核桃AFLP银染技术体系的建立

摘    要:为了建立准确、高效、实用的核桃AFLP体系,应用于核桃品种鉴别、优异基因的分子标记,本试验针对核桃叶片中多糖、酚类等次生代谢物比较多的特点,采用改良CTAB法,通过添加抗氧化物质,得到了适合核桃AFLP分析的基因组DNA提取方法;在此基础上进行了AFLP体系中主要参数的优化研究,比较了两种银染方法的效果,并从64对选择性扩增引物中筛选出了18对适合核桃AFLP分析的引物,建立了能得到清晰指纹分析图像的核桃AFLP技术体系。

Abstract:To establish exact,highly effective and practical walnut AFLP system and molecular mark method applied to identification of walnut species and differentiation of genes,a genome DNA extraction method suitable for walnut AFLP analysis was developed.Due to presence of the substances such as polyhexose and hydroxybenzen in walnut leaves,this method was achieved by modifying CTAB method and addition of vC,PVP40,DIECA.AFLP system with clear image for fingerprint analysis has been established by optimization of parameters involved in the AFLP system and comparison of different silver staining methods and screening 18 pairs of primers suitable for walnut AFLP analysis from 64 pairs of primers for selective amplification.
