为了研究长期干旱条件下Mdm-miR160-MdARF17-MdHYL1 调控网络在苹果生长发育中的作用,以GL-3和Mdm-miR160e OE、MdARF17 RNAi、MdmARF17 OE、 MdHYL1 OE及MdHYL 1 RNAi转基因苹果为试验材料,进行为期3个月的长期干旱处理,测定苹果株高、根干质量、根冠比以及叶片净光合速率、胞间CO2浓度、气孔导度、蒸腾速率、叶绿素荧光值和水分利用效率。结果表明,在长期干旱胁迫下, Mdm-miR160e OE转基因苹果植株更矮化,根冠比、根干质量、光合能力及水分利用效率大于GL-3植株,具有更强的耐旱性; MdARF17 RNAi植株比GL-3更矮,且具有更大的根冠比、根干质量、光合能力和水分利用效率,而MdARF17 OE与之相反; MdHYL1 RNAi植株对长期干旱胁迫更敏感,其株高大于野生型且具有更小的根冠比、更低的光合能力和水分利用效率,而 MdHYL1 OE植物对长期干旱条件具有更强的适应性,生长发育表型与RNAi植株相反。综上所述,Mdm-miR160- MdARF17-MdHYL1调控网络在苹果响应长期干旱过程中发挥了重要功能,为苹果抗旱分子育种提供了候选基因。  相似文献

ABSTRACT

The aim of this study was to examine the usefulness of physical and chemical fractionation in quantifying soil organic matter (SOM) in different stabilized fraction pools. Soil samples from three land use types in Lorestan province, Southwest Iran were examined to account for the amount of organic carbon and nitrogen in different SOM fractions. Size/density separation and chemical oxidation methods were applied to separate the SOM fractions including particulate organic matter (POM), Si + C (silt and clay), DOC (dissolved organic C), rSOM (oxidation-resistant organic carbon and nitrogen) and S + SA (sand and stable aggregates). The values obtained for TOC, TN, and HWC were highest in forest lands followed by the range and agricultural lands. Among the SOM fractions, S + SA showed the highest values (5.75, 5.77 and 20.6 g kg?1 for agriculture, range and forest lands respectively) followed by POM, Si + C, rSOM, and DOC. The concentrations of C and N in the labile fractions obtained the higher values than in the stabilized fractions. Forest lands had the highest amounts of organic C and N among all fractions whereas agricultural lands showed highest values for inorganic C content of soils in different fractions.  相似文献

研究了10.8%草甘膦·2,4-D水剂(AS)对柑橘园杂草的田间防治效果.结果表明,10.8%草甘膦·2,4-D AS制剂用量15 750~31 500 mL/hm2几乎对所有禾本科、阔叶类和其他类杂草均有较好的防治效果,防效与41%草甘膦异丙胺盐AS制剂用量4 116 mL/hm2相当,显著优于72% 2,4-D丁酯EC制剂用量8 100 mL/hm2的防效,作用较快,持效期较长.  相似文献

This study aimed to evaluate the effect of sorghum and sweet potato on the bioavailability of iron, gene expression of proteins involved in iron metabolism and the plasma antioxidant capacity in animals fed with whole sorghum grains processed by dry heat or extrusion, combined or not with sweet potato flour with high content of carotenoids. Five experimental groups were tested (n = 7): dry heat sorghum flour (DS); extruded sorghum flour (ES); whole sorghum flour + sweet potato flour (DS + SP); extruded sorghum flour + sweet potato flour (ES + SP) and positive control (FS). The evaluations included: hemoglobin gain, hemoglobin regeneration efficiency, gene expression of divalente metal transporter 1 (DMT-1), duodenal citochroma B (DcytB), ferroportin, hephaestin, transferrin and ferritin and total plasma antioxidant capacity (TAC). The ES + SP group showed higher (p < 0.05) expression of DcytB, ferroportin and hephaestin when compared to the control group. The DS group showed high (p < 0.05) expression of DMT-1 and the ES showed high mRNA expression of transferrin and ferritin. The changes in the sorghum physicochemical properties from extrusion process reduced the iron and phytate content, and increased the gene expression of proteins involved in iron metabolism, improving iron bioavailability. The combination of sweet potato and sorghum flour (dry or extruded) improved the iron capture and total antioxidant capacity, probably due to the presence of β-carotene and antioxidant compounds.  相似文献

