摘    要:重叠延伸PCR技术是一种通过寡聚核苷酸链之间重叠的部分互相搭桥、互为模板进行PCR扩增,从而获得目的DNA基因片段的方法。该技术在目的基因的人工合成及外源蛋白在宿主中的高效表达等方面显示出良好的应用前景。综述了重叠延伸PCR技术的原理、反应条件的优化及其应用。

Abstract:Gene splicing by overlap extension is an approach for recombining DNA molecules at precise junctions irrespective of nucleotide sequences at the recombination site.The primers are designed so that the ends of the products contain complementary sequences.When the two products are mixed,de-natured,and reannealed,the strands having the matching sequences at their 3’ends overlap and act as primers for each other.This simple and widely ap-plicable approach has great advantages over other recombinant DNA techniques in synthesis of gene.This article summarized the principle,reaction condi-tions,and applications of the SOE-PCR.
