摘 要: | 根据近等基因系分析原理,以大白菜细胞质雄性不育系CMS-2及其保持系B-2为材料.对大白菜细胞质雄性不育基因及其保持基因进行RAPD标记研究。引物S391在不育系中有特异性扩增,得到一条约700bp的特异片断;引物S425在保持系中有特异性扩增,得到一条约600bp的特异片,分别命名为S391-700bp、S425-600bp。采用Blastn进行核苷酸同源序列比较,结果发现S391-700bp存在一个开放阅读框架,这一开放阅读框架与植物叶绿体丝氨酸乙酰基转移酶DNA部分片段有很高的同源性;S425-600bp是新发现的一段DNA序列。根据测序结果设计特异引物,结果表明本研究所获得的两个标记片断都能确定其特异性。

Abstract: | According to the isogenic line analysis theory, RAPD analysis was undertaken in the cytoplasm male sterile(CMS) line CMS-2 and its maintainer line B-2 in Chinese cabbage. And get two special amplified fragment of S391-700bp and S425-600bp Separate in sterile line and its maintainer line.The results showed there is a open reading frame in S391-700bp and the identities is high between S391-700bp and chloroplast serine acetyltransferase gene;And the S425-600bp is a new DNA fragment. |
