芝麻种质资源RAPD分析及其遗传多样性

摘    要:利用RAPD分子标记方法,选取60个随机引物,对从我国保存的芝麻种质资源中接续取的19个种质进行遗传多样性分析,选出扩增效果好的14个多态性引物,并用于统计分析,14个引物共扩增出142条带,其中多态性带61条,占43%,平均每个引物有4.36条,变化范围2~7条,依据聚类分析结果可将19个供试材料分为4大类,地理来源较远的国外种质与我国本地种质之间遗传差民较大.利用RAPD标记技术在基因水平上分析芝麻种质资源遗传多样性完全可行.

Abstract:RAPD technique with 60 random primers was used to detect the genetic diversity of 19 sesame germplasms selected from a great quantity of sesame resources in China. 14 polymorphic primers were selected for statistic analyses. 142 RAPD markers were obtained and 43% of them were polymorphic. Each polymorphic primer can amply 4.36 bands on average with rang from 2 to 7 bands. The 19 sesame germplasms could be divided into four groups according to dendrogram of cluster. The largest genetic difference was found between overseas germplasms and germplasms from China. It is feasible to evaluate genetic diversity of sesame germplasms by using RAPD technique.
