雄黄兰花蕾离体培养技术探讨

摘    要:以雄黄兰花蕾为外植体,以MS为基本培养基,通过添加不同浓度的BA、NAA等激素,探讨外源激素对其愈伤组织诱导、不定芽分化、增殖与不定根形成的影响,并建立了微繁体系;通过采取不同驯化炼苗措施,总结了一整套炼苗与栽培技术。试验表明:选用MS+6-BA2.0mg.l-1+NAA0.2 mg·l^-1培养基,其外植体诱导率达84.4%,不定芽诱导率达68.2%,选用MS+6-BA2.0mg·l^-1的培养基进行增殖培养,可得到优良的雄黄兰小种球,球茎增殖系数达4.9;在1/2MS培养基中培养15d,球茎生根率达98%;瓶苗常规驯化后选用国产泥炭土炼苗,成活率在90%以上;冬末春初繁殖的生根苗定植大田后当年开花。

Abstract:Exogenous effect of crocomia crocosmiflora flower buds external implantations of callus induced, adventitious bud differentiation, multiplication and form adventitious root were approached by MS culture media and different concentration BA, NAA. Micro-system of breeding was established. The technique of culture nursery stock was summarized by different culture measures. The results showed Crocomia crocosmiflora isolated culture induced rate was 84.4% by MS +6-BA 2.0 mg · l^-1 + NAA 0.2mg · l^-1. its adventitious buds induced rate was 68.2%, and form excellent small seed ball, and coefficient of corm multiplication was 4.9% by MS +6-BA 2.0 mg· l^-1. The strike rate of seed ball was 98% after 15 days in 1/2MS. Its bottle seedlings domesticated commonly that the rate of survival was over 90% which cultured in the local peaty. Seedling of strike will be flower planted field during the end of winter and first of spring multiply.
