儿茶素EGCG对Caco-2细胞α-葡萄糖苷酶活性的抑制作用研究

摘    要:试验对Caco-2细胞膜上蔗糖酶和麦芽糖酶这两种α-葡萄糖苷酶活性进行不同培养时间检测,并以儿茶素单体EGCG作为抑制剂,探究其对Caco-2来源的蔗糖酶和麦芽糖酶的抑制效果。结果表明,Caco-2细胞培养18 d后膜表面具有较高活性的蔗糖酶和麦芽糖酶,可用于进行α-葡萄糖苷酶抑制剂筛选。儿茶素EGCG具有蔗糖酶和麦芽糖酶抑制活性,抑制IC50分别为31.08μg/m L和36.44μg/m L。

Abstract:This research used the Caco-2 cell monolayer model in vitro to simulate the α-glucosidase activity at different culture days. EGCG was used as an inhibitor to investigate the inhibition activities on sucrase and maltase in Caco-2 cells. The results illustrated that Caco-2 could be used to estimate α-glucosidase inhibitory activity after 18 days with high sucrase and maltase activities. EGCG showed inhibitory effect on sucrase and maltase in Caco-2 cells, with the IC50 values of 31.08μg/mL and 36.44μg/mL, respectively.
