I—ELISA特异诊断棘球蚴（包虫）病的研究

采用亲和层析技术纯化的兔抗棘球蚴抗体，建立了酶联免疫吸附抑制试验（Ｉ—ＥＬＩＳＡ）。取代反应和阻断反应均无非特异反应；检测健康对照血清１２６份（人３６，猪９０），均为阴性；与猪囊尾蚴病血清１３６份（人４６，猪９０）、细颈囊尾蚴病血清３８份（羊２２、猪１６）无交叉反应；对棘球蚴病血清１８３份（人６１、羊７１、猪５１）进行检测，其阳性符合率分别为８８．５％（５４／６１）、９８．６％（７０／７１）、９６．１％（４９／５１）。     

Effect of YIGU capsule on IGF-I mRNA and protein expression in rat osteoblasts in vitro

AIM: The effects of YIGU capsule on proliferation and IGF-I mRNA protein expressions in osteoblasts were studied. METHODS: (1) Forty 12-month old Sprague-Dawley female rats were divided randomly into four groups (YIGU capsule high dose group, medium dose group and low dose group; saline group), the drug-containing serum and control serum were prepared. (2) The new-born Sprague-Dawley rat osteoblasts were cultured with different YIGU capsule drug-containing serum at different concentrations and different exposure time. MTT method was used to observe proliferation of osteoblasts. (3) RT-PCR method was used to measure the relative IGF-I mRNA levels and ELISA method was used to measure IGF-I secretion at different exposure time. (4) ELISA method was used to measure IGF-I secretion at different exposure time. RESULTS: (1) Proliferation of osteoblasts was more than the control groups after 48, 72 and 96 h, respectively (P<0.01); (2) The relative IGF-I mRNA levels and IGF-I protein expression were higher than those in control group after 48, 72 and 96 h, respectively (P<0.01 or P<0.05). CONCLUSIONS: It was suggested that YIGU capsule drug-containing serum promoted proliferation, IGF-I mRNA and protein expression. These results may be parts of the mechanisms of YIGU capsule to prevent and treat osteoporosis.     

玉米HD-ZIP I亚家族基因鉴定及表达分析

转录因子是植物响应逆境胁迫的重要调节因子,在其整个生长发育过程中发挥着重要的作用。HD-ZIP家族蛋白是植物中特有的一大类转录因子,包含4个亚家族(HD-ZIP I～IV),其中HD-ZIP I亚家族成员主要参与干旱、渗透压等极端环境和ABA及乙烯等激素处理的响应过程。本文采用隐马可夫模型(HMM)在玉米参考基因组中鉴定到17个HD-ZIP I亚家族成员,这些基因不均匀分布于玉米6条染色体上,与水稻的亲缘关系要近于拟南芥。玉米HDZIP I亚家族基因在玉米7种组织中表现出多种表达模式,具有明显的组织表达特异性。另外, HD-ZIP I亚家族基因对高盐、淹水及冷害等不同的逆境胁迫处理呈现出不同的响应模式及响应程度差异。5种不同激素处理后,玉米HD-ZIP I亚家族基因也表现出复杂的响应模式。这些结果为进一步解析玉米HD-ZIP I亚家族基因的生物学功能和作用机理提供了一定的参考价值。     

Effects of Nrf2 overexpression on proliferation,activation and collagen type I synthesis in cultured hepatic stellate cells stimulated by ethanol

AIM：To investigate the effects of nuclear factor E2-related factor 2 (Nrf2) overexpression on the proliferation, cell cycle distribution, collagen type I (Col I) synthesis and alpha-smooth muscle actin (α-SMA) expression in cultured hepatic stellate cell line HSC-T6 stimulated by ethanol. METHODS：Cultured HSC-T6 cells were transfected with pEGFP-Nrf2 or pEGFP-N1 (empty vector) plasmid by liposome transient transfection. The cells were divided into control group, ethanol group, ethanol+pEGFP-Nrf2 group and ethanol+pEGFP-N1 group. The mRNA expression of Nrf2, α-SMA and Col I was determined by RT-PCR, and their protein expression was detected by Western blotting. Cell proliferation was assessed by MTT assay, and cell cycle was detected by flow cytometry. RESULTS：The pEGFP-Nrf2 plasmid was successfully transfected into HSC-T6 cells, and the mRNA and protein expression of Nrf2 was higher than other three groups 48 h after transfection (P<0.05). Compared with control group, the cell proliferation and the mRNA and protein expression of α-SMA and Col I in ethanol group and ethanol+pEGFP-N1 group were significantly increased (P<0.05), and the numbers of HSC-T6 cells were decreased in G1 phase and increased in S phase (P<0.05), without significant differences between the two groups (P>0.05). Meanwhile, the cells in ethanol+pEGFP-Nrf2 group showed significantly decreased proliferation level, down-regulated mRNA and protein expression of α-SMA and Col I, higher numbers in G1 phase and lower numbers in S phase compared with ethanol group and ethanol+pEGFP-N1 group (P<0.05). CONCLUSION：Nrf2 overexpression could significantly down-regulate the expression of α-SMA and Col I and cause G1/S phase arrest in HSC-T6 cells cultured with ethanol, thus inhibiting the proliferation and activation of the cells.     

