家蚕五龄后部丝腺蛋白质构成与茧层量的关系

摘    要:家蚕 5龄期是后部丝腺细胞大量合成并分泌蚕丝主要成分丝素蛋白的时期 ,对这一时期后部丝腺细胞的蛋白质构成进行研究 ,有利于发现丝腺细胞分泌丝素蛋白有关的功能蛋白质 ,阐明蚕茧高产的机理。采用蛋白质双向电泳和图像分析技术 ,研究发现家蚕同一品种的不同个体 ,或同一个体不同部位的后部丝腺细胞蛋白质的构成基本一致。该结果反映了家蚕同一品种不同个体后部丝腺细胞的新陈代谢水平基本没有差异 ,家蚕品种个体间茧层量差异 ,可能是个体生长发育和后部丝腺细胞数的不同等原因造成 ,而与每一个体后部丝腺细胞的新陈代谢关系不显著。该结果也说明在目前双向电泳技术和染色技术的条件下进行家蚕后部丝腺蛋白质组研究时 ,从家蚕同一品种的任一个体、任一部位提取后部丝腺细胞的蛋白质样品 ,所得的蛋白质双向电泳结果基本相同

Abstract:The fifth instar of silkworm is the stage when the cells of posterior silkgland greatly synthesize and secrete fibroin protein, the main component of silk. The protein composition of posterior silkgland in this stage is studied so that the related functional proteins that occur during the secretion of fibroin protein by posterior silkgland will be found and the high production mechanism of cocoon will be elucidated. It was found that protein composition of posterior silkglands from different individuals of the same race and different parts of posterior silkglands from the same individual were almost the same by using two-dimensional electrophoresis of protein and image analysis system. The results suggested that there was little difference among metabolic level of individuals in the same race.Therefore, the difference of cocoon shell weight among individuals in the same race was probably induced by difference of development and growth among individuals, cell number of posterior silkgland and so on. Howerer, the relation between cocoon shell weight among individuals in the same race and metabolism of posterior silkgland in different individuals in the same race was no significantly difference. The protein samples extracted from any individual or any part of posterior silkgland in the same race had no influence on the proteome results by two-dimensional electrophoresis of protein and stain techniques at present.
