真核翻译延伸因子(eukaryotic translation elongation factor,eEFs)是一种重要的多功能调控蛋白,eEF1β是eEF1的组成部分,在蛋白质生物合成过程中发挥着重要的作用。本文通过RT-PCR扩增克隆小麦(Triticum aestivum L.)的eEF1β基因,并命名为TaeEF1β。氨基酸同源性分析发现,TaeEF1β具有高度保守性,且其保守结构域位于137~226 aa处。qRT-PCR结果表明,中国小麦花叶病毒(Chinese wheat mosaic virus,CWMV)侵染小麦植株后,可以诱导TaeEF1β基因转录水平的上调表达。另外,本文也进一步分析了TaeEF1β基因在小麦根、茎、叶的表达水平和CWMV侵染不同时间点的表达情况。 相似文献

In accordance with the deficient elongation of the Stretching-bending Rectifier, as well as the severe wear and slipping of V-belt , the driving system velocity ratio of rectifier and the calculation formula of the theoretical elongation are deduced, which are based on the analysis of its structure and reeling principle. The carrying capacity of the driving system is also analyzed and it reaches the main reason for the elongation of the stretching-bending rectifier that cannot reach request, which is caused by the step less torsion of fixed output of the continuously variable transmission. The improvement measure which can be achieved by increasing the stiffness of the spring to increase the output torsion of the continuously variable transmission is proposed. Short basal internodes are important for lodging resistance of rice(Oryza sativa L.).Several canopy indices affect the elongation of basal internodes,but uncertainty as to the key factors determining elongation of basal internodes persists.The objectives of this study were(1)to identify key factors affecting the elongation of basal internodes and(2)to establish a quantitative relationship between basal internode length and canopy indices.An inbred rice cultivar,Yinjingruanzhan,was grown in two split-plot field experiments with three N rates(0,75,and 150 kg N ha−1 in early season and 0,90,and 180 kg N ha−1 in late season)as main plots,three seedling densities(16.7,75.0,and 187.5 seedlings m−2)as subplots,and three replications in the 2015 early and late seasons in Guangzhou,China.Light intensity at base of canopy(Lb),light quality as determined from red/far-red light ratio(R/FR),light transmission ratio(LTR),leaf area index(LAI),leaf N concentration(NLV)and final length of second internode(counted from soil surface upward)(FIL)were recorded.Higher N rate and seedling density resulted in significantly longer FIL.FIL was negatively correlated with Lb,LTR,and R/FR(P<0.01)and positively correlated with LAI(P<0.01),but not correlated with NLV(P>0.05).Stepwise linear regression analysis showed that FIL was strongly associated with Lb and LAI(R2=0.82).Heavy N application to pot-grown rice at the beginning of first internode elongation did not change FIL.We conclude that FIL is determined mainly by Lb and LAI at jointing stage.NLV has no direct effect on the elongation of basal internodes.N application indirectly affects FIL by changing LAI and light conditions in the rice canopy.Reducing LAI and improving canopy light transmission at jointing stage can shorten the basal internodes and increase the lodging resistance of rice. 相似文献

