提取的灰枣叶片DNA得率约为0.370~0.530μg/mg,A260/A280约为1.90~2.00,A260/A230在2.00以上,分子量在23 kb以上,能够被EcoRⅠ,HindⅢ酶切。其中,第2种处理方法提取效果较好。

Abstract:Four methods,including CTAB method,were used to extract the genomic DNA from the leaves of Ziziphus jujube from Xinzheng county,Henan province.The prepared DNA samples were identified by ultraviolet spectrophotometer analysis,agarose gel electrophoresis,etc.The results showed that the DNA production rate were 0.370-0.530 μg per milligram of fresh leaves,A260/A280=1.90-2.00 and A260/A230>2.00.The molecular weight was more than 23 kb.The second method was the best for extraction of DNA from the leaves of Ziziphus jujube from Xinzheng.
