Effects of Large Saddle Panels on the Biomechanics of the Equine Back During Rising Trot: Preliminary Results

The saddle panels, directly in contact with the horse's back, are likely an important element to optimize the fitting of the saddle, the comfort of the horse, and subsequently, the pain management in dorsalgic horses. The aim of this study was to better understand the effect of the saddle panels on the horse's back, by evaluating a prototype saddle (comfort panels: CP) compared to a standard saddle (STD). The horse's back movements were measured using inertial measurement units (IMUs) fixed at the levels of thoracic vertebrae T6, T12, T16 (under the saddle) and lumbar vertebrae L2 and L5. The centers of mass (COMs) of the horse and the rider and limb's protraction-retraction angles, pressure between saddle and horse's back, and force on the stirrups were measured using respectively 2D motion capture, pressure mat and force sensors in the stirrup leather. Three horses were trotted at the rising trot (sitting: left diagonal-rider seated; standing: right diagonal-rider standing) by the same rider. To compare saddles, linear mixed-effects regression models were used. The estimated means (SE) were calculated. During sitting phase, pressure in the cranial and middle areas of the saddle significantly increased for CP compared to STD (+0.9 (0.2) kPa and +1.0 (0.1) kPa, respectively) whereas caudal pressure decreased (−1.8 (0.4) kPa). Concurrently, the range of motion of angles T12-T16 and T16-L2 under the saddle significantly increased (+1.8 (0.2)° and +2.3 (0.3)°, respectively). The results showed that modifications of the panels' shape not only affect the pressure distribution but also the kinematics of the thoracic and lumbar regions of the equine back.     

Protective efficacy of an H5/H7 trivalent inactivated vaccine produced from Re-11, Re-12, and H7-Re2 strains against challenge with different H5 and H7 viruses in chickens

We developed an H5/H7 trivalent inactivated vaccine by using Re-11, Re-12, and H7-Re2 vaccine seed viruses, which were generated by reverse genetics and derived their HA genes from A/duck/Guizhou/S4184/2017(H5 N6)(DK/GZ/S4184/17)(a clade 2.3.4.4 d virus), A/chicken/Liaoning/SD007/2017(H5 N1)(CK/LN/SD007/17)(a clade 2.3.2.1 d virus), and A/chicken/Guangxi/SD098/2017(H7 N9)(CK/GX/SD098/17), respectively. The protective efficacy of this novel vaccine and that of the recently used H5/H7 bivalent inactivated vaccine against different H5 and H7 N9 viruses was evaluated in chickens. We found that the H5/H7 bivalent vaccine provided solid protection against the H7 N9 virus CK/GX/SD098/17, but only 50–60% protection against different H5 viruses. In contrast, the novel H5/H7 trivalent vaccine provided complete protection against the H5 and H7 viruses tested. Our study underscores the importance of timely updating of vaccines for avian influenza control.     

Zerumbone attenuates MPP+-induced cytotoxicity in human neuroblastoma SH-SY5Y cells by inhibition of oxidative stress

AIM: To investigate the role of zerumbone (ZER) in 1-methyl-4-phenylpyridinium (MPP⁺)-induced cytotoxicity of human neuroblastoma SH-SY5Y cells. METHODS: Human neuroblastoma SH-SY5Y cells were cultured in vitro and the protective effect of ZER against MPP⁺-induced cytotoxicity was measured by CCK-8 assay. Flow cytometry was used to determine the apoptosis and reactive oxygen species (ROS). The expression of Parkinson disease protein 7 (PARK7) was knocked-down by using PARK7-specific short hairpin RNA (shRNA). The protein levels of PARK7, nuclear factor E2-related factor 2 (Nrf2) and heme oxygenase-1 (HO-1) were determined by Western blot. RESULTS: MMP⁺ remarkably reduced the cell viability in a dose-dependent and time-dependent manner. The SH-SY5Y cell injury model was established by treatment with MPP⁺ at 600 μmol/L for 24 h. ZER up-regulated the protein levels of PARK7 and Nrf2 (P<0.05), alleviated apoptosis (P<0.05), and reduced ROS production (P<0.05) in the SH-SY5Y cell injury model. Meanwhile, N-acetyl-L-cysteine (NAC) had the similar functions. Moreover, significant reductions in the protein levels of Nrf2 and HO-1 (P<0.05), and obvious increases in apoptosis (P<0.05) and ROS level (P<0.05) were demonstrated in PARK7-knockdown cells. CONCLUSION: ZER protects SH-SY5Y cells against MPP⁺-induced cytotoxi-city, which may be related to activation of PARK7/Nrf2/HO-1 pathway, and subsequent attenuation of oxidative stress and apoptosis.     

