Systemic acquired resistance (SAR) is induced following inoculation of Peronospora tabacina sporangia into the stems of Nicotiana tabacum plants highly susceptible to the pathogen. Previous results have shown that accumulation of acidic β-1,3-glucanases (PR-2's) following induction of SAR by P. tabacina may contribute to resistance to P. tabacina. We showed that up-regulation of the PR-2 gene, PR-2d, following stem inoculation with P. tabacina, is associated with SAR. Studies using plants transformed with GUS constructs containing the full length promoter from PR-2d or promoter deletions, provided evidence that a previously characterized regulatory element that is involved in response to salicylic acid (SA), may be involved in regulation of PR-2d following induction of SAR with P. tabacina. This work provides evidence that regulation of PR-2 genes during P. tabacina-induced SAR may be similar to regulation of these genes during infection of N-gene tobacco by TMV or following exogenous application of SA, and provides further support for the role of SA in regulation of genes during P. tabacina-induced SAR.  相似文献

Abstract

In Japan, soybean is usually cultivated in fields that have been converted from rice paddies, and poor seedling establishment due to pre-emergence seedling damping-off is often observed during the rainy season. In this study, the factors that cause the damping-off in flooded soil were investigated under high soil moisture conditions in a greenhouse and in agricultural fields. In sterilized soil sampled from a soybean field, seedlings emerged well under 48-hr flooded conditions. In unsterilized soil, soybean seeds treated with the fungicide, mancozeb+metalaxyl exhibited much higher emergence rates even under 48-hr flooded conditions than the seeds treated with oxytetracycline +streptomycin, benomyl, or flutolanil. Pythium, Phytophthora, Mucorales, Trichoderma, Geotrichum-like microorganisms, and some fungi producing conidia in a false head, were isolated from decayed seedlings. Of the isolated microorganisms, oomycete microorganisms, Pythium helicoides, other Pythium sp., and Phytophthora sp. were pathogenic to soybeans under flooded conditions. As the length of the flooding period increased, pre-emergence seedling rot also increased. However, the pathogenic oomycetes rarely caused pre-emergence seedling rot under conditions without flooding. Furthermore, under flooded conditions, the damage caused by these pathogens was reduced by treating the seeds with mancozeb+metalaxyl. These results indicate that oomycete microorganisms are involved in pre-emergence seedling damping-off under flooded soil conditions.  相似文献

Lettuce downy mildew caused by Bremia lactucae has long been a model for understanding biotrophic oomycete–plant interactions. Initial research involved physiological and cytological studies that have been reviewed earlier. This review provides an overview of the genetic and molecular analyses that have occurred in the past 25 years as well as perspectives on future directions. The interaction between B. lactucae and lettuce (Lactuca sativa) is determined by an extensively characterized gene-for-gene relationship. Resistance genes have been cloned from L. sativa that encode proteins similar to resistance proteins isolated from other plant species. Avirulence genes have yet to be cloned from B. lactucae, although candidate sequences have been identified on the basis of motifs present in secreted avirulence proteins characterized from other oomycetes. Bremia lactucae has a minimum of 7 or 8 chromosome pairs ranging in size from 3 to at least 8 Mb and a set of linear polymorphic molecules that range in size between 0.3 and 1.6 Mb and are inherited in a non-Mendelian manner. Several methods indicated the genome size of B. lactucae to be ca. 50 Mb, although this is probably an underestimate, comprising approximately equal fractions of highly repeated sequences, intermediate repeats, and low-copy sequences. The genome of B. lactucae still awaits sequencing. To date, several EST libraries have been sequenced to provide an incomplete view of the gene space. Bremia lactucae has yet to be transformed, but regulatory sequences from it form components of transformation vectors used for other oomycetes. Molecular technology has now advanced to the point where rapid progress is likely in determining the molecular basis of specificity, mating type, and fungicide insensitivity.  相似文献