Expression of natural killer cell stimulatory receptor NKG2D and its ligand MICA in Tibetan patients with gastric cancer at Qinghai plateau

AIM: To investigate the expression of natural killer cell stimulation receptor D(NKG2D) in peripheral blood and the expression of major histocompatibility complex class I chain-related protein A(MICA) on the gastric carcinoma tissue and the neighboring non-cancerous tissue in Tibetan patients with gastric cancer at Qinghai plateau. METHODS: Samples of 33 patients and 20 healthy persons were collected to detect NKG2D in peripheral blood by Flow cytometry analysis. The expressions of MICA in part of the correspondent tissues were examined by RT-PCR and immunohistochemistry.RESULTS: The expression of NKG2D in the plateau of Tibetan patients with gastric cancer group (13.47 ±5.26)% was significantly lower than that in healthy groups (32.62±10.08)%. The mRNA level of MICA in gastric cancer was significantly higher than that in neighboring non-cancerous tissue (P<0.05). The expressions of MICA proteins showed a significant difference between the neighboring non-cancerous tissue (21.21%, 7/33) and the gastric carcinoma tissue (78.79%, 26/33), between the high differentiation (60.00%, 3/5) and the moderate (72.73%, 8/11), low differentiation (88.24%, 15/17). The expressions of MICA were not correlated with tumor size, sex, age, lymph node metastasis of the patients (P>0.05). Spearman rank correlation displayed that the expressions of MICA mRNA were positively correlated with the MICA protein level (r=0.903, P<0.01).CONCLUSION: The immune escape in Tibetan patients with gastric cancer at Qinghai plateau probably relates to the down-regulation of NKG2D and the up-regulation of its ligand MICA. The activity of NK cells and the anti-cancer cellular immunity level are descending in the plateau of Tibetan patients with gastric cancer. The decrease in the receptor NKG2D is a reason for the activity of NK cell descending.     

武威市极大风速气候特征分析及其推算

利用武威市观测站2005~2013年最大风速与极大风速观测数据,拟合武威市最大风速与极大风速之间关系,从而推算1980~2004年极大风速,对武威市1980年以来极大风速气候特征进行分析。结果表明,1980年以来武威市极大风速共出现4个峰值,分别出现在1984、1985、1993和2013年,尤其是1993年,极大风速达30.5 m/s,历史灾情记录显示1993年的"5.5黑风"破环力极强,给武威的农业生产带来了极大的影响,1994~2012年属于极大风速较小阶段,但由于此阶段武威市设施农业发展迅速,仍是给当地生产造成的严重的损失,利用第Ⅰ型极值分布方法推算武威市未来10、30、50、100年内可能出现的最大风速,推算结果分别为28.6、31.3、32.6、34.3m/s,在今后设施农业的规划和发展应加强大风灾害的防范。     

‘72杨’韧皮部的构造与理化性能

将‘72杨’、杉木和毛竹的木质部进行对比试验,观察并测试‘72杨’韧皮部的解剖构造以及理化特性,为其高值化利用提供基础数据。使用场发射环境扫描电镜、X射线衍射仪、傅里叶变换红外光谱仪等设备和NREL标准对‘72杨’韧皮部的微观结构、结晶度、化学成分等物化性质进行测定与分析。研究结果表明,‘72杨’韧皮部中韧皮薄壁细胞和筛管分子占细胞总面积的(81.9±1.8)%,结构相对简单。‘72杨’韧皮部气干密度为0.358 g/cm3,绝干密度为0.321 g/cm3;而木质部的相应密度较高,分别为0.497和0.482 g/cm3;‘72杨’韧皮部结晶度仅为19.4%,比木质部低8.7%。‘72杨’韧皮部纤维素、半纤维素、木质素的含量分别为28.7%,11.1%,24.1%,均低于木质部中相应成分的含量,且木质化程度低,半纤维素以木糖为主。此外,由红外谱图发现‘72杨’韧皮部含有单宁、酚类、胼胝质等物质。‘72杨’韧皮部具有低密多孔、结构疏松、结晶度低、木质素含量低、抽提物含量高等特点。因此,‘72杨’韧皮部特别有利于机械(能耗低)或化学(抗降解屏障低)降解以及物化改性(多孔、可及性强),可提取酚类、单宁、胼胝质等物质用于工业应用,研究结果可为‘72杨’韧皮部的高值化利用提供重要的理论依据。     

Molecular characterization of Ascaridia galli from Bangladesh and development of a PCR method for distinguishing A. galli from Heterakis spp.

We analyzed the nuclear ribosomal internal transcribed spacer (ITS) 1 and ITS2 sequences for Bangladesh isolates of Ascaridia galli, and we determined that the sequences were unreliable as molecular markers for distinguishing A. galli from other Ascaridia species, because the sequences showed high identity with that of A. columbae. However, the ITS1 sequences were available for designing PCR primers distinguishable between Ascaridia galli and Heterakis spp. Bangladesh isolates of A. galli constituted a monophyletic clade along with other geographical isolates in the cytochrome c oxidase subunit I (COI) phylogenetic tree, however, we could not clarify the phylogenetic relationships between A. galli and other Ascaridia spp., because their available sequences in GenBank were very few. The developed PCR method using DNA from A. galli and Heterakis spp. eggs would enable differential diagnosis of the individual infections in the future.     

THE REIMPROVEMENT FOR IMPULSE OF G. I. TAYLOR THEORY OF PLASTIC DEFORMATION IN IMPACT OF CYL1NDICAL PROJECTILES

In this paper the reimprovement for G. I. Taylor theory of plastic deformation in impact of cylindical projectiles is done. The impulse in the reimproved theory is G. I. Taylor' s 1/2, and 3/4 of G. I. Taylor' s improved theory. The analytical solution of the reimproved theory shows that it gives better agreement with exprimental results than those of the original Taylor' s theory and, the improved theory.
