彩色马蹄莲“柠檬”组织培养研究

摘    要:以彩色马蹄莲品种“柠檬”的块茎为外植体,研究不同的消毒方法和激素水平对彩色马蹄莲离体繁殖的影响,并筛选适合彩色马蹄莲试管苗生长的培养基。结果表明,在初代试验中对彩色马蹄莲的块茎消毒以0.2%升汞处理15 min为宜;MS+1.0-2.0 mg/L6-BA+0.1-0.2 mg/L NAA为适宜的愈伤组织诱导培养基;在继代培养基MS+0.5 mg/L6-BA+0.1 mg/L NAA上,愈伤组织能够分化出状态良好的丛生芽并继续增殖;在培养基MS+0.1mg/L IBA上,生根率达100%,试管苗生长健壮,根系良好。

Abstract:In this tissue culture experiment, bulbs of Zantedeschia hybrida ' Ning Meng' were used as explants. The results showed that the best time of 0. 2% HgCl2 disinfectant was 15 min. and the best calli induction medium was MS + 1.0 -2.0 mg/L 6-BA + 0. 1 -0. 2 mg/L NAA. The calli would grow into cluster buds in the medium of MS + 0. 5 mg/L 6-BA +0. 1 mg/L NAA, and the buds grew well with new callus proliferation. The rooting rate was 100% in the medium of MS +0. 1 mg/L IBA.
