伞滑刃线虫属 4个种rDNA的PCR扩增

摘    要: 从161份病松树样本中分离、鉴定出拟松材线虫(Bursaphelenchus mucronatus)、霍夫曼伞滑刃线虫(B.hofmanni)和赫列尼库斯伞滑刃线虫(B.hellenicus)3个种,其中后二者是中国新记录种,华山松和云南松分别是这两个种的世界新寄主记录。本试验以松材线虫(B.xylophilus)为对照,用伞滑刃属线虫rDNA相关引物对这4种伞滑刃线虫进行PCR扩增,进一步印证形态鉴定的可靠性。试验结果表明,霍夫曼伞滑刃线虫、赫列尼库斯伞滑刃线虫与松材线虫、拟松材线虫,均扩增到约800 bp的分子片段,为霍夫曼伞滑刃线虫和赫列尼库斯伞滑刃线虫的鉴定从分子生物学角度提供了一定依据。

Abstract:From Dec. 1999 to Dec. 2001, Bursaphelenchus mucronatus, B.hofmanni and B. hellenicus were isolated and identified from 161 infected pine samples of 34 counties/cities in Yunnan pine forest areas. B.hofmanni and B.hellenicus were new records of Bursaphelenchus in China according to their morphologic characteristics, while Pinus armandii and P.yunnanensis were their new hosts recorded in the world. Molecular technique was applied to further verify the reliability of morphologic identification and Bursaphelenchus nematode rDNA primer was used for the PCR amplification with B.xylophilus as the control. The results showed that a 800 bp fragment was amplified in all four species, which provided the molecular evidence for the identification of B.hofmanni and B.hellenicus.
