AIM: To investigate the effects of maternal limb ischemic preconditioning (LIP) on the mitochondrial structures and functions of the hippocampal neurons induced by reoxygenation in the intrauterine distress fetal rats. METHODS: Pregnant rats (n=40) were randomly divided into 4 groups: sham (S) group, LIP group, fetal distress (FD) group and LIP+FD group. Intrauterine ischemia model was established through the experimental design. The ultrastructure of the mitochondria in CA1 area of the hippocampus was observed. The mitochondrial membrane potential and reactive oxygen species (ROS) were measured. The content of ATP and MDA in the hippocampus tissue was detected. The activity of Mn-SOD was observed. RESULTS: Compared with sham group, the ultrastructure of mitochondria in CA1 area of the hippocampus was damaged in FD group and LIP+FD group. The mitochondrial membrane potential, the content of ATP and the activity of Mn-SOD were decreased. However, the content of ROS and MDA was increased. Compared with FD group, the ultrastructure of mitochondria in CA1 area of the hippocampus was intact in LIP+FD group. Furthermore, the reduced mitochondrial membrane potential and ATP content were inhibited. The activity of Mn-SOD was increased, but the content of ROS and MDA was decreased in LIP+FD group. CONCLUSION: Limb ischemia preconditioning inhibits the damage the mitochondria of fetal hippocampal neurons induced by reoxygenation in the intrauterine distress fetal rats.  相似文献

AIM:To investigate the effects of electroacupuncture (EA) pretreatment on survival, brain injury and cognitive function in rats after limb ischemia/reperfusion (LI/R). METHODS:One hundred and thirty-two healthy male SD rats weighing 255~300 g were randomly divided into 3 groups (n=44 each):sham operation group (sham), LI/R group and LI/R plus EA pretreatment (IL/R+EA) group. The LI/R model was made by a method that the bilateral femoral arteries were occluded for 3 h with atraumatic microclips followed by 48 h of reperfusion. In sham group, sham operation was performed. The EA pretreatment was conducted twice a day for 14 d prior to the LI/R event. EA pretreatment included the following acupoints:Baihui (GV20), Zusanli (ST36) and Xuehai (SP10). The survival rate within 7 d following LI/R was calculated. The changes of cognitive function were detected 48 h after reperfusion using Morris water maze test. The cerebral water content was determined by detecting the wet and dry weight. Microglial cells were evaluated following immunolabeling of Iba1 (a marker of microglia). The protein level of cleaved caspase-3 in the hippocampus was measured by Western blotting. The neuronal apoptosis was detected using TUNEL method. Meanwhile, the changes of pathological structure in hippocampus were observed under light microscope. The activity of MPO and SOD, and the content of ROS and MDA were also investigated. RESULTS:Compared with sham group, the Iba1 positive cells, the protein level of cleaved caspase-3, the apoptotic index, and the levels of ROS/MDA and MPO activity in LI/R and LI/R+EA groups increased significantly. The normal hippocampal neurons reduced, and SOD activity decreased significantly in hippocampus. The survival rates of the rats within 7 d decreased, the latency and swimming distance increased, and the number of crossing the platform reduced. Compared with LI/R group, the above indexes in LI/R+EA group were markedly improved. CONCLUSION:EA stimulation improves the survival rate and cognitive dysfunction, and reduces brain damage in LI/R rats by preventing microglial activation and attenuating oxidative stress.  相似文献

The prevalence of capped hock in 5601 post-weaning pigs from 93 pig farms in England was 17.2%. The prevalence increased with age. Once adjusted for age, the lowest prevalence of capped hock was observed in pigs kept on soil floors (usually covered with deep straw bedding). There was no significant increase in the risk of capped hock in pigs kept on solid concrete floors with deep straw bedding. However, pigs kept on solid concrete with some, or the entire pen, sparsely bedded and pigs kept on partially or fully slatted floors had an approximately threefold increased risk of capped hock. This did not vary significantly between these four floor types. This was in contrast to the associated risks for bursitis in the same pigs, where as the floor went from highly resilient (straw and solid floors) to hard and perforated (fully slatted) the risk of bursitis increased in a similar way to a dose response. No other variables that were measured were associated with a change in risk for capped hock, while observation of pigs slipping or slip marks and wet, dirty and worn pens were also associated risks for bursitis. These results indicate that capped hock and bursitis are both affected by exposure to floors, but in different ways. The prevalence of capped hock was associated only with floor hardness, with deep straw protecting the pigs, while bursitis was associated with both changes in bedding depth (hardness), floor material (soil versus concrete) and floor construction (solid versus slatted floors) and in factors associated with locomotion (slipping and slip marks). These results indicate that the aetiology of capped hock and bursitis might differ.  相似文献

