Emmer wheat (Triticum dicoccon Schrank, 2n = 4x = 28) consists in a hulled wheat; its cultivation has been drastically reduced during the last century as a consequence of its low yield. Recently, its agronomic and nutritive values, as well as the increase of popularity of organic agriculture, have led to a renewed interest making its cultivation economically viable in the marginal lands with an increase of the cultivated areas. In Italy, it mainly survives in few marginal lands of central and southern Italy, where local varieties, adapted to the natural environment from where they originate, are used; moreover, some selected lines have also been developed. In the present work, agro-morphological and qualitative traits, together with molecular analyses of 20 emmer accessions consisting of Italian landraces, breeding lines, and cultivars, were performed. The field experiments were conducted for two consecutive years (2001/2002–2002/2003) in two locations: Viterbo in central Italy, and Foggia in south Italy. The analyzed emmer wheat accessions showed a good amount of genetic variability for both evaluated agro-morphological and molecular traits. This study illustrates an increase in earliness, GY, TW, TKW, and YI going from landraces, breeding lines to cultivars, while the variability does not change proportionally.  相似文献

Genetic diversity was investigated in 73 accessions of emmer wheat (Triticum dicoccon Schrank) from 11 geographical regions using a set of 29 simple-sequence repeat (SSR or microsatellite) markers, representing at least two markers for each chromosome. The SSR primers amplified a total of 357 different alleles with an average of 12.31 alleles per locus. The number of fragments detected by each primer ranged between 6 (Xgwm1066) and 21 (Xgwm268). Null alleles were detected in nine of the 29 primers used. A high level of gene diversity index was observed. Across the 29 primers, gene diversity ranged from 0.60 (Xgwm46) to 0.94 (Xgwm655), with a mean of 0.82. There was a highly significant correlation (r=0.882; p<0.01) between gene diversity index and the number of loci, showing the number of loci per se is a strong indicator of diversity. Analysis of genetic diversity within and among eleven geographical regions revealed most of the genetic diversity of the total sample resided within regions. The coefficient of gene differentiation (Gst = 0.27) showed that the genetic variation within and among the 11 geographical regions was 73 and 27%, respectively. High value of mean number of alleles per locus was found in Iran (4.86) followed by Morocco (4.10) and Armenia (4.03). On the contrary, lower mean number of alleles per locus was detected in Yemen (2.83). The average gene diversity index across regions ranged from 0.52 (Slovakia) to 0.67 (Morocco) with an average of 0.60. Multivariate techniques of principal component analysis and clustering were employed to examine genetic relationship among the 73 emmer wheat accessions vis-à-vis geographical regions of collections. The genetic distance coefficients for all possible 55 pairs of regional comparisons ranged from 0.63 (between Iran and Armenia, Georgia and Azerbaijan, Georgia and Slovakia) to 0.97 (between Morocco and Yemen, Spain and Georgia, and Turkey and Iran) with a mean of 0.82. From the PCA results, a two dimensional plot of PC1 versus PC2 was constructed. The scatter plot of the first two principal components which explained altogether 27% of the total variation depicted the presence of a clear pattern of geographical differentiation except in few cases like accessions from Caucasian region. Similar pattern of genetic relationships among accessions was observed in cluster analysis. The study provided genetic information of emmer wheat in relation to geographical regions of origin. The information could be utilized in crop improvement, germplasm conservation programs, and in further investigation.  相似文献

Enzymatic activity often reduces the nutritional value of wheat flour during food manufacturing, causing compound degradation and/or heat damage. The choice of wheat varieties with low enzymatic activity could therefore help to preserve the nutritional quality of food. The aim of this research was to evaluate polyphenol oxidase, alpha-amylase and beta-amylase activities in whole meal flours of 59 accessions belonging to different wheat species and subspecies, cropped in two years. The extraction pH (7.0), reaction pH (5.5) and reaction temperature (45 °C) were determined by preliminary trials. The ANOVA highlighted significant differences for all enzymes among species/subspecies and, for amylases, between cropping years; however, the year influence was overwhelming only for alpha-amylase. Einkorn showed the highest polyphenol oxidase (362.1 ± 9.46 U/g DM) as well as the lowest alpha-amylase (0.20 ± 0.006 CU/g DM) and beta-amylase (12.0 ± 0.36 B3U/g DM) activities. The embryo/scutellum had the highest polyphenol oxidase and alpha-amylase values, followed by the bran and the endosperm; in contrast, beta-amylase was evenly distributed in the bran and the endosperm, and was absent in the embryo/scutellum.  相似文献

