目的研究冻存对内皮生长晕细胞增殖能力和成血管能力的影响,探讨内皮生长晕细胞冻存和复苏的可行性。方法分离脐血中单个核细胞,采用贴壁培养法培养扩增内皮生长晕细胞,免疫组织化学染色和荧光染色法鉴定其内皮细胞特性。扩增后的细胞采用浓度为650μmol/L(体积比为50mL/L)和1040μmol/L(体积比为80mL/L)二甲基亚砜的培养基冻存至液氮中,24h后复苏并观察冻存细胞的复苏率、复苏后细胞的增殖能力和成血管能力的改变。结果采用贴壁法培养的细胞具有多种内皮细胞特性。细胞采用含不同浓度二甲基亚砜(50mL/L和80mL/L)的培养基冻存后,其复苏率差异无统计学意义,细胞的增殖能力在复苏后36h内50mL/L、80mL/L二甲基亚砜组均较未冻存组减弱(P<0.05),72h后三组间比较差异无统计学意义。而复苏后6h内成血管速率较未冻存组减弱(P<0.05),但30h后三组间比较差异无统计学意义。结论内皮生长晕细胞可以进行冻存和复苏。  相似文献

BACKGROUND: Small volume resuscitation has been advocated as a beneficial therapy for endotoxemia in horses but this therapy has not been investigated in a prospective manner. The objective of this study was to determine the cardiopulmonary effects of small-volume resuscitation using hypertonic saline solution (HSS) plus Hetastarch (HES) during experimental endotoxemia in anesthetized horses. HYPOTHESIS: Treatment of horses with induced endotoxemia using HES-HSS does not alter the response of various cardiopulmonary indices when compared to treatment with either small- or large-volume isotonic crystalloid solutions. ANIMALS: Eighteen healthy horses were randomly assigned to 1 of 3 groups. Anesthesia was maintained with halothane. Endotoxemia was induced by administering 50 microg/kg of Escherichia coli endotoxin IV. The horses were treated over 30 minutes with 15 mL/kg of balanced polyionic crystalloid solution (control), 60 mL/kg of balanced polyionic crystalloid solution (ISO), or 5 mL/kg of HSS followed by 10 mL/kg of HES (HSS-HES). METHODS: Prospective randomized trial. RESULTS: Cardiac output (CO) after endotoxin infusion increased significantly (P < .05) from baseline in all groups, whereas mean central venous pressure increased significantly (P < .05) in the ISO group only. Mean pulmonary artery pressure increased from baseline (P < .05) in horses treated with isotonic fluids and HSS-HES. There was no effect of treatment with HSS-HES on CO, systemic vascular resistance (SVR), mean arterial pressure, blood lactate concentrations, or arterial oxygenation. CONCLUSIONS AND CLINICAL IMPORTANCE: The use of HSS-HES failed to ameliorate the deleterious hemodynamic responses associated with endotoxemia in horses. The clinical value of this treatment in horses with endotoxemia remains unconfirmed.  相似文献

AIM: To investigate the effect of scutellarin on resuscitating rat non-heart-beating marginal donor (NHBMD) livers with extracorporeal liver perfusion in vitro. METHODS: Twenty-four male Wistar rats were randomly divided into 3 groups: group A (n=8), the livers were subject to 15~20 min of warm ischemia; group B (n=8) and group C (n=8), the livers were subject to 45 min of warm ischemia. All the livers were treated by extracorporeal liver perfusion (ECLP), and scutellarin solution was add to the perfusion fluid in group C with the intervals of 1 h. The data of bile production and the markers of hepatocyte reperfusion injury of the livers,alanine aminotransferase (ALT) and endothelin-1 (ET-1), in each group were tested. The liver tissues in each group were obtained before perfusion and 1 h, 3 h and 6 h after perfusion, and then observed under transmission electron microscope to investigate the changes of ultrastructures. RESULTS: With the time of perfusion went on, the livers in group B and group C demonstrated recovery of synthe-tic function after 3 h and 6 h of perfusion, and the production of bile in group B and group C was not significantly different from that in group A after 6 h of perfusion (P>0.05). The levels of ALT and ET-1 in group B and group C were significantly higher than those in group A after 1 h, 3 h and 6 h of perfusion (P<0.05), and those in group C was significantly lower than those in group B after 3 h and 6 h of perfusion (P<0.05). The ultrastructural destruction of the liver cells in both group B and group C was more serious than that in group A. After 3 h and 6 h of perfusion, the destruction of the liver cells was partly recovered, and the recovery degree in group C was more obvious than that in group B. CONCLUSION: Scutellarin may play a significant role in resuscitating rat NHBMD livers with extracorporeal liver perfusion in vitro.  相似文献

[目的]探索降解毒死蜱的新的微生物资源.[方法]应用最大或然数(MPN)法,通过添加适量复苏促进因子(Rpf)从多种土壤及污水生物处理系统中分离活的非可培养(VBNC)资源菌种,根据文献报道的已知毒死蜱降解菌株的系统关系综合分析,初步筛选出与已知降解毒死蜱菌种近缘的VBNC菌株并进行降解试验.[结果]共筛选出6株具有毒死蜱降解能力的菌株,其中菌株CHZYR63对毒死蜱的生物降解能力最强,经Box-behnken响应面法优化后降解率可达70.15%.[结论]可培养化VBNC菌种资源在残留农药降解方面有较大的利用价值,为揭示受染环境中VBNC微生物的形成及其活性化、环境污染物质的共代谢等微生物群体的相互作用机制提供了参考.  相似文献

Background: Central venous pressure (CVP) is used in many species to monitor right‐sided intravascular volume status, especially in critical care medicine. Hypothesis: That hypohydration in adult horses is associated with a proportional reduction in CVP. Animals: Ten healthy adult horses from the university teaching herd. Methods: In this experimental study, horses underwent central venous catheter placement and CVP readings were obtained by water manometry. The horses were then deprived of water and administered furosemide (1 mg/kg IV q6h) for up to 36 hours. Weight, CVP, vital signs, PCV, total protein (TP), and serum lactate were monitored at baseline and every 6 hours until a target of 5% decrease in body weight loss was achieved. The spleen volume was estimated sonographically at baseline and peak volume depletion. Linear regression analysis was used to assess the association of CVP and other clinical parameters with degree of body weight loss over time. Results: There was a significant association between CVP and decline in body weight (P < .001), with a decrease in CVP of 2.2 cmH2O for every percentage point decrease in body weight. Other significant associations between volume depletion and parameters measured included increased TP (P= .007), increased serum lactate concentration (P= .048), and decreased splenic volume (P= .046). There was no significant association between CVP and vital signs or PCV. Conclusions and Clinical Importance: These findings suggest that CVP monitoring might be a useful addition to the clinical evaluation of hydration status in adult horses.  相似文献