Background

Many chemotherapy protocols have been reported for treatment of canine appendicular osteosarcoma (OSA), but outcome comparisons in a single population are lacking.

Objective

To evaluate the effects of protocol and dose intensity (DI) on treatment outcomes for carboplatin and doxorubicin‐based chemotherapy protocols.

Animals

Four hundred and seventy dogs with appendicular OSA.

Methods

A retrospective cohort study was performed comprising consecutive dogs treated (1997–2012) with amputation followed by 1 of 5 chemotherapy protocols: carboplatin 300 mg/m2 IV q21d for 4 or 6 cycles (CARBO6), doxorubicin 30 mg/m2 IV q14d or q21d for 5 cycles, and alternating carboplatin 300 mg/m2 IV and doxorubicin 30 mg/m2 IV q21d for 3 cycles. Adverse events (AE) and DI were evaluated. Kaplan–Meier survival curves and Cox proportional hazards regression were used to compare disease‐free interval (DFI) and survival time (ST) among protocols.

Results

The overall median DFI and ST were 291 days and 284 days, respectively. A lower proportion of dogs prescribed CARBO6 experienced AEs compared to other protocols (48.4% versus 60.8–75.8%; P = .001). DI was not associated with development of metastases or death. After adjustment for baseline characteristics and prognostic factors, none of the protocols provided a significant reduction in risk of development of metastases or death.

Conclusions and Clinical Importance

Although choice of protocol did not result in significant differences in DFI or ST, the CARBO6 protocol resulted in a lower proportion of dogs experiencing AEs, which could be advantageous in maintaining high quality of life during treatment. DI was not a prognostic indicator in this study.  相似文献

This article describes the lower limb and hoof conformation of a population of semi-feral Mongolian horses living on an open tundra/steppe environment. Data were collected from a convenience sample of 120 Mongolian horses used in the 2011 Mongolian Derby. Digital images of the hooves were obtained, and the lower limb conformation was assessed by four veterinarians involved in the screening of the horses offered for the derby. The horses were predominantly geldings (96%, 100/104), approximately 8.6 ± 2.5 years old, and 137 ± 8 cm at the withers. None of the horses were subjected to routine hoof trimming. Based on a 7-point linear score, lower limb conformation was normal, with a trend (>1 linear score deviation) slightly toward carpal valgus, mildly offset cannon (third metacarpal), and valgus at the matacarpophalangeal joint. Hoof measurements were within the norm for horses of this size. Fetlock valgus was associated with a smaller hoof width:length ratio (P = .016). None of the other hoof measurements were significantly associated with abnormal conformation scores. Overall, few conformation abnormalities were observed, and hoof shape and size was within the normal expected range for horses of this size. The hoof conformation in this population of Mongolian horses represented the natural interaction of the hoof with the environment.  相似文献

Dogs suffering from Golden Retriever muscular dystrophy (GRMD) present symptoms that are similar to human patients with Duchenne muscular dystrophy (DMD). Phenotypic variability is common in both cases and correlates with disease progression and response to therapy. Physical therapy assessment tools were used to study disease progression and assess phenotypic variability in dogs with GRMD. At 5 (T0), 9 (T1), 13 (T2) and 17 (T3) months of age, the physical features, joint ranges of motion (ROM), limb and thorax circumferences, weight and creatine kinase (CK) levels were assessed in 11 dogs with GRMD. Alterations of physical features were higher at 13 months, and different disease progression rates were observed. Passive ROM decreased until 1 year old, which was followed by a decline of elbow and tarsal ROM. Limb and thorax circumferences, which were corrected for body weight, decreased significantly between T0 and T3. These measurements can be used to evaluate disease progression in dogs with GRMD and to help discover new therapies for DMD patients.  相似文献