The increasing demand for traditional and natural foods has renewed the interest in hulled wheat species. Among these, Triticum dicoccum Schübler has survived in Italy only in a few hilly and mountainous areas of central and southern Italy. As a rule, emmer is cultivated in marginal areas with organic farming procedures that use very low N inputs, since this wild species is characterised by low yield, long and weak culms that easily lodge under windy conditions. Unfortunately, there is a lack of information on crop productivity and N effect for this crop. Research was carried out to study the effect of cultivation year (2004 and 2005), plant origin (Garfagnana, Leonessa and Molise) and N dressing (N0, N30, N60 and N90 kg ha−1) on crop yield and grain quality of emmer grown in south-central Italy. Fertilizer was split at seeding, tillering and stem elongation. Tested parameters were highly influenced either by crop origin or N application (biomass, hulled and threshed grain yield, spikes m−2, spikelets per spike, kernel weight, plant height, lodging, kernel ash and proteins). Several parameters augmented as fertilization rate increased (hulled and unhulled grain yield, biomass accumulation, spikes m−2, kernels m−2, protein content); 1000-kernel weight showed an opposite trend and in some cases no differences were noticed among fertilized treatments (plant height and spikelets per spike). Molise was the most productive population, closely followed by Garfagnana. The present research rejected some common belief that emmer has to be grown without N dressing, and crop undergoes lodging in marginal mountainous areas. Besides, grain yield of N90 and N60 treated emmer was only 6 and 18% lower compared to the five most important durum wheat varieties cultivated in the same area, suggesting emmer as a possible alternative crop to durum wheat in marginal areas of Mediterranean-type agro-ecosystems.  相似文献

采用十二烷基磺酸钠 -聚丙烯酰胺凝胶电泳 (SDS- PAGE)方法 ,对 172份来自以色列的野生二粒小麦的高分子量谷蛋白亚基 (HWM- GS)组成进行了鉴定和分析。结果发现 ,这 172份野生二粒小麦 1B染色体上的HMW- GS有 19种变异类型 ,比普通小麦 1B染色体上 HMW- GS的变异类型丰富得多 ,其中有 8种 (6 * 8* ,6 8* ,2 2 ,6 * 8,7* 9,7* 9* ,7* 8* ,7* 8)为普通小麦中不常见类型 ,各个亚基的分布频率不同 ,17 18亚基分布频率最高 (2 9.6 5 % ) ,并且存在 3种普通小麦中不存在的特殊亚基 (图 1中加 ?的类型 ) ;1A染色体上的 HMW- GS有4种变异类型 ,其中一个亚基 (自定义为 1* )以前未见报道。发现野生二粒小麦 HMW- GS组成中 Glu- B1位点的 17 18亚基和 Glu- A1位点的 1亚基出现频率比普通小麦中出现的频率高。这些试验数据可为野生二粒小麦上特异HMW- GS的进一步研究与利用提供理论依据。  相似文献

Durum wheat (Triticum turgidum L. var. durum) is used predominantly for pasta products, but there is increasing interest in using durum for bread-making. The goal of this study was to assess the bread-making potential of 97Emmer19, an Emmer wheat (Triticum turgidum L. var. dicoccum) and in breeding lines derived from crosses of 97Emmer19 with adapted durum wheat cultivars. 97Emmer19 and its progeny were evaluated in 2005 and 2006 along with five durum wheat cultivars. Three bread wheat (Triticum aestivum L.) cultivars were included as checks to provide a baseline of bread making quality observed in high quality bread wheat cultivars. 97Emmer19 exhibited higher LV than all the durum wheat checks and approached the LV achieved with the bread wheat cultivar ‘AC Superb’. Breeding lines derived from 97Emmer19 had higher LV than those of the durum wheat checks, confirming that this trait was heritable. In general, durum wheat cultivars with elevated gluten strength and/or increased dough extensibility were noted to have higher LV. Dough extensibility appeared to be a more critical factor as gluten strength increased. These results indicate that there is potential to select for genotypes with improved baking quality in durum breeding programs.  相似文献

