AIM: To investigate the effect of growth hormone receptor (GHR) knockdown on nuclear factor-κB (NF-κB) activity and inflammatory cytokine production stimulated by growth hormone (GH) in 3T3-L1 adipocytes. METHODS: The specific siRNA for GHR was transfected into 3T3-L1 adipocytes to silence GHR expressions. The effects of GH on NF-κB activation and inflammatory cytokine production in 3T3-L1 adipocytes transfected with siRNA-GHR or siRNA-control were measured by dual-luciferase system analysis, real-time RT-PCR and ELISA. RESULTS: The protein expression of GHR was diminished after transfection with GHR specific siRNA. Dual-luciferase reporter system analysis revealed that GHR knockdown resulted in attenuation of GH-stimulated NF-κB activation in the 3T3-L1 adipocytes. GHR knockdown ameliorated the GH-induced production of inflammatory cytokines TNF-α, IL-1β, IL-6, MCP-1 and MIP-1α in the 3T3-L1 adipocytes. CONCLUSION: Knockdown of GHR might be efficacious to prevent GH-induced inflammatory responses in the 3T3-L1 adipocytes.  相似文献

Efficient utilization of isabgol (Plantgo ovata Forsk.) straw biomass through preparation of bio-active organic fertilizer in a natural composting process has not been studied. In this study, phosphate rock and silicate mineral (mica) powder were used as natural sources of phosphorus (P) and potassium (K), respectively. Cow dung slurry as a natural decomposer was mixed with the straw biomass at a 10:1 (weight/weight) ratio along with mineral powder. Then, nutrient-mobilizing bio-inoculants were used in the composting process after attaining thermal stabilization. The agronomic effectiveness of the resulting bio-active compost (BAC) as a bio-organic fertilizer was compared with that of conventional organics (farmyard manure and vermicompost) and chemical fertilizer (CF) by growing isabgol under field conditions. Composting with the natural sources of P and K along with the bio-inoculants increased the total nitrogen (13.6 g kg-1), P (38.7 g kg-1), and K (31.2 g kg-1) contents in the final product (i.e., BAC) compared with composting without the mineral powder and bio-inoculants. Application of BAC remarkably improved the seed yield (2.5%) and husk quality of isabgol in comparison with conventional organics and CF. Compared with CF, BAC significantly boosted the economic yield of isabgol by improving the husk recovery (2.5%) and mucilage yield (4.12%). Furthermore, BAC significantly improved the soil quality by increasing organic carbon (C), available nutrients, and microbial biomass C contents, as well as enzyme activity. The positive correlation between soil and plant parameters also highlighted the benefits of BAC for isabgol production through soil quality improvement. Therefore, it can be considered as a zero-waste technology, whereby a large quantity of straw biomass generated from isabgol cultivation, which contains essential nutrients, can be recycled back to the soil. Furthermore, BAC can be effectively used as a bio-active organic fertilizer, particularly in systems where chemical inputs are restricted, such as organic agriculture.  相似文献

目的探讨直肠前壁低度恶性间质瘤的诊断及手术治疗效果.方法回顾性分析3例直肠前壁低度恶性间质瘤患者的临床资料.结果3例患者均行手术治疗,经病理及免疫组化证实均为直肠低度恶性间质瘤.其中,2例完整切除,1例未完整切除.未完整切除者术后16个月死亡,另2例现无瘤存活.结论直肠前壁低度恶性间质瘤临床虽极少发生,但术前仍可作出初步诊断,病理及免疫组化可确诊,术中要尽可能完整切除肿瘤,这是治疗的关键,可明显改善预后.  相似文献

Oesophageal leiomyosarcoma has yet to be reported in dogs. This retrospective case series describes the case management and clinical outcome of four dogs with oesophageal leiomyosarcoma treated by marginal excision alone. Histological features used to determine tumour grade included capsular invasion, percent necrosis, pleomorphism and mitotic rate. All tumours were designated grade 1 leiomyosarcoma. Excision of all grossly evident tumour tissue was achieved in two of the four cases; however, histopathologic evaluation showed tumour cells at the surgical margins in one of these two cases. Two dogs had grossly incomplete excision. Two dogs died from unrelated conditions, one 3 years and 5.5 months after surgery, the other at 65 days. One dog had persistent mega‐oesophagus and was lost to follow‐up 388 days after surgery and one dog is still alive (last follow‐up 405 days after surgery). Despite large tumour size and incomplete excision, surgical removal of low‐grade leiomyosarcomas can result in long‐term resolution of clinical signs.  相似文献
