ABSTRACT

The exploitation of phosphate mines generates an important quantity of phosphate sludge that remains accumulated and not valorized. In this context, composting with organic matter and rhizospheric microorganisms offers an interesting alternative and that is more sustainable for agriculture. This work aims to investigate the synergetic effect of arbuscular mycorrhizal fungi (AMF), phosphate-solubilizing bacteria (PSB) and phospho-compost (PC), produced from phosphate-laundered sludge and organic wastes, and their combination on plant growth, phosphorus solubilization and phosphatase activities (alkaline and acid). Inoculated mycorrhizae and bacteria strains used in this study were selected from plant rhizosphere grown on phosphate-laundered sludge. Significant (p < .05) increases in plant growth was observed when inoculated with both consortia and PC (PC+ PSB+ AMF) similar to those recorded in plants amended with chemical fertilizer. Tripartite inoculated tomato had a significantly (p < .05) higher shoot height; shoot and root dry weight, root colonization and available P content, than the control. Co-inoculation with PC and AMF greatly increased alkaline phosphatase activity and the rate of mycorrhizal intensity. We conclude that PC and endophytic AMF and PSB consortia contribute to a tripartite inoculation in tomato seedlings and are coordinately involved in plant growth and phosphorus solubilization. These results open up promising prospects for using formulate phospho-compost enriched with phosphorus-solubilizing microorganisms (PSM) in crop cultivation as biofertilizers to solve problems of phosphate-laundered sludge accumulation.  相似文献

As part of a study of the processes involved in litter biodegradation following sewage sludge (SS) addition, the variations over 14 months of phosphatase activities in a cork oak litter (Quercus suber L.) were investigated. A field experimental design was carried out using the litter-bag method on both a fertilized plot receiving SS applications (twice ) and a control plot. Acid (APH) and alkaline (BPH) phosphatases were measured, along with several biotic and abiotic variables potentially involved in the regulation of these enzymes. These included moisture, temperature, pH, water-extractable inorganic P (PW), culturable heterotrophic bacteria and fungi. Sludge addition had significant effects on all the variables measured. Indeed, sludge increased significantly BPH activities, available PW, microbial densities (i.e. bacteria and fungi) and pH in the fertilized plot. In contrast, APH activities decreased significantly following sludge addition. As a consequence, the BPH/APH ratio increased markedly and immediately in the fertilized plot, but only after the 1st amendment. Following the 1st preconditioning SS amendment, the 2nd fertilization had fewer effects on biological variables, because of summer dryness. The different properties examined varied significantly with incubation time, and most were significantly related to the seasonal patterns of litter moisture in this Mediterranean forest ecosystem. Hence, sewage sludge application modified the intensity of microbial responses to environmental factors, but biological patterns regulating P turn-over were maintained.  相似文献

California halibut Paralichthys californicus is an important commercial species with high aquaculture potential in Baja California Sur, México. To optimize the feeding process using live prey and/or inert diets, we evaluated alkaline proteases, pepsin, trypsin, chymotrypsin, leucine aminopeptidase, lipase, α-amylase, and acid and alkaline phosphatase activities on starved larvae and larvae fed live prey. Highest activities were observed for alkaline protease, trypsin, chymotrypsin, leucine aminopeptidase, and alkaline phosphatase in feeding larvae than starved larvae on day 4 after hatching. At day 5, a sizeable increase in all enzymatic activities was detected in feeding larvae. Alkaline protease, trypsin, chymotrypsin, and alkaline phosphatase decreases progressively from day 5 until day 18. At day 18, a slight pepsin activity was observed. This was considered an indicator of the start of digestive system maturation. We concluded that total enzymatic equipment for this species is complete between day 18 and 30 after hatching. Based on this evidence, early weaning from live prey to inert feed would be possible at this time.  相似文献

A comparison of intra‐ and extracellular acid phosphatase, alkaline phosphatase and phytase activity in six fungi is reported. A strong linear relationship between intra versus extracellular fungal acid phosphatase (R2 = 0.94), alkaline phosphatase (R2 = 0.96), and phytase (R2 = 0.97) is observed. Three‐fourth of acid phosphatase were generally present inside the fungal cells and only 25 % were released extracellularly after a three weeks period. Phytase shows the reverse trend where thirty nine times higher extracellular phytase activity was noticed than present inside the fungal cells. The extracellular enzymes are found 60 % more efficient in the hydrolysis of phytin than their intracellular counterpart but they are at par in the hydrolysis of glycerophosphate. The results clearly demonstrated that phytase types of phosphatases mostly occur outside the fungal cells whereas most of the acid phosphatase and alkaline phosphatase are located inside the cells.<?show $6#>  相似文献