The efficacy of a new fungicide fluopicolide in suppression of Phytophthora blight caused by Phytophthora capsici was evaluated under laboratory and field conditions. Studies with 51 P. capsici isolates from vegetable crops in Georgia, USA, indicated that 5.9% of the isolates were resistant, 19.6% were intermediately sensitive, and 74.5% were sensitive to 100 μg ml−1 of mefenoxam based on in vitro mycelial growth. EC50 values of fluopicolide in inhibiting mycelial growth of 25 isolates, representing resistant, intermediately sensitive, and sensitive to mefenoxam, ranged from 0.05 to 0.35 μg ml−1 with an average of 0.2 μg ml−1 EC50 values of fluopicolide in suppressing zoospore germination and sporangium production of the 25 isolates ranged from 1.1 to 4.5 μg ml−1 and 0.3–9.0 μg ml−1, respectively. Evaluation of a collection of 150 P. capsici isolates from vegetables and irrigation ponds found none of the isolates were resistant to 10 μg ml−1 of fluopicolide. Field experiments were conducted to determine the efficacy and application methods of fluopicolide for control of P. capsici on squash in spring 2007 and 2009. Fluopicolide applied through drip irrigation or as a foliar spray at 86.6 or 115.4 g ha−1 consistently provided significant disease reduction and increased squash yield. Results with fluopicolide were similar or slightly superior to mefenoxam applied at recommended rate. Fluopicolide applied at 57.7 g ha−1 did not provide consistent satisfactory disease suppression. The results indicated that fluopicolide was effective in suppression of different stages of the life cycle of P. capsici and could be a viable alternative to mefenoxam for managing Phytophthora blight in squash production.  相似文献

BACKGROUND: Pythium insidiosum is an aquatic oomycete that causes severe segmental thickening of the canine gastrointestinal (GI) tract, resulting in weight loss, vomiting, diarrhea, and death. Infection in dogs previously has been observed primarily in the southeastern United States. OBJECTIVE: To describe the clinicopathologic and epidemiologic findings associated with GI pythiosis in 10 dogs from California. METHODS: Dogs were initially identified on the basis of supportive clinical findings and routine histology. Pythiosis was confirmed in each dog with at least one of the following: immunoblot serology, enzyme-linked immunosorbent assay serology, immunohistochemistry, and culture followed by species-specific polymerase chain reaction, rRNA gene sequencing, or both. RESULTS: Between September 2003 and December 2006, GI pythiosis was confirmed in 1 dog from central California and 9 dogs that lived within a 30-mile radius of Davis, CA. Seven of 8 dogs for which environmental data were available had frequent access to flooded rice fields or other water sources. Esophageal lesions were present in 2 of 10 dogs. Common laboratory findings included eosinophilia (7/9), hypoalbuminemia (9/9), and hyperglobulinemia (8/9). Median survival time was 26.5 days (range, 0-122 days), and the disease was ultimately fatal in all 10 dogs. CONCLUSIONS AND CLINICAL IMPORTANCE: The geographic distribution of pythiosis has widened in recent years to include the western United States. Factors that may have contributed to this change include altered rice-farming practices and landscape irrigation. Veterinarians in California should be familiar with the clinicopathologic features associated with GI pythiosis to aid in early diagnosis and effective treatment.  相似文献

A clinical epidemiological study was conducted among 34 rural properties located within the Brazilian Pantanal region and nearby areas between 2007 and 2010. The diagnosis of equine pythiosis was based on antibody detection (by enzyme-linked immunosorbent assay), polymerase chain reaction, histopathological analysis, and cultures positive for Pythium insidiosum. The majority of the affected animals (85%) were in the Pantanal biome, which had a higher disease prevalence (0.9%-66.7%) than that of the Cerrado (2.7%-33.3%). The disease was more prevalent in the rainy season (January-March), with an abrupt fall in the number of cases during the drought period (April–September; correlation of R2 = 0.77; P < .01). Generally, the average prevalence of equine pythiosis in both regions was 5%, with mortality and lethality rates of 1.3% and 23.1%, respectively, in the Pantanal and 2.3% and 45.5%, respectively, in the Cerrado. However, the treatment with immunotherapy may have underestimated these numbers, especially in the Pantanal. Animals older than 1 year were 8.09 times more affected by the disease than younger animals in the same environment (P < .05). A correlation between the anatomical area of the lesion and the type of skin color was also observed. Approximately 73% of the lesions were found in dark-pigmented areas, and animals with a dark coat color were affected more frequently. These findings highlight the importance of hematophagous insects in the epidemiology of pythiosis because these areas are preferred for blood feeding.  相似文献

