口蹄疫病毒R株致弱前后的基因变异研究

摘    要:为研究口蹄疫病毒(FMDV)致弱前后基因组变化的关系,本研究根据GenBank上发表的口蹄疫病毒全基因序列数,设计了8对引物,采用RT-PCR方法分别扩增出FMDVR株及其致弱毒株(R304)编码区的8段基因,将各片段克隆至pMD18-T载体,并进行序列测定。比较和分析编码区各个基因(LP、VP4、VP2、VP3、VP1、2A、2B、2C、3A、3B、3C、3D)的核苷酸序列和氨基酸序列的变异。结果表明,R株和R304株基因组区全长均为6966nt,编码2322个氨基酸,致弱后共有110个核苷酸发生变化,编码氨基酸有32个发生变化;其中3A基因的氨基酸序列变化最大,其次为LP和VP1,而2A和2C的氨基酸未发生变异。该研究对口蹄疫病毒的抗原和毒力研究有重要的参考价值。

Abstract:In this study,eight DNA fragments covered the full elngth genome of Foot-and-mouth disease virus R strain and its attenuated variant were amplified by RT-PCR using specific primers. Each fragment was cloned to pMD18-T vector and sequenced. The results showed that the genome of R strain and the attenuated strain shared the same ORF of 6 966 nt which encoded for 2322 amino acid.