Brown spot needle blight (BSNB), a disease of pine trees caused by the fungus Lecanosticta acicola, has been known in Slovenia since 2008 and in Croatia since 1975. Recent outbreaks in Slovenia prompted this study to compare L. acicola populations in these two neighbouring European countries. Sixty-nine isolates collected from three pine species (Pinus mugo, P. halepensis and P. nigra) were used to determine the phylogenetic relationships, genetic structure, and reproductive strategy of the pathogen. EF1-α sequences showed that Slovenian and Croatian isolates share a common ancestry with individuals from central and northern Europe. Population structure analysis revealed four distinct population clusters of L. acicola in these two countries, generally corresponding to their respective geographic location and host. An unequal ratio of mating types and a low overall genetic diversity in the population indicated a strong influence of asexual reproduction. Although some of the oldest recorded European occurrences of BSNB are from Croatia, this study provided no evidence that the population studied in Croatia was the source of the sampled outbreaks in Slovenia. The aim of this work was to study the fatty acid (FA) bioconversion ability in Eurasian perch fed with diets differing in their polyunsaturated fatty acids (PUFA) from n‐3 and n‐6 series content at two development stages: adults in exogenous vitellogenesis, and juveniles during the on‐growing phase. Duplicate groups of adults and juveniles were fed for 12 weeks with four diets: D1 and D2, two diets prepared with fish oil partially or totally as the lipid source, and so containing long‐chain PUFA (LC‐PUFA). Those two diets differed by their n‐3/n‐6 FA dietary ratio (0.2 and 7.0, respectively), D1 being characterized by a high n‐6 LC‐PUFA level, while D2 had a high level of n‐3 LC‐PUFA. D3 and D4 were constituted only with vegetable oils, and were therefore devoid of LC‐PUFA. D3 was characterized by a high level of 18:2 n‐6 (n‐6/n‐3 ratio of 0.3), while D4 was characterized by a high level of 18:3 n‐3 (n‐3/n‐6 ratio of 1.9). Both groups of fish were able to elongate and desaturate the 18:3 n‐3 precursor into eicosapentaenoic acid and docosahexaenoic acid, regarding the FA profile of livers. Furthermore, total elongation/desaturation from [1‐14C]18:3 n‐3 of LC‐PUFA was higher in fish fed with the high dietary 18:3 n‐3 level compared to the diet rich in n‐3 LC‐PUFA. By opposition, the bioconversion of 18:2 n‐6 into LC‐PUFA was limited, regarding the elongation/desaturation activity of LC‐PUFA from [1‐14C]18:2 n‐6. In view of the great ability for bioconversion of n‐3 FA, linseed oil is a promising alternative to fish oil in formulating feed for juveniles perch as there were no differences in terms of specific growth rate between the treatments, but adults undergoing maturation should have at least partially LC‐PUFA in their diet, particularly arachidonic acid (ARA) which is important during maturation, as breeders are not able to bioconvert 18:2 n‐6 into ARA. 相似文献

Single phytotoxicity of two representative phthalate esters (PAEs), di-n-butyl phthalate (DnBP) and bis(2-ethylhexyl) phthalate (DEHP), was tested in mung bean (Vigna radiata) seedlings germinated for 72 h in soils spiked with varying concentrations (0-500 mg kg-1 soil) of DnBP or DEHP. PAEs added at up to 500 mg kg-1 soil exerted no significant effect on germination but both pollutants significantly inhibited root elongation (P 〈 0.01); DEHP inhibited shoot elongation (P 〈 0.01) and DnBP depressed biomass on a fresh weight basis (P 〈 0.05). Seedling shoot and root malondialdehyde (MDA) Contents tended to be stimulated by DnBP but inhibited by DEHP. However, increases in superoxide dismutase, peroxidase, ascorbate peroxidase and polyphenol oxidase activities, as well as glutathione (GSH) content, were induced at higher concentrations (e.g., 20 mg kg-1) of both compounds. Accumulation of proline in both roots and shoots and the storage compounds, such as free amino acids and total soluble sugars, in whole plant was induced under the stress exerted by both PAEs. The general responses of mung bean seedlings indicated higher toxicity of DnBP than DEHP on primary growth, during which root elongation was a more responsive index. MDA and GSH were more sensitive parameters in the roots than in the shoots and they might be recommended as physiologically sensitive parameters to assess the toxicity of PAE compounds in soils in future long-term studies. 相似文献

为今后利用SYBR GreenⅠ荧光定量法研究粘虫EF-1a和AK基因的表达情况,设计合成用于PCR扩增的引物,运用RT-PCR方法克隆EF-1a和AK基因片段,并进行序列同源性分析;然后采用SYBR Green Ⅰ荧光定量法设计特异性引物进行RT-PCR扩增,通过熔解曲线和标准曲线分析等建立实时RT-PCR方法。结果表明,从粘虫中克隆获得EF-1a和AK基因片段长度分别为622 bp和1020 bp,分别编码207个氨基酸和339个氨基酸;序列同源性分析结果表明,粘虫EF-1a与鳞翅目昆虫家蚕、桦尺蠖和柑桔凤蝶的EF-1a氨基酸序列同源性为95%以上;AK与鳞翅目昆虫草地贪夜蛾、烟蚜夜蛾、棉铃虫、斜纹夜蛾和甜菜夜蛾的AK氨基酸序列同源性为98%以上。熔解曲线和标准曲线结果显示,设计的EF-1a和AK基因引物均可应用于实时RT-PCR扩增。 相似文献
