摘    要:利用法国农业科学院-明尼苏达大学的辐射杂种克隆板(IMpRH),将猪细胞视黄醇结合蛋白基因2(RBP2)和细胞视黄酸结合蛋白基因2(CRABP2)分别定位在猪13、4号染色体上。利用RT-PCR方法,克隆到了CRABP2的CDS全长。利用半定量的RT-PCR方法,对RBP2和CRABP2基因在成年五指山猪12种不同组织(肺、骨骼肌、脾、心脏、胃、大肠、淋巴结、小肠、肝、大脑、肾、脂肪)中的组织表达谱进行了研究。在这两个基因中共检测到了4个单核苷酸多态位点。在RBP2-C117T多态位点,利用基于聚合酶链式反应的限制性片段长度多态性(PCR-RFLP)方法,对其在莱芜黑猪、五指山小型猪、贵州小型猪、广西巴马小型猪和通城猪群中的分布进行了研究。在通城猪群中,进行了不同基因型与性状间的关联分析,发现其不同的基因型与肌肉大理石纹评分、肌肉嫩度高度相关。在猪的生产和育种中,这一多态位点可能会成为有用的分子标记。

Abstract:The retinoid(vitamin A and its derivatives) plays a critical role in vision,growth,reproduction,cell differentiation,and embryonic development.Using the INRA IMpRH panel,porcine cellular retinol binding protein gene 2(RBP2) and cellular retinoic acid binding protein gene 2(CRABP2) were assigned to porcine chromosome 13 and 4,respectively.The entire coding sequence(CDS) of porcine CRABP2 was amplified using RT-PCR,and deduced amino acid sequence of porcine CRABP2 was compared to human corresponding protein.The mRNA distributions of the two genes in adult Wuzhishan miniature pig tissues(lung,skeletal muscle,spleen,heart,stomach,large intestine,lymph code,small intestine,liver,brain,kidney,and fat) were examined.A total of four SNPs were identified in porcine RBP2 and CRAPB2.Using PCR-RFLP method,the RBP2-C117T SNP locus was analyzed in the Laiwu,Wuzhishan,Guizhou,Bama pig breeds,and Tongcheng pig population.Significant associations were found between different genotypes of RBP2-C117T with marbling score(MS.) and muscle shear force(MSF) traits.This SNP may be used as genetic marker in pig breeding and production.
