草菇子实体超氧化物歧化酶的检测

摘    要:采用聚丙烯酰胺垂直平板凝胶电泳法测定草菇蛋期至成熟期子实体不同部位超氧化物歧化酶(SOD)。结果表明蛋期子实体有1~2条酶带,伸长期和成熟期有2~4条酶带。菌柄是子实体衰老历程中 SOD 同工酶酶谱变化最敏感的部位。还观察了不同期子实体及其提取液于4℃低温贮藏4天后 SOD 同工酶的变化,并对草菇衰老机制及其与贮藏过程中 SOD 同工酶变化的关系进行了探讨。

Abstract:A horizontal polyacrylamide slab gel electrophoresis analysis on the superoxide dismutase (SOD) isozymes in the different parts of the fruit- body during egg stage up to mature stage.The results showed that there were 1-2 SOD isozyme bands in the egg stage,2-4 bands in the elon- gating and mature stages.The SOD isozymes of the stipe seem to be the most sensitive among the changeable ones in the senescence process of fruitbodies. Changes of SOD isozymes were observed during storage.Determinati- ons of the fruitbodies at different stage and their extracted solutions of the enzyme at 4℃ for 4 dayswere made.The possible machanism of senescence and its relationship to the SOD isozyme changes during storage were discussed. It is speculated that this work may be of value in the selection of sto- table straw mushrooms and their cultivaton
