Development of a rapid assay for the diagnosis of Myxobolus cerebralis in fish and oligochaetes using loop-mediated isothermal amplification 总被引:1,自引:0,他引:1
A loop-mediated isothermal amplification assay was developed for the rapid detection of Myxobolus cerebralis in both fish and oligochaete hosts. The assay was optimized to amplify parasitic DNA by incubation with Bst DNA polymerase and a set of six specially constructed primers at 65 degrees C for 60 min. The amplification products were detected visually using SYBR Green I dye which gave identical results to gel electrophoresis analysis. Parasite DNA was detected from infected oligochaetes, and from the anal fin, caudal fin, dorsal fin and operculum of clinically infected fish. This 'Myxo-LAMP' assay has a detection limit similar to that of a polymerase chain reaction assay (10(-6)), but is more rapid and only requires a water bath for amplification and is therefore practical for simple and rapid diagnosis of infected tissue. 相似文献

Raw green gram contains trypsin inhibitor (5.6 × 10–3 units/0.1 g) while cooking destroyed the trypsin inhibitor in green gram. No destruction was observed in the case of lysine, methionine, cystine, threonine and tryptophan. Sulphur containing amino acids have been found to be the only limiting amino acids in green gram from studies on blood levels of amino acids and PER experiments. Raw green gram, when fed as a sole source of protein promoted an extremely low growth rate, (13.2 g/4 weeks) and PER (0.77) in rats. Cooking slightly improved the nutritive value of the gram, as indicated by increase in gain in body weight (24 g/4 weeks) as well as PER (1.22). The presence of inhibitors in green gram did not appear to affect some of the essential amino acids as observed from the blood amino acid levels. 相似文献

Identifying and quantifying the links between water resources and food production is crucial in addressing the intensified conflicts between water scarcity and food security. We proposed an integrated framework for quantifying relationships between food and water based on the concept of green water (GW), blue water (BW) and crop water productivity (CWP). An estimation method coupling hydrologic model and crop and water statistics was developed and validated to quantify basin-scale GW, BW and CWP in breadbasket basins of China. A basin-scale GW and BW assessment method was developed by using the Soil and Water Assessment Tool (SWAT). Monthly-step calibration and validation were performed at 15 discharge flow stations in seven first-order river basins of the country. The coefficient of determination (r2) and Nash-Sutcliffe Efficiency (NSE) in calibration stage ranged from 0.18 to 0.95, and −4.22 to 0.93, respectively; while in validation period, r2 ranged from 0.02 to 0.97 and NSE ranged from −266.7 to 0.96. The simulated available soil water was validated against the observed soil moisture data, and the results showed that the model can reflect the yearly average values of soil water storage. Overall, the modeling performance for river basins with 4.94 million km2 of drainage areas in total was acceptable. The simulated hydrologic components were then coupled with crop-and-water-statistics-based estimation method for assessing basin-scale CWP on four staple grain crops, i.e. rice, wheat, maize, and soybean. The results were validated by comparing with the similar investigations in China and around the globe. Fogging with formaldehyde of citrus packinghouses when the fruit are absent is a practice to control conidia of Penicillium digitatum (Pers.) Sacc., the cause of citrus green mold. Replacements for formaldehyde in these facilities are needed because of worker and environmental health issues. To evaluate the effectiveness of candidate sanitizers, craft wood sticks with conidia of P. digitatum were attached throughout commercial citrus ethylene degreening rooms and either water alone or the sanitizers were applied. The rooms were 20 ± 2 °C and humidified to 85–90% relative humidity. Aldehydes, peroxygen compounds, sodium hypochlorite, chlorine dioxide, quaternary ammonium, alcohols, one phenolic compound, and one organic acid were applied with a compressed air assisted atomizer or fan atomizer in a volume of approximately 6 L per 100 m3 of room volume dispensed over a 3 h period. Rates applied were expressed as active ingredient per m3 of room volume. All were compared to formaldehyde applied at 1.98 g m−3 of room volume. After 24 h, the craft wood sticks were retrieved, and germination of the conidia assessed. Five sanitizers reduced germination of conidia by more than 95% and equaled formaldehyde in effectiveness. They were (effective rates): (1) glutaraldehyde (0.1 g m−3); (2) hydrogen peroxide (4.4 g m−3); (3) Citrisol (1.0 g m−3), a proprietary mineral oxychloride oxidizer; (4) acetic acid (5.3 g m−3); and (5) peracetic acid (2.4 g m−3). The toxicity of effective sanitizers was determined by exposure of P. digitatum conidia for 10 min to concentrations of each and the proportion of survivors used to estimate EC50 and EC99 concentrations. The toxicity of the sanitizers in this assay did not predict their effectiveness when applied by fogging, probably because other factors, such as distribution, persistence, droplet size, or vapor pressure also influenced their effectiveness. 相似文献

