生药防风不定芽分化影响因素的研究

摘    要:以防风愈伤组织为材料,研究了培养基、激素组合、光照、愈伤诱导周期和继代培养次数对愈伤组织不定芽分化的影响。结果表明:适合防风愈伤组织芽分化的最佳培养基为MS+NAA0.2+KT0.2+6-BA1.0;继代培养的周期为25d,继代培养2次防风不定芽的分化率较高。随着光照强度从5001x到20001x的增加,防风愈伤组织产生的芽数增加,丛生芽健壮,光照强度20001x最适合防风愈伤分化出不定芽。

Abstract:By using the Saposhnikovia divaricata callus as the test materials,we have researched the effects of the substrate、Hormone combination 、light,callus induction period and the number of subculture on callus adventitious bud differentiation.The result showed that the best substrate used bud polarization of culture the callus,it is MS+NAA0.2+KT0.2+6-BA 1.0;the suitable callus subculture period for the Saposhnikovia divaricata was 25 d,under two subcultures,the rate of callus adventitious bud differentiation was higher.As the light intensity increased from 5 001 x to 20 001 x,the shoots produced by the Saposhnikovia divaricata callus increased,the caespitosa shoots were robust,the light intensity 20 001 x was the best condition for the Saposhnikovia divaricata calli emergence.
