以菊苣无菌苗的子叶为外植体,研究其在附加不同浓度NAA和6-BA的培养基上的分化情况.结果表明:在不同培养基上,菊苣子叶均能诱导出淡黄色的愈伤组织和分化形成不定芽,但不同基因型菊苣的出愈率和不定芽诱导存在明显差别.诱导Rossa di Verona tardiva selez prelude和Rossa di treviso菊苣愈伤组织形成和芽分化的最佳培养基为MS+NAA 0.5 mg/L+6-BA 2 mg/L;而诱导In miscuglio菊苣愈伤组织和芽分化的最佳培养基为MS+NAA 0.5 mg/L+6-BA 3 mg/L.  相似文献

实验旨在研究孕酮对蛋鸡CaBP-d28k基因表达的影响。对产蛋高峰期蛋鸡进行孕酮和孕酮受体拮抗剂(RU-486)处理,利用实时荧光定量PCR技术(QRT-PCR),测定在蛋壳形成过程中输卵管子宫部钙结合蛋白(CaBP-d28k)、孕酮受体(PGR)的表达量;同时对血液中钙离子和孕酮浓度变化进行测定。结果表明:孕酮处理后,血液中钙离子浓度显著降低(P<0.05),输卵管子宫部CaBP-d28k表达量也显著降低(P<0.05);孕酮受体拮抗剂处理后,血液中钙离子浓度显著升高(P<0.05),输卵管子宫部CaBP-d28k表达量极显著升高(P<0.01)。结果提示,在蛋壳钙化过程中,孕酮对CaBP-d28k表达和钙离子转运起负调控作用。  相似文献

Summary The establishment of representative core collections and back-up reserve collections was proposed to facilitate effective management and to promote utilization of large base collections. The priorities of the developing national PGR programmes, in the organization of components of their base collections, are different from those of IARCs since the strengths as well as limitations of the two systems vary. Unlike most IARCs, national programmes have the option of networking their active collections, maintained at several eco-sites, and linking this network to the base collection kept under long-term storage. This keeps open the option to develop situation-specific subsets for an effective germplasm utilization. There is a pressing need for a scientific re-examination of the concept of germplasm core in order better to apply it in developing core subsets in the national PGR programmes. The formulation of situation-specific subsets is advocated, as the system would be directed to users' requirements or addressed to gene bank managers' resource constraints. Arguments given in favour of developing core subsets, rather than a single core are: (i) logical; (ii) population genetic; and (iii) germplasm usage considerations. The in vitro formation of newly formed adventitious buds and shoots from internodal branch segments was studied on 12-month-old plants of Citrus aurantium L. cv. Brazilian. The effects of 6-Benzyladenine (BA) and α-Naphthalene acetic acid (NAA) treatments were evaluated on adventitious bud and shoot regeneration. High rates of bud initiation and shoot development were obtained both with BA supplemented medium, in the range from 1 mg L−1 to 3 mg L−1, and with 0.1 mg L−1 NAA supplemented medium. NAA concentrations above 1 mg L−1 significantly reduced bud initiation and shoot elongation. The results obtained using different in vitro culture vessels such as Petri dishes, tubes and glass culture jars were compared. The highest adventitious bud induction was observed in Petri dishes for internodes cultured in 2 mg L−1 BA supplemented medium, with 95% responsive explants forming 9.0 ± 2.4 adventitious buds. The adventitious buds observed in Petri dishes reached a maximum height of 1 mm, with no further development, while some of the adventitious shoots cultured in tubes and glass culture jars grew over 1 cm in height. A shoot regeneration gradient of the internodes collected along the branch axis was noticed, with basal ones exhibiting higher regeneration frequency.  相似文献

Towards the selection of taxa for plant genetic conservation   总被引:1,自引:1,他引:1  
The signing and ratification of the Convention on Biological Diversity has resulted in increased scientific and public interest in the conservation and utilisation of biological diversity. This has in turn triggered the need to develop more effective methodologies to conserve biological diversity for the benefit of all humanity. One of the first factors to be considered when conserving botanical diversity is the efficient and effective selection of the target taxa. The aim of this paper is not to set detailed priorities for the genetic conservation of the plant genetic resources of any particular crop or species complex, but to draw attention to the factors that should be considered when formulating priorities. The factors that are considered important when selecting plant genetic resource targets are: current conservation status; potential economic use; threat of genetic erosion; genetic distinctiveness; ecogeographic distribution; biological importance; cultural importance; cost, feasibility and sustainability; legislation; ethical and aesthetic considerations; and priorities of the conservation agency undertaking the conservation. Each of these factors is discussed in turn. Although it is not possible to provide a single methodology for the selection of plant genetic resource targets at this time, it is hoped that the consideration the factors discussed will make the selection of target taxa more object, make better use of the limited conservation resources and thus enhance the process of genetic conservation as a whole.  相似文献

Plant growth regulators (PGR) have potential to increase grain yield and may also alter grain protein levels of cereal crops. A 3-yr field experiment with spring barley ( Hordeum vulgare L.) and wheat ( Triticum aestivum L.) cultivars was conducted to determine whether ethephon treatment increased protein concentration, protein yield, and grain yield. A greenhouse experiment was also conducted to evaluate the response of barley grain protein concentration to gradual addition of ethephon (2.2 × 10-3 mM) or chlormequat (5.8 × 10-3 mM) solution after an thesis. Under field conditions, ethephon treatment increased barley and wheat grain protein concentrations by as much as 16 % but decreased grain yield so that protein yield increases were small or did not occur. For Laurier barley, total gram N content was increased by up to 20 % in one year of this study. However, over the 3 years, an inverse relation existed between grain protein and yield. Greenhouse data showed that i) the gradient in grain size and protein concentration among spikelets of a spike, which is established before anthesis, was not affected by either chlormequat or ethephon; and ii) chlormequat increased grain protein by 7 to 11 % whereas ethephon increased protein concentration by up to 13 % in one of the two experiments. Our data indicate that PGR can alter protein accumulation in the grain, and thus, the quality of bread wheat and feed barley crops can be increased m regions with a short crop-growing season. However, a portion of the increase in gram protein concentration is due to decreased starch deposition, which is associated with yield reductions. The greenhouse data confirmed that a portion of the increase in grain protein concentration due to PGR application is caused by increased protein accumulation in the barley grain.  相似文献

Maize (Zea mays L.) yield is a function of the number harvested kernels per unit land area and the individual kernel weight (KW). Kernel weight and its development show a wide variability due to the genotype, the environment, the crop management, and all possible interactions. Commercial maize hybrids differ markedly in the patterns (rate and duration of kernel growth) behind differences in final KW. The same can be observed when public or elite proprietary maize inbred lines are analyzed. To progress in our understanding of KW variability, we reviewed and discussed current knowledge for analyzing kernel growth as an integrated system, modulated by processes linking different levels of organization (the different kernel tissues, the whole kernel, the plant, the canopy). Ideas on how to integrate this knowledge towards the development of a multi-hierarchical scale framework for predicting KW under different growth environments are currently needed, as they have high relevance for dissecting the genetic basis of kernel growth and maize yield definition at the canopy level.  相似文献
