The pharmacokinetics of difloxacin (Dicural) was studied in a crossover study using three groups (n = 4) of male and female Friesian calves after intravenous (i.v.), intramuscular (i.m.) and subcutaneous (s.c.) administrations
of 5 mg/kg body weight. Drug concentration in plasma was determined by high-performance liquid chromatography using fluorescence
detection. The plasma concentration–time data following i.v. administration were best fitted to a two-compartment open model
and those following i.m. and s.c. routes were best fitted using one-compartment open model. The collected data were subjected
to a computerized kinetic analysis. The mean i.v., i.m. and s.c. elimination half-lives (t1/2β) were 5.56 ± 0.33 h, 6.12 ± 0.42 h and 7.26 ± 0.6 h, respectively. The steady-state volume of distribution (Vdss) was 1.12 ± 0.09 L/kg and total body clearance (ClB) was 2.19 ± 0.1 ml/(min. kg). The absorption half lives (t1/2ab) were 0.38 ± 0.027 h and 2.1 ± 0.09 h, with systemic bioavailabilities (F) of 96.5% ± 6.4% and 84% ± 5.5% after i.m. and s.c. administration, respectively. After i.m. and s.c. dosing, peak plasma
concentrations (Cmax) of 3.38 ± 0.13 μg/ml and 2.18 ± 0.12 μg/ml were attained after (tmax) 1.22 ± 0.20 h and 3.7 ± 0.52 h. The MIC90 of difloxacin for Mannheimia haemolytica was 0.29 ± 0.04 μg/ml. The AUC/MIC90 and Cmax/MIC90 ratios for difloxacin following i.m. administration were 120 and 11.65, respectively and following s.c. administration were
97.58 and 7.51, respectively. Difloxacin was 31.7–36.8% bound to calf plasma protein. Since fluoroquinolones display concentration-dependent
activities, the doses of difloxacin used in this study are likely to involve better pharmacodynamic characteristics that are
associated with greater clinical efficacy following i.m. administration than following s.c. administration. 相似文献
The statistical analysis of hormones sampled throughout the production cycle is complicated because factors such as age and weight at the measuring date interfere. Spline curves constructed from pieces of low-degree, random-effects polynomials could be used for a more accurate analysis of data. Concentration of insulin-like growth factor-1 (IGF-1), weight gain, and concentrate intake of Parda de Montaña (PM) (n = 27) and Pirenaica calves (n = 14) were modeled with a spline model according to age at weaning, pre-weaning concentrate feeding, and breed. At birth, calves were randomly assigned to early weaning (EW) at 90 d or traditional weaning (TW) at 150 d. During lactation, half of PM calves received concentrates (S), whereas the remainder received no concentrates (NS). After weaning, calves received concentrates on an ad libitum basis until they reached a weight of 450 kg. The spline model had better likelihood than a polynomial of 6 degrees or a split-plot model. Serum IGF-1 concentration was greatly affected by age at weaning and pre-weaning concentrate feeding, but not by breed. In NS calves, IGF-1 concentration was greater in EW than in TW calves from 120 to 300 d, irrespective of breed. During lactation, S calves had greater IGF-1 concentration than NS calves. After weaning, EWNS calves reached the IGF-1 concentration of EWS calves after 4 mo on concentrates, whereas TWNS calves attained IGF-1 concentration of TWS calves after only 2 mo, because of their increased concentrate intake relative to TWS calves. Concentration of IGF-1 was positively correlated with the immediate weight gains and intake, but it was not a good predictor of performance in the long term. 相似文献
This study was conducted from 1992-2002 on 65,534 individual milk records and fat percentage in 473 herds, in six provinces of Khuzestan, Mazndaran, W. Azerbaijan, E. Azerbaijan, Ardabil and Gilan. The data was analyzed by SAS sotfware using GLM procedure. The heritability and breeding value of 1,195 animals were calculated by DFRML procedure. The average milk yield per lactation, days of lactation, fat percentage and LSM of fat percentage were 1,513 kg, 202 days, 5.04 and 6.77, respectively. The estimated heritability of milk was 0.16. The LSM of average milk production in the provinces of Gilan, Mazandaran, E. Azerbaijan, W. Azerbaijan, Khuzestan and Ardabil were: 1,452, 1,586, 1,382, 1,183, 2,135 and 1,189 kg, respectively. These results indicated that Khuzestan province has the highest potential in the field of milk production. To study the effect of weaning stress on dairy buffalo calves,5 healthy calves were chosen and weaned,and the blood samples were collected at 1 day before weaning,and 1,14 and 21 days after weaning to analysis blood physiology and biochemistry indexes and immune function.The results showed that the count of white blood cell,lymphocyte,erythrocyte,monocytes and hematocrit,aspartate aminotransferase activity,blood glucose,cortisol and immunoglobulin G contents of dairy buffalo calves were no significant difference between before and after weaning treatments (P > 0.05).The count of neutrophils at 21 d after weaning were significantly lower than that of 1 d before weaning of dairy buffalo calves (P < 0.05).However,lactate dehydrogenase and alanine aminotransferase activity in weaned treatments were significantly higher than those of animals in the treatment before weaning (P < 0.05).The serum urea nitrogen of dairy buffalo calves at 21 d after weaning was significantly higher than that of 1 d before weaning (P < 0.05) and hemoglobin content at 21 d after weaning was significantly lower than that at 1 day before weaning (P < 0.05).It indicated that the physiological and immunity function of dairy buffalo calves were challenged under weaning stress. 相似文献
This review summarizes the results of a study on rotaviruses isolated from calves affected by neonatal diarrhea.
The results indicated that rotavirus infection is widespread and supported the evidence for an etiologic role of these viruses in neonatal diarrhea. Differences in virulence among bovine rotaviruses appeared also to be confirmed.
Conventionally reared calves were fully susceptible to the experimental infection induced by rotaviruses originating from heterologous hosts, i.e. monkeys, pigs and rabbits.
When rotavirus strains of bovine, simian and rabbit origin were compared by cross neutralization tests, it was found the simian and porcine strains were indistinguishable and both appeared to relate antigenically to the bovine strain.
Finally, it was proven that feeding newborn calves with colostrum and first milk of their dams, previously vaccinated with an inactivated adjuvanted rotavirus vaccine, could prevent the neonatal diarrhea from occurring. 相似文献