Practices that minimize the rate of soil degradation, increase crop yields and raise farm income are key to sustaining agricultural productivity in the hills of Nepal. The use of farmland is undergoing rapid changes in response to increasing population pressure, deforestation and subsistence needs. Against this background, this study examined the impact of an agroforestry intervention project on farm income based on a sample of subsistence farm households in Dhadhing district. The project was implemented by Nepal Agroforestry Foundation in 1993/94 to increase fodder production through the promotion of agroforestry. A total of 223 households (82 with project and 141 without project) were interviewed during May–October 1998 to collect information on production and agroforestrys' impact on farm income. The benefit-cost analysis showed that the agricultural system including agroforestry was more profitable than the conventional one. The results also showed that the introduction of mulberry trees for sericulture could further enhance the profitability of an agroforestry-based system. Thus, agroforestry has great potential for enhancing food production and farmers' economic conditions in a sustainable manner through its positive contributions to household income.This revised version was published online in November 2005 with corrections to the Cover Date. 相似文献

The effects of two pruning regimes on the above-ground biomass allocation and nodulation of Gliricidia sepium (Jacq.) Walp. (Leguminosae: Robinieae) were studied in a cut-and-carry forage production system under humid tropical conditions in Guadeloupe, French Antilles. The grass layer composed of a mixture dominated by Paspalum notatum Flügge (80%) and Digitaria decumbens Stent. The pruning regimes were partial pruning (ca. 50%) every two months and complete pruning every six months. The complete pruning caused an almost complete turnover of N2 fixing nodules. The nodule biomass decreased after the partial pruning, but the turnover was not complete. The nodule to foliage biomass ratio followed the same pattern under both treatments, and the values of the ratio converged towards the end of the experimental period. The maxima of standing nodule biomass were 7.2 and 13.0 kg ha−1 in the partially and completely pruned trees, respectively. The cumulative leaf fodder harvest was higher under partial pruning management, due to smaller litter loss. The branch biomass production was higher under complete pruning management. Grass production was not affected by the pruning pattern of G. sepium. It was concluded that the partial pruning management produces more fodder in the studied association, and the nodulation probably adjusts to the canopy N requirements. The potential N release to soil in the turnover of nodules of G. sepium (max. 0.82 kg ha−1) is negligible compared to the N export in tree and grass fodder harvest, 190 and 215 kg ha−1 in partially and completely pruned plots, respectively. This paper examines the first-best instruments for biodiversity maintenance in commercial boreal forests when landowners behave either in Faustmannian or Hartmanian way. Using an extended Hartman model, we show that biodiversity conservation requires both prolonged rotation age and leaving retention trees. While the former promotes some old growth species, the latter create new structural elements of decaying and dead wood, which can sustain a variety of saprolyxic species. A fully synchronized combination of retention tree subsidy and tax instrument is needed both to lengthen the privately optimal rotation period and to provide an incentive to leave retention trees. Using Finnish data we illustrate empirically the sizes of instruments. When combined with a harvest tax, the retention tree subsidy is 1000 and 750 € in the Faustmann and in the Hartman model, respectively. When used with a timber subsidy or a site value tax, the retention tree subsidy is 1700 €/ha in both models. The harvest tax rate varies over the range 40–65% in the Faustmann model and 20–40% in the Hartman model, while timber subsidy is between 0.5–1.0% and site value tax is about 1.75%. 相似文献

Vacant land, a product of population and economic decline resulting in abandonment of infrastructure, has increased substantially in shrinking cities around the world. In Cleveland, Ohio, vacant lots are minimally managed, concentrated within low-income neighborhoods, and support a large proportion of the city’s urban forest. We quantified abundance, richness, diversity, and size class of native and exotic tree species on inner-city vacant lots, inner-city residential lots, and suburban residential lots, and used i-Tree Eco to model the quantity and economic value of regulating ecosystem services provided by their respective forest assemblages. Inner-city vacant lots supported three times as many trees, more exotic than native trees, and greater tree diversity than inner-city and suburban residential lots, with the plurality of trees being naturally-regenerated saplings. The urban forest on inner-city vacant lots also had two times as much leaf area and leaf biomass, and more tree canopy cover. The quantity and monetary value of ecosystem services provided by the urban forest was greatest on inner-city vacant lots, with exotic species contributing most of that value, while native taxa provided more monetary value on residential lots. The predominately naturally-regenerated, minimally managed exotic species on vacant land provide valuable ecosystem services to inner-city neighborhoods of Cleveland, OH. 相似文献

ABSTRACTThe present study reports the biochemical composition of Perna viridis from the southwestern coast of India. A balanced essential to nonessential amino acid ratio (0.7:1.1) along with the optimum and balanced quantities of vitamins, mineral nutrients, and low cholesterol contents characterized P. viridis. The n-3/n-6 polyunsaturated fatty acid ratio of P. viridis was found to be 3.7:5.3 and therefore may serve as an alternative to balance the higher intake of n-6 fatty acids. High oxyradical scavenging capacity (24–32%) and total phenolics (2–5 mg GAE?1) suggest the nutraceutical potential of P. viridis. 相似文献
