The activity levels of succinate dehydrogenase (SDH) and glucose‐6‐phosphate dehydrogenase (G6PD) were assessed in various tissues of Cyprinus carpio var communis which had been exposed to lethal concentrations of group‐II pyrethroids (deltamethrin, cypermethrin, fenvalerate and fluvalinate) for a period of 72 h. The results indicated a steady decrease in SDH activity with a concomitant increase in G6PD activity. The decreased SDH activity indicated inhibition of SDH at mitochondrial level and the increased G6PD activity an enhancement of an alternative pathway of carbohydrate metabolism, viz the hexose monophosphate shunt (HMP) or pentose phosphate pathway as a biochemical adaptation to overcome the toxic stress. © 2001 Society of Chemical Industry  相似文献

枇杷果实发育过程中山梨醇含量及相关酶活性的变化   总被引:1,自引:0,他引:1  
以果型和品质差异较大的黄肉枇杷(Eriobotrya japonica Lindl.)‘解放钟’和白肉枇杷‘白梨’的果实为试材,测定果实发育过程中山梨醇、果糖、葡萄糖含量及与山梨醇代谢相关酶--山梨醇脱氢酶(Sorbitol dehydrogenase,SDH)和山梨醇氧化酶(Sorbitol oxidase,SOX)活性的变化,并探讨山梨醇代谢与枇杷果实生长发育和品质形成的关系。结果表明:2个枇杷品种果实中山梨醇含量均随果实发育进程呈下降趋势,且‘白梨’枇杷果实中山梨醇含量高于‘解放钟’。在花后35~80 d,2个枇杷品种幼果中葡萄糖和果糖含量均较低且相对稳定,而花后95~140 d果实中葡萄糖和果糖含量逐渐上升,并在花后125~140 d果实成熟期急剧增加而达到峰值;果实中葡萄糖含量在2个品种间差异不显著(p0.05),花后125~140 d的果实成熟期,‘白梨’果实中果糖含量显著高于‘解放钟’(p0.05)。果实发育过程中SDH和SOX的活性均呈先降后升的变化趋势。说明2个枇杷品种果实发育过程中山梨醇、果糖、葡萄糖含量及SDH和山SOX的活性表现出相似的变化规律,但果实山梨醇含量与SDH和SOX的活性在果实膨大期均存在显著差异(p0.05),这可能是黄肉枇杷与白肉枇杷品质差异的关键。  相似文献

为研究萨福克羊×杜泊羊×湖羊杂交羊(萨杜湖杂交羊)生长发育情况及其对当地生态环境的适应能力,采用生产性能常规测定、显微测定和全自动血液生理生化分析仪分别测定了不同年龄阶段萨杜湖杂交羊的体重、体尺指标、羊毛品质(包括不同部位羊毛的净毛率、细度、有髓毛和无髓毛占比、自然长度和伸直长度等指标)和成年羊血液生理生化指标。结果表明:萨杜湖杂交羊早期生长速度较快,初生阶段及周岁时公羊体斜长均显著高于母羊(P<0.05);3月龄时公羊体高、体斜长和胸围均显著高于母羊(P<0.05);6月龄时母羊体重极显著高于公羊(P<0.01),成年时期公羊体重和体高均显著高于母羊(P<0.05);其余各生长阶段内,公母羊间体重及体尺差异不显著(P>0.05)。其成年时平均净毛率较低,为45.21%;长度适中:平均自然长度为4.55cm,伸直长度为6.22cm;细度良好,为17.60μm。公母羊血液生理生化指标及其同国内绵羊参考值相比存在一定的差异,其中母羊血小板总数(PLT)和血小板压积(PCT)均极显著高于公羊(P<0.01),公羊甘油三酯(TG)和白蛋白(ALB)含量显著高于母羊(P<0.05),其余各项指标均无显著差异(P>0.05)。综上所述,萨杜湖杂交羊在生长发育等方面都有不同程度的改进和提高,本试验结果可为该品种杂交羊进一步开发利用提供参考依据。  相似文献

试验旨在研究有机磷农药氧化乐果对雄性小鼠睾丸琥珀酸脱氢酶(SDH)和精液质量的影响作用。将40只健康昆明雄性小鼠随机分为4组,即低、中、高3个染毒组和对照组,其中染毒组分别灌胃1.0、2.0、4.0 mg/kg氧化乐果,对照组灌胃生理盐水,采用等容量灌胃法(0.2 mL/10 g体重),连续灌胃14 d。随着氧化乐果染毒剂量的逐渐增加,小鼠睾丸琥珀酸脱氢酶(SDH)活力、精子密度、存活率均明显降低,与对照组相比差异显著(P<0.05);小鼠体重、睾丸重、附睾及其脏器系数与对照组相比差异均不显著(P>0.05)。氧化乐果对小鼠精子密度和精子存活率有一定的损伤作用,对睾丸琥珀酸脱氢酶有明显的抑制作用。  相似文献

以鲫鱼Carassius cuvieri未成熟的卵细胞为材料,研究了玻璃化冻存对鲫鱼卵细胞的存活率及琥珀酸脱氢酶(SDH)活性的影响。结果表明:鲫鱼卵细胞存活率在1~6 d下降明显,6~8 d下降趋势减弱,保持相对稳定;琥珀酸脱氢酶活性呈显著下降(P<0.05)。玻璃化冻存液VSd组的冻存效果最好,其组分为180.0 g·L-1 1,2-丙二醇,110.0 g·L-1甲醇,0.1 mol·L-1 海藻糖和40.0 g·L-1 聚乙二醇。该玻璃化冻存液配方及方法在一定时间内保存鲫鱼未成熟卵细胞具有一定的应用价值。  相似文献

