摘    要:以葛花总异黄酮含量及回收率为考察指标,选用大孔树脂对葛花总异黄酮进行分离纯化。结果表明,X-5树脂对葛花总异黄酮有较好的吸附和解吸效果,最佳工艺条件为:洗脱剂为70%乙醇;洗脱剂用量为3倍树脂体积;流速为2 mL/min;葛花总异黄酮上样液的质量浓度为2 mg/mL,速率为2 mL/min;冲洗杂质用水体积为20 mL。试验表明,该法简单可行,分离效果好,经X-5处理后的葛花异黄酮含量可达54%。

Abstract:Based on the content and recovery rate of isoflavones, the method to extract total isoflavanoids from Flos puerariae using Macroporous resin was evaluated in the current work. The result showed that X - 5 type resin have better adsorption and desorption effect for total Gehua isoflavanoids. The best elution agent was 70% ethanol, whose best volume was three time as that of resin, and its flow velocity was 2 mIJmin. The eoncent of total Gehua isoflavanoids in the samples should be 2 mg/mL, and the velocity was 2mL/min. This method was simple and feasible with good separation effect. After purified by X-5, the extract of isoflavanoids could be up to 54%.