同期排卵处理母牛体温和活动量变化规律及效果分析

为探究同期排卵处理母牛体温和活动量变化规律及不同同期排卵技术处理效果，指导同期排卵技术优化。本研究自动监测了18头20月龄左右同期排卵（GnRH-PG-GnRH）处理荷斯坦母牛和17头产后40~60 d预同期排卵（PG-PG-GnRH-PG-GnRH）处理荷斯坦母牛的体温和活动量，应用自动检测系统进行母牛发情监测。结果发现，同期排卵处理母牛发情时阴道温度平均升高（0.43±0.20）℃，持续（12.37±2.73）h；活动量平均升高（18.28±18.61）倍，持续（11.00±1.68）h；排卵时阴道温度平均下降（0.20±0.10）℃，持续（11.00±1.68）h。自动化发情监测显示，同期排卵处理母牛7头发情并排卵；预同期排卵母牛GnRH处理前全部发情排卵。两种同期排卵处理，虽可改变母牛性周期进程，促进母牛性周期同步化，但均难以使母牛性周期完全同步。因此，将同期排卵-定时输精和发情鉴定技术科学结合才能取得更好的繁殖效果。     

CRISPR/cas系统及其在家禽上的应用研究进展

CRISPR/cas是一种获得性免疫防御系统,广泛存在于细菌和古细菌中。Strep tococcus pyogenes cas9(Spcas9)作为目前研究最多、最清楚的效应蛋白,因其酶活性高和靶点广而被应用于生命科学各个领域,但Spcas9分子量大、脱靶率高等缺点在一定程度上限制了其应用。随着CRISPR系统技术被深入开发,一些新效应蛋白被发现,本文就CRISPR/cas系统分类、原理、新效应蛋白发现及cas9(S.Pyogenes)多种变构体技术开发在及家禽上的应用进行综述,为相关领域的研究提供参考。

交沙霉素等四种药物对雏鸡隐孢子虫病的预防效果

对经口感染(4.0×10~5个/鸡)贝氏隐孢子虫(Cryptosporidium baileyi)卵囊的雏鸡在其饲料中分别添加交沙霉素(4.0g/kg)、复方新诺明(4.9g/kg)、大蒜素(400mgkg/)、甲硝唑(2.0g/ks),任鸡自由采食,连用7d.结果表明,实验鸡潜隐期延长,粪中卵囊数显著降低,体重明显增加,血液学值及血清酶学指标与健康对照鸡无明显差异.由此证明,这4种药物对实验性雏鸡隐孢子虫病有不同程度的预防作用.

导入AGPase基因的转基因可育水稻及其经济性状的研究

以农杆菌介导法将淀粉合成关键酶AGPase(腺嘌呤二磷酸葡萄糖焦磷酸化酶）基因导入云南地方优质水稻合系35，38，39，41，42中，获得了大量的可育转基因植株。PCR扩增检测，外源基因已稳定整合进水稻基因组中。大量的农艺性状及淀粉含量研究表明，转基因水稻种子的千粒重比对照明显提高，但总淀粉含量没有显著变化。