在鱼类胚胎发育过程中,胰岛素样生长因子-II(insulin-like growth factors-II,IGF-II)起着关键的作用。本研究根据Gen Bank收录的鲑鳟鱼IGF-II基因序列设计引物,以哲罗鱼(Hucho taimen)肝RNA提取物为模板,利用RT-PCR方法扩增出哲罗鱼IGF-II基因开放阅读框。将目的基因IGF-II与原核表达载体p SUMO连接构建出重组表达载体p SUMO-IGF。将该重组质粒转化到大肠杆菌Rosetta中进行目的蛋白的诱导表达。SDS-PAGE电泳分析显示,约在40 k D处含有清晰的条带,与预期结果相符;目的蛋白以包涵体形式存在。对包涵体进行变性/复性后获得较纯的目的蛋白,利用ELISA和MTT方法对目的蛋白进行免疫学活性及生物学活性分析。ELISA结果显示该目的蛋白能够与商品化的抗鲑鳟鱼IGF-II的抗体发生特异性反应,并且呈现抗原浓度依赖性,该结果说明本研究获得了具有良好免疫原性的IGF-II蛋白;MTT方法测定IGF-II蛋白对鲤(Cyprinus carpio)上皮细胞(epitheliaoma papulosum cyprini,EPC)和虹鳟(Oncorhynchus mykiss)性腺细胞(rainbow trout gonad,RTG-2)的增殖效果来鉴定IGF-II蛋白的生物学活性,结果显示所表达的哲罗鱼IGF-II蛋白能够有效的刺激EPC细胞和RTG-2细胞增殖。该结果表明利用原核表达系统获得的哲罗鱼IGF-II蛋白具有良好的生物学活性。本研究为哲罗鱼的生长模式和生长繁殖的研究奠定了基础。  相似文献

A 1‐year‐old, female spayed, domestic shorthair cat presented for blepharospasm of the right eye. Slit‐lamp biomicroscopic examination showed focal corneal ulceration and presumptive keratomalacia of the right eye. Examination of the left eye was normal apart from a focal endothelial opacity. Within the first 24 h of medical management, the right eye developed marked corneal edema and globular anterior protrusion of the corneal surface consistent with feline acute corneal hydrops (FACH). Surgical management consisted of a bridge conjunctival graft, nictitating membrane flap, and temporary tarsorrhaphy. Resolution of corneal edema and pain occurred in the right eye within 24 days. Spectral domain optical coherence tomography (SD‐OCT) of the anterior segment was performed in both eyes. Conjunctival tissue from the bridge graft precluded examination of deeper corneal structures in the right eye. The left eye displayed a focal separation of the corneal endothelium and Descemet's membrane from the overlying stroma. These SD‐OCT findings are similar to the analogous syndrome found in humans and represent a potential etiology for FACH of the right eye in the case presented here. Unfortunately, the cat was lost to follow‐up and the progression of this lesion to FACH in the left eye could not be determined.  相似文献

[目的]了解2.0%好普水剂对辣椒病毒病的防治效果和使用剂量,为其推广应用及辣椒病毒病的防治提供参考.[方法]以2%菌克毒克水剂为对照药剂,通过小区试验研究了2.0%好普水剂对辣椒病毒病的防治效果.[结果]2.0%好普水剂对辣椒病毒病有较好的防治效果,移栽后56 d第3次药后15 d调查,2.0%好普水剂稀释500倍、650倍对辣椒病毒病的防效分别为88.69%、80.35%,与2%菌克毒克水剂稀释260倍的防效相当.[结论]在辣椒上使用,推荐使用浓度为稀释500倍或650倍喷雾.  相似文献
