Infections Investigation of ALV and REV in Guangxi Local Poultry Breeds and Sequence Analysis of gp85 Gene of ALV Isolates

In order to investigate the infection status of avian leukosis virus (ALV) and avian reticuloendotheliosis virus (REV) in the major local breeds of Qinzhou,Guangxi,totally 953 samples of egg white,cloaca swab and serum of Ma duck,Shitou goose,Tiejiao-Ma chicken,turkey and pigeon were collected from the representing flocks and detected by the commercial ELISA kits.ALV was isolated for the ALV p27 positive samples by culturing on DF-1 cells,and gp85 gene was sequenced.The results showed that the detections of ALV were negative in the samples except those of Tiejiao-Ma chicken,while REV antibody was found positive in Ma duck,Tiejiao-Ma chicken and turkey.The nucleotide sequences of gp85 gene of two isolates shared 94.5% identity with each other,and shared 86.9% to 94.9% with reference strains.The amino acid sequences of gp85 gene of two isolates shared 91.5% identity with each other,and shared 84.0% to 91.6% with reference strains.There were many variable sites in the hyper variable region hr1 and hr2,and the vr2 and vr3 variable regions were relatively conservative.Phylogenetic tree analysis showed that the two isolates shared the highest homology with SCAU11-XG strain.     

Pathogenesis of HIV-associated dementia and its potential drug targets

HIV-associated dementia (HAD) is a serious complication of AIDS patients. With the wide application of highly active antiretroviral therapy (HAART), the replication of HIV is under effective control and the incidence of HAD is also declined. However, a relatively mild HIV-associated symptom of dementia, called minor cognitively motor disorder (MCMD), become the problem and can not be neglected in the treatment of AIDS. The neural injury caused by HIV may be mediated mainly by macrophages, microglia and astrocytes, though we can not rule out the direct damage on neurons by HIV proteins. To date, the precise mechanism of neural damage caused by HIV remains unclear. The present review tries to figure out the recent progress of pathogenesis and potential drug targets for HAD.     

噬菌体Bp7尾丝蛋白gp37的原核表达与鉴定

为了研究噬菌体尾丝蛋白gp37在识别宿主菌大肠埃希菌K12过程中的作用,PCR扩增获得gp37基因C端1 161bp序列,以EGFP为荧光标签,构建重组表达质粒pET-28a-EGFP-gp37,在大肠埃希菌BL21(DE3)中IPTG诱导表达重组蛋白EGFP-gp37,亲和层析纯化蛋白。SDS-PAGE及Western blot检测结果表明,重组蛋白EGFP-gp37在大肠埃希菌BL21(DE3)为可溶性表达,表达量占菌体总蛋白量的23%。EGFP-gp37融合蛋白与大肠埃希菌K12相互作用,激光共聚焦显微镜检测结果显示,尾丝蛋白gp37能够吸附到宿主菌K12表面,表明尾丝蛋白gp37参与宿主菌受体的识别和吸附过程。     

深圳市罗湖区415名社区人群亚健康现况调查

目的 了解深圳市罗湖区社区居民亚健康状态现况及人群分布特点,为亚健康状态的干预提供相关依据.方法 采用多阶段随机抽样,对深圳市罗湖区10个社区健康服务中心415名社区居民采用症状自评量表(SCL-90)进行亚健康测评和身心症状分析.结果 调查对象的SCL-90总分及总均分与全国常模比较差异无统计学意义(P＞0.05),亚健康状态主要表现为躯体症状和精神病性症状;相对于女性,男性易出现偏执症状;相对于高文化人群,低文化人群易出现躯体化、抑郁、焦虑以及敌对、精神病性等障碍;相对年轻人群,老年人群易出现躯体化、强迫、焦虑及抑郁等症状.结论 深圳市罗湖区居民身心健康状况良好,但仍存在一些危害健康的亚健康症状,必须加强社区居民健康教育,对不同健康状态的人群进行相应干预.     

松材线虫及其近缘种热激蛋白90基因(hsp90)克隆与序列分析

热激蛋白90家族(heat shock protein 90(HSP)family)是一种进化保守的蛋白质,在分子伴侣功能、细胞循环调控、抗原传递方面发挥着重要作用。本研究用RT-PCR和RACE技术,克隆得到了松材线虫(Bursaphelenchus xylophilus)、拟松材线虫(Bursaphelenchus mucronatus)和豆伞滑刃线虫(Bursaphelenchus doui)热激蛋白90基因的cDNA全长序列,分别命名为Bx-hsp90、Bm-hsp90和Bd-hsp90,(GenBank登录号分别为:GU013561、HMO34943和HM347331),其cDNA全长分别为2255、2193和2232bp,开放阅读框均为2127bp,编码708个氨基酸,5'端非编码区的长度分别为33、13和13bp,3'端非编码区的长度分别为95、53和92bp。其DNA全长分别为2440、2388和2407bp,包含3个内含子。三者氨基酸序列的一致性为98.78%。进化分析表明,松材线虫、拟松材线虫、豆伞滑刃线虫与秀丽小杆线虫(Caenorhabditis elegans)的亲缘关系较近,推测三...     