High temperature has a negative impact on wheat grain quality and reduces market value. Emmer wheat (Triticum dicoccon Schrank), one of the earliest domesticated wheat species, is a source of genetic diversity for the improvement of heat and drought tolerance in modern wheat. However, the potential of emmer wheat for the improvement of grain physical quality under high temperature stress is little studied. A diverse set of 184 emmer-based hexaploid lines was developed by crossing emmer wheat with hexaploid wheat and backcrossing once to hexaploid wheat. These materials, seven hexaploid recurrent parents and seven commercial cultivars, were evaluated at two times of sowing (E1 and E2) in the field, in 2015–2016. The materials were genotyped using a 90 K SNP platform and these data were used to estimate the contribution of emmer wheat to the progeny. Significant phenotypic and genetic variation for grain physical quality traits including protein content and test weight was observed. High temperature significantly increased protein content and decreased test weight. Large scale field phenotyping identified emmer progenies with improved grain characteristic compared to their respective parents and commercial cultivars in both environments. A few families consistently produced higher trait means across environments compared to their recurrent parents. The emmer wheat parent contributed between 1 and 37% of the genome in emmer-based genotypes. Selected emmer derived lines with superior protein content and test weight, tended to have a greater genetic contribution from the emmer parent, ranging from 12 to 37% and 7–37% in E1 and E2, respectively. It was concluded that new genetic variation for seed traits, such as protein content and test weight, can be introduced to hexaploid wheat from emmer wheat. The newly developed emmer derivatives identified with enhanced grain quality under high temperature stress can potentially be used to improve grain quality through breeding.  相似文献

Emmer wheat is hulled wheat that was wide cultivated in Spain at the past. Actually, the most of this germplasm is conserved in Germplasm Banks, and only two small populations have been found in Asturias (North of Spain) in a recent collecting mission. In this work, a collection of 31 Spanish emmer lines developed from identical number of accessions of two Germplasm Banks was analysed for morphological spike traits and seed storage protein composition. Up to seven different botanical varieties were detected, which suggest the presence of a wide diversity, although lower than the historically described 10 botanical varieties. At level of seed storage proteins, the lines showed a high diversity, although the new alleles were present with low frequency in materials with scarce agronomic interest for the farmers (var. atratum, var. lagascae or var. pycnurum). This last circumstance could translate in a lost of variability by genetic drift.  相似文献

Emmer and spelt are two species of hulled wheats that once were widely cultivated in Spain. Today on much reduced extension, these crops are still grown in Asturias (Northern of Spain). A recent expedition to this region was made with the purpose of collecting representative samples of the genetic variability, and compares these materials with those collected previously and conserved in Germplasm Banks. During the field trips, 35 populations were collected with a high diversity, although it was lower than that described in the past. The results of this comparison indicated that great part of this diversity is in risk of further losses in the remain native hulled wheat, mainly due to a progressive introduction of foreigner spelt with better harvest characteristics.  相似文献

Today, emmer wheat, T. turgidum subsp. dicoccon, widely grown in the past is a candidate crop for sustainable agriculture in Italy. As part of a research project aimed at the enhanced use of the hulled wheat germplasm, molecular characterization was carried out to understand the genetic structure of the crop and to identify accessions of interest. A collection of 194 accessions was analyzed with 15 microsatellite loci (SSRs), while only a sample of 38 accessions was tested with 19 RFLP probes. The marker loci were selected on the basis of their independent genomic distribution. Genetic distances and allelic frequencies were calculated for each marker class. The genetic relationships were visualized with dendrograms. RFLP loci were, on average, less polymorphic than SSRs. An average Dice's genetic distance of 0.22 for RFLPs vs 0.38 for SSRs was detected, while an expected average heterozygosity per locus of 0.23 for RFLPs vs 0.26 for SSRs was also estimated. With a least number of 10 loci per marker class it was possible to identify each genotype. The most diverse accessions had different geographic origins. Germplasms from Italy and Ethiopia appear to belong to a more primitive genepool, given that a group of accessions from these countries were genetically differentiated from a Russian-Iranian group.  相似文献