Responses of the major mitochondrial respiratory enzyme succinate dehydrogenase (SDH) activity in gill and muscle tissue of tilapia (Oreochromis mossambicus) were investigated upon their transfer to a new pond habitat. Sixty healthy adult fish were procured from each of the three ponds used and were distributed in three cages in equal numbers; the cages were placed in each of the other two ponds as well as into original rearing pond. Four fish were removed from each cage every week and were sacrificed for the assay of SDH activity in gills and muscles. Transfer of fish from a stress condition to a benign environment and vice versa resulted in an increase and decrease in SDH activity of fish, respectively. The intensity of increase upon transfer was again dependent upon the amount of stress in the new habitat. In general, the SDH activity of the test fish strongly correlated with the ammonia concentration (P?<?0.05) of water regardless of habitats. Time required for the introduced fish to acclimatize with new environmental conditions was dependent upon the environmental milieu of both its initial and final place of rearing and hence their physiological states. Differences in water quality, especially ammonia concentration, between its original place of rearing and the place of transfer were found to be responsible for the differences in acclimation period of two or three weeks. Relatively less difference in ammonia (2.68 for gills and 3.20 for muscles) between the initial place of rearing and new habitat resulted in acclimation of the fish one week earlier (second week) than the relatively wide difference (4.46 for gills and 5.62 for muscles) for third week. It may be reasonable to conclude that the varied responses of the mitochondrial respiratory enzyme of exogenously introduced fishes with the differences in the water quality—especially the ammonia concentration of ambient water between the original fish holding pond and growout pond—can be used to predict the time that will be required for the exogenously introduced fish to fully acclimatize with the new habitat.  相似文献

Testosterone (T) administration to maleAnabas testudineus significantly stimulated the activities of cytochrome oxidase, -glycerophosphate dehydrogenase (-GPDH) and Mg2+ adenosine triphosphatase (Mg2+ ATPase) and inhibited lactate dehydrogenase (LDH), cytosolic and mitochondrial malate dehydrogenases (MDH). The activities of succinate dehydrogenase (SDH), glucose-6-phosphate dehydrogenase (G-6-PDH) and catalase were unaffected by testosterone treatment. Administration of estradiol-17 (E2) in female fish, significantly stimulated cytochrome oxidase activity, inhibited Mg2+ ATPase, SDH, catalase and cytosolic and mitochondrial MDH activity, and was without effect on other enzymes studied.The simultaneous injections of actinomycin D or chloramphenicol and T or E2 prevented the hormonal influence on hepatic enzyme activities. The present study demonstrates that inA. testudineus sex steroids influence hepatic oxidative metabolism by a mechanism sensitive to the action of inhibitors of protein synthesis.  相似文献

小麦是世界主要粮食作物之一,但籽粒赖氨酸含量低,影响其营养品质。为了通过生物技术提高小麦籽粒中赖氨酸含量,选用小麦LKR-SDH基因靠近3'端385 bp的保守序列,以水稻pGluB为启动子,拟南芥FAD2第一内含子为间隔序列,pUC18为中间栽体,构建LKR-SDH基因的hpRNA表达盒。然后将hpRNA表达盒插入到pAHC25的HindⅢ酶切位点上,成功构建小麦LKR-SDH基因RNAi植物表达载体pAHC25-TaLKR/SDH-RNAi。通过限制性内切酶酶切鉴定,表明pAHC25-TaLKR/SDH-RNAi载体构建正确。进一步采用基因枪法将其转入到扬麦158中,获得了部分小麦转化植株。旨在为小麦高赖氨酸品种的培育提供候选材料。  相似文献

对玻璃化冻存后羊Ovis aries软骨细胞的存活率和琥珀酸脱氢酶(SDH)活性进行了初步研究。结果表明:细胞存活率随着冻存时间的延长而逐渐下降,但下降速度不明显;SDH活性在玻璃化冻存1 ~ 20 d变化幅度较大,20 ~ 30 d下降趋势减弱,保持相对稳定。从各项指标来看,认为玻璃化溶液VSb组的冻存效果最好,在经过5,10, 15,20和30 d的冻存后,其存活率分别为85.47% ± 1.78%,80.73% ± 1.81%,78.62% ± 2.06%, 76.35% ± 2.58%和73.83% ± 1.49%,细胞SDH活性吸光度值分别为0.49 ± 0.064,0.444 ± 0.073,0.394 ± 0.039,0.354 ± 0.082和0.339 ± 0.053。该玻璃化冻存方法对长期冻存羊软骨细胞具有一定的实际应用意义。图4表2参17  相似文献

The aim of the present study is to investigate the protective effect of bilberry extract on liver damage in restraint stressed mice. A remarkable increase in alanine aminotransferase (ALT) and reactive oxygen species (ROS) levels was observed in stressed mice. Treatment with bilberry extract restored ALT and ROS to normal levels, and enhanced mitochondrial complex II activity that was lowered in restraint stressed mice. The mitochondrial electron transfer chain (ETC)-related gene expression was measured by RT-PCR, and a significant up-regulation of complex II mRNA was observed for SDHA, B, C and D mRNA in bilberry extract-treated group compared with that in stressed group. Bilberry extract administration also profoundly elevated the Na+-K+-ATPase activity and mitochondria membrane potential (?Ψm), which was reduced in the stressed group. Bilberry extract exhibited protective effect by scavenging free radicals and attenuating mitochondrial dysfunction in the liver of restraint stressed mice. It may be used as a promising therapeutic agent in preventing and delaying the life-related disease.  相似文献