Activity of carboxylic acid amide (CAA) fungicides against Bremia lactucae   总被引:1,自引:1,他引:0  
Four carboxylic acid amide (CAA) fungicides, mandipropamid (MPD), dimethomorph (DMM), benthiavalicarb (BENT) and iprovalicarb (IPRO) were examined for their effects on various developmental stages of Bremia lactucae, the causal agent of downy mildew in lettuce, in vitro and in planta. Spore germination in vitro or on leaf surfaces was inhibited by all CAA fungicides (technical or formulated). MPD was more effective in suppressing germination than DMM or BENT, whereas IPRO was least effective. CAA induced no disruption of F-actin microfilament organisation in germinating spores of B. lactucae. CAA applied to germinating spores in vitro prevented further extension of the germ tubes. When applied to germinated spores on the leaf surface they prevented penetration. Preventive application of CAA to intact plants inhibited infection. MPD was more effective in suppressing infection than DMM or BENT, whereas IPRO was least effective. Curative application was effective at ≤3 h post-inoculation (hpi) but not at ≥18 hpi. CAA (except IPRO) applied to upper leaf surfaces inhibited spore germination on the lower surface and hence reduced infection. CAA suppressed sporulation of B. lactucae on floating leaf discs and when sprayed onto infected plants two days before onset of sporulation. BENT and DMM were more effective in suppressing sporulation than MPD or IPRO. Epidemics of downy mildew in shade-house grown lettuce were suppressed by CAA. A single spray applied to five-leaf plants before transplanting controlled the disease for 50 days. The results suggest that CAA are effective inhibitors of spore germination and therefore should be used as preventive agents against downy mildew of lettuce caused by B. lactucae.  相似文献

Several plant pathogenic oomycetes have been under investigation using modern molecular approaches. Genome sequencing and annotations are underway or near to completion for some of the species. Pathogen-associated molecular pattern molecules (PAMPs) and effector molecules perform inter- and intracellular tasks as adaptation factors and manipulators of the defence network. Hundreds of secreted putative effectors have been discovered and conserved molecular patterns such as RXLR and EER motifs have been identified and used for classifications. PAMPs and effectors are recognized directly or indirectly by the pattern recognition receptors at the cell surface including receptor-like kinases and receptor-like proteins, and/or by nucleotide binding site–leucine rich repeat proteins within the cytoplasm. The current knowledge of effectors, immune receptors and the defence network, will help us understand the ‘intricate genetic dance’ between the oomycete pathogens and their hosts. This review concentrates on the recent findings in oomycete-plant interactions.  相似文献

Bioassays testing the fungicide sensitivity against metalaxyl of Peronospora tabacina isolates collected in German tobacco fields in 2005 revealed the presence of two phenotypes, resistant and sensitive. DNA fingerprints using SSR and minisatellite primers allowed separation of the samples into two groups. The differences in amplification patterns coincided with the sensitive and resistant reaction of the isolates in metalaxyl bioassays. New primers were developed which allowed PCR-based detection of P. tabacina and differentiation of the metalaxyl-sensitive and the metalaxyl-resistant phenotype, respectively. Screening of recent blue mold isolates from Germany and other European countries for metalaxyl sensitivity with leaf disk bioassays coincided completely with the PCR-based identification of the two phenotypes. In Germany, exclusively resistant isolates were found between 2002 and 2004. These still dominate. Since 2005 the co-occurrence of sensitive isolates has been shown. No similar monitoring has been done in any other European country. However, we found the resistant phenotype reaction in a French isolate of 2004 and in two out of three Italian isolates in 2007. Two isolates from Poland and Bulgaria in 2007 were sensitive to metalaxyl. For all 58 isolates tested since 2002 the metalaxyl bioassay-based resistance type and the PCR-based tests for sensitive and resistant genotype coincided. Using genotype-specific primers for future population studies may help to trace the sources of the pathogen from which yearly propagation starts.  相似文献
