Acid soluble collagen (ASC) and pepsin soluble collagen (PSC) were isolated from rohu skin with the yield of 64.2 and 6.8% (dry weight basis), respectively. Both collagens had glycine as the major amino acid with imino acid content of 196–202 residues/1,000 residues and were characterized as type I collagen with molecular composition of (α1)2α2-heterotrimer. Fourier transform infrared spectra of both collagens were similar, with no shift in wavenumber of all amide bands. The Tmax value of ASC and PSC was 36.40 and 35.48°C, respectively. The zero surface net charge of ASC and PSC was found at pH 5.9 and 5.3, respectively.  相似文献

Summary Self incompatibility (SI) can be used to alleviate costly hand emasculation and pollination in F1 hybrid chrysanthemum seed production. SI, however, disrupts the progression of inbreedig (selfing or full-sib mating). Consequently, inbreds are selected for breakdown of the SI system or the presence of pseudo-self compatibility (PSC). PSC inbreds, recombinant inbreds, and noninbred cultivars were selfed and/or intercrossed to determine PSC expression across environments and generate 1–3 inbred generations (I1–I3). Percent PSC ranged from 0–68.8% for inbred parents, 0.2–99.7% for recombinant inbreds, and 0.6–25.7% for noninbred cultivars. There was no indication of end-of-season PSC. The majority of parents (78%) were classified as low PSC and this trend continued in the I1 (70.1%), I2 (65.6%), and I3 (83.6%) generations; mid PSC was rarer (11.9–18.8%) and high PSC the least common (4.5–15.6%). PSC distributions were primarily continuous, rather than discrete, indicating quantitative inheritance. In several inbred families, 100% of the I1 individuals were SI; this was not correlated with parental PSC level. Inbred families derived from selfing low and mid PSC parents were the most likely to reach extinction due to inbreeding depression. High PSC was not highly heritable, since I1 progeny were predominantly SI or had low to mid PSC levels. Most 153-1 values for PSC:SI segregations (1:1, 1:3, 3:1) were not significant at the 5% level. Realized heritability (H R ) estimates for PSC ranged from a low of 0.05% to 10.19%, although increased H R values did not account for inbreeding depression or genetic mechanisms preventing selection for high PSC. The highest individual %PSC increased over the high parent with SI × low PSC or SI × mid PSC parents in all cases; progeny means did so only in SI × low PSC. Low × low crosses were split evenly between an increase and decrease in progeny or highest individual mean. Since all low × low crosses and low selfs produced 43–50% of the progeny with PSC levels higher than the parents, it appears that most low parents possess some unexpressed PSC genes. Mid PSC parents responded similarly to low PSC genotypes. An increase in PSC was found when crossing SI × PSC parents, illustrating a threshold effect. PSC × PSC crosses (high × low, low × low) produced SI, low, and mid PSC I1 individuals but did not show a heterotic effect, since the PSC parents had already surpassed the PSC threshold. Selfing high PSC parents, however, suggested different genetic control. Progeny and highest individual means behaved the same; 83% decreased and 17% remained the same for PSC levels. The drop in PSC indicated non-additive gene action. Thus, the PSC threshold with additive gene action holds when selection for higher PSC levels is being done from low and mid PSC parents, but once high PSC levels are obtained, non-additive gene action prevails.Abbreviations self pollinated - H R Realized Heritability - PSC Pseudo-Self Compatibility - SC Self Compatibility - SI Self Incompatibility  相似文献

以紫蛇尾Ophiopholis mirabilis为原料检测其基本成分,并用盐酸对紫蛇尾进行脱钙处理,通过L9(34)正交试验对其胶原蛋白的提取工艺进行了优化。结果表明:紫蛇尾含水分51.80%,灰分42.91%,蛋白质1.68%,脂肪0.29%,总糖0.03%;酶促溶性胶原蛋白(PSC)提取工艺的优化条件为脱钙后的紫蛇尾干粉(g)与提取液(mL)的固液比为1∶15,胃蛋白酶加酶量3%,17℃下提取4 d,PSC得率为52.65%。对纯化后的酸促溶性胶原蛋白(ASC)和PSC进行紫外扫描分析,结果显示,ASC、PSC的最大吸收波长均在235 nm左右。对PSC的氨基酸组成分析表明,脯氨酸和羟脯氨酸的含量分别为10.1%和2.13%,是较典型的Ⅰ型胶原蛋白。  相似文献

The performance of electronic nose (E-nose) for Chinese Cymbidium scent profiling has been evaluated. Changes in scent profiles of two Cymbidium ensifolium cultivars have been monitored at different flowering stages (initial flowering, full flowering, and terminal flowering) and different times combined with two gas collecting devices. Samples were collected by static headspace (SHS) method. How E-nose can be used for pattern recognition and for studying the releasing of flower scent were proposed. Data obtained were subjected to principal component analysis (PCA) and discriminant function analysis (DFA). PCA was performed on the initially instrumental data to explore the structure of each data set and such result showed that the sensory data contained information related to the cultivar and to time spots. DFA was performed to improve the results, leading to clear separations between the sample groups. Gas collecting device did not seriously affect the result of PCA and DFA. Relative aroma intensity (RAI) was proposed as an alternative concept to compare scent intensity between samples on different time points. These results demonstrate the potential application of the E-nose to evaluate the scent profile of flower.  相似文献

Abstract

Iron (Fe)‐enriched concretions, a complex natural matrix with high chemical heterogeneity and phosphate‐sorption capacity, is widespread in soils with restrictive drainage in Greece. However, the phosphorus (P) status and related characteristics of Fe‐enriched concretions in agricultural soils in areas where P fertilization is mainly inorganic are relatively unknown. Active noncrystalline Fe and aluminum (Al) oxides (Feox, Alox), oxalate extractable P (Pox), P sorption capacity (PSC), and the degree of P saturation (DPS) of Fe‐enriched concretions from agricultural imperfectly drained soils in central Greece were determined using the acid ammonium oxalate method. The concretions contain 13 times as much Feox, twice as much Alox, and almost 15 times as much Pox than the surrounding soil matrix. Pox accounted for 50–80% of total P of the soil concretions, indicating strong accumulation of noncrystalline P components (Al‐ and Fe‐P). The PSC, expressed as a 0.5 (Alox+Feox), ranged from 184.7 to 314 mmol kg?1, demonstrating the strong affinity of the Fe‐enriched concretions for P. The DPS, which represents the fraction of concretion sorbent surface coverage by P, was computed as 100 (Pox/PSC) with values ranging from 6 to 13% (mean=8%). The results of this study indicate that the Fe‐enriched concretions, due to their high noncrystalline Fe and Al oxides content, act as major sink of phosphate, controlling the location, mobility, and dynamics of P in agricultural soils with restrictive drainage.  相似文献