Biochemical and hematological analyses of blood and serum were performed in pigs with regional ileitis and in wasting pigs with a negative necropsy. In sera from pigs with regional ileitis the levels of total protein, albumin, transferrin, alkaline phosphatase and zinc were significantly decreased compared with normal pigs of the same age. The number of white blood cells, and the concentration of Cortisol and α1-antitrypsin were significantly increased. In wasting pigs with early signs of regional ileitis or with a negative necropsy the same blood changes were observed but to a less degree. It was concluded that a wasting syndrome after weaning may precede regional ileitis. Concerning the etiology of regional ileitis the significance of malabsorption and wasting syndrome in combination with invasion of intestinal intercellular microorganisms is discussed.  相似文献

The effect of interspecific complementary and competitive root interactions and rhizosphere effects on primarily phosphorus (P) and iron (Fe) but also nitrogen (N), potassium (K), calcium (Ca), zinc (Zn), and manganese (Mn) nutrition between mixed cropped peanut (Arachis hypogaea L.) and barley (Hordeum vulgare L.). In order to provide more physiological evidence on the mechanisms of interspecific facilitation, phosphatase activities in plant and rhizosphere, root ferric reducing capacity (FR), Fe-solubilizing activity (Fe-SA), and rhizosphere pH were determined. The results of the experiment revealed that biomass yield of peanut and barley was decreased by associated plant species as compared to their monoculture. Rhizosphere chemistry was strongly and differentially modified by the roots of peanut and barley and their mixed culture. In the mixed cropping of peanut/barley, intracellular alkaline and acid phosphatases (AlPase and APase), root secreted acid phosphatases (S-APase), acid phosphatases activity in rhizosphere (RS-APase), and bulk soil (BS-APase) were higher than that of monocultured barley. Regardless of plant species and cropping system, the rhizosphere pH was acidified and concomitantly to this available P and Fe concentrations in the rhizosphere were also increased. The secretion Fe-solubilizing activity (Fe-SA) and ferric reducing (FR) capacity of the roots were generally higher in mixed culture relative to that in monoculture treatments which may improve Fe and Zn nutrition of peanut. Furthermore, mixed cropping improved N and K nutrition of peanut plants, while Ca nutrition was negatively affected by mixed cropping.  相似文献

The objective of this study was to investigate the growth of coffee (Coffea arabica L. cv. ‘Bourbon’) in soil amended with compost, vermicompost, and bokashi at 25%, 50%, and 66% while mycorrhizal colonization and activity of acid and alkaline phosphatases and urease in soil were monitored. Treatments with 25% compost as well as higher application rates of compost and bokashi (50% or 66%) increased coffee growth. Shoot fresh weight increased 5 times when 25% bokashi or compost was applied, 8 times when 50% was applied and 10 times when 66% was applied compared to the control plants without organic fertilizer. Acid and alkaline phosphatase and urease activity increased when organic fertilizer was applied to soil. Application of organic fertilizers increased the growth and fresh weight of root and shoot of coffee plants and improved the root mycorrizal colonization and enzymatic activity of the soil.  相似文献

With increasing size of inoculum (0.025, 0.25, 2.5 g of wet Staphylococcus aureus per 100 ml broth) maximal coagulase-activity (256–512 U/ml) was detected in the supernatant after 30 min-2 hr of incubation at 37°C. Alpha- and beta-hemolysins appeared later (after 8–24 hr of incubation). Acid/alkaline phosphatase could be demonstrated from the beginning of incubation, but their maximal activity in culture supernatant could be demonstrated after 8–24 hr of incubation (Table 1). Thus, size of inoculum and time of incubation have a greater significance in the optimal production of some staphylococcal substances than generally assumed.  相似文献

把铜锈环棱螺(Bellamya aeruginosa)暴露于不同浓度的有毒微囊藻藻液中(对照组:只投喂四尾栅藻(Scenedes musquadricanda);混合藻组:50%四尾栅藻+50%铜绿微囊藻(Microcystis aerugirtosa);蓝藻组:只投喂铜绿微囊藻),用酶联免疫检测法(Enzyme—linked immuno sorbentassay,ELISA)检测藻液和肝组织中藻毒素浓度,藻液中包括藻相和水相的总微囊藻毒素(MC)浓度分别为:对照组0ug·L^-1;混合藻组(14.47±1.22)ug·L^-1;蓝藻组(29.47±2.43)ug·L^-1。螺在两种不同毒素浓度藻液中暴露15d后再移人四尾栅藻藻液中降解15d。结果表明,暴露期间,混合藻组、蓝藻组螺肝组织中MC含量均为先增加后减少,再增加,且同期混合藻组MC含量都明显高于蓝藻组;作为机体代谢生物标志物的酸性磷酸酶(ACP)和碱性磷酸酶(ALP)活性随MC浓度及其暴露时间发生相应变化;作为解毒生物标志物的谷胱甘肽硫转移酶(GST)活性在混合藻组先被诱导后被抑制,在蓝藻组初期变化不明显后表现为诱导。在15d降解过程中,混合藻组和蓝藻组MC含量均持续下降;机体生物标志物ACP、AIJP和GST活性表现为不同程度的降低。本试验结果为ACP、ALP和GST活性作为MC胁迫的生物标志物提供了一些依据。  相似文献