有机酸对液培玉米吸收137Cs和90Sr的影响

利用液培方法研究胡敏酸钠等有机酸盐作用下玉米幼苗对放射性核元素137Cs和90Sr的吸收速率。研究结果表明:利用有机酸盐可以降低玉米幼苗对放射性核元素的吸收速率。1低活度放射性核素水平下,对照(不加有机酸盐)的玉米幼苗中发现了97%放射性核元素137Cs,而高活度放射性核素水平中为61%;有机酸盐不同程度地降低了玉米幼苗中放射性核元素137Cs的量。在低活度水平中四苯硼钠降低137Cs的量最多, 玉米仅吸收12%,而酒石酸钾钠处理中玉米吸收量最高,为45.5%。对137Cs的吸收率,在胡敏酸钠处理中为23.9%, EDTA-Na处理中为41.3%;在高活度放射性核素水平中吸收率的顺序为四苯硼钠11%,胡敏酸钠26%,酒石酸钾钠28.7%,EDTA-Na 37%。2低活度水平下,对照中放射性核元素90Sr的56%被玉米幼苗吸收,而高活度水平中达61%。有机酸盐在低活度水平中降低放射性核元素90Sr的顺序为胡敏酸钠 23.9%,EDTA-Na 26%,酒石酸钾钠28%和四苯硼钠36%;在高活度水平中为四苯硼钠11%,EDTA-Na 24%,胡敏酸钠26%,酒石酸钾钠 31%。3低活度和高活度水平中,四苯硼钠和胡敏酸钠降低玉米幼苗体内的放射性核元素性质为最佳。4所有有机酸盐处理与对照之间的差异都达到极显著水平。     

Differential HSP90α expression in fish hepatocytes from polluted estuary during summer

The molecular chaperone, heat shock protein 90α (HSP90α), plays an important role in protein folding, degradation of denatured proteins and steroid activation. It is essential for the maintenance of cellular integrity and survival when induced in response to environmental, physical and chemical stresses. In the present investigation the effect of environmental stress on HSP90α expression was examined in grey mullet Mugil cephalus living in either a contaminated (Ennore) or uncontaminated (Kovalam) estuary over two seasons: Hepatocytes were isolated from grey mullet of both estuaries. Oxidative stress was determined along with HSP90α in these fish. Additionally, immunohistochemical changes were studied to confirm the HSP90α expression. Comparison of the results revealed enhanced hepatocyte oxidative stress and HSP90α expressions in fish from Ennore to a significant extent than fish from Kovalam. Also, the results showed significant seasonal variations with maximum expression observed during summer compared to the monsoon season. Overexpression of HSP90α in hepatocytes exposed to chronic environmental stress by pollutants may confer differential effects on cell survival by protecting against oxidative stress induced changes. The results also indicate that seasonal variations have significant effect on the HSP90α expression.     

利用90K基因芯片进行小麦株高QTL分析

为给小麦株高标记辅助选择提供可供选择的分子标记,并进一步对株高QTL进行精细定位及相关基因克隆,以小麦骨干亲本周8425B和小偃81衍生的包含102个家系的RIL群体(F_8)为材料,利用90K芯片标记构建高密度遗传图谱,在3个环境下对株高进行QTL检测。结果表明,所构建的图谱含有9 290个SNP标记,覆盖了小麦21条染色体的63个连锁群,图谱总长3 894.64cM,平均标记密度为0.42cM。共检测到9个控制株高的QTL,分布于1B、4A、4D、6B、7A、7B和7D染色体上,变异解释率为2.23%~16.25%。QPh.nafu.4D、QPh.nafu.4A、QPh.nafu.1B-2与前人定位到的位置相同或相近。QPh.nafu.7A具有较大的LOD值(8.17)和变异解释率(14.69%),为主效QTL。QPh.nafu.6B、QPh.nafu.7B-1、QPh.nafu.7B-2均能在多个环境下使用多种QTL检测方法定位到,可能为新的较稳定的控制株高的QTL。
