卵泡从原始卵泡发育为成熟卵泡,直至排卵、黄体发育等过程都受到精密的调控,产生大量的优势卵泡是绵羊产多羔及实现快速扩繁的关键因素。研究发现,相关信号通路和转录因子通过影响绵羊卵泡中卵母细胞、颗粒细胞的生长,进而调控卵泡的发育成熟,对这些信号通路进行深入了解,有助于探索卵泡发育的调控机制,早日实现绵羊高效繁育。Notch是卵泡发育过程中发挥重要作用的高度保守信号通路,PI3K/AKT/mTOR信号通路各成员都是广泛存在于细胞内的信号转导分子,在卵泡发育早期发挥了主要作用,还有间隙连接(gap junction,GJ)和跨带突触(transzonal projections,TZPs)等物理连接方式,在细胞间的交流通讯起到重要作用。作者详细介绍了Notch信号通路、PI3K/AKT/mTOR信号通路、间隙连接及跨带突触的结构功能在绵羊卵泡发育中的调控作用,为进一步探明绵羊卵泡发育的调控机制提供参考。  相似文献

AIM To observe the effect of naringenin on cardiac injury in ischemia/reperfusion (I/R) rats, and to explore whether the role of naringenin is involved in PI3K/AKT signaling pathway and endoplasmic reticulum stress and its related apoptotic pathways. METHODS SD rats (n=48) were randomly divided into sham operation (sham) group, model (I/R) group, naringenin treatment (NAR) group and naringenin+LY294002 (NL) group. Myocardial I/R injury model was prepared by ligation of left anterior descending coronary artery of rats for 30 min followed by reperfusion for 120 min. After reperfusion, the serum levels of cardiac troponin I (cTnI) was measured by ELISA. HE staining, TTC staining and TUNEL staining were used to detect the myocardial histopathological changes, myocardial infarction area and myocardial cell apoptotic rate. The mRNA levels of endoplasmic reticulum stress-related indicators glucose-regulated protein 78 (GRP78), C/EBP homologous protein (CHOP) and caspase-12 were detected by RT-qPCR. The protein levels of cleaved caspase-3, GRP78, CHOP, caspase-12, p-PI3K and p-AKT were determined by Western blot. RESULTS Compared with I/R group, the serum content of cTnI, myocardial pathological damage, myocardial infarction area and myocardial cell apoptotic rate were significantly reduced (P<0.05), the protein levels of cleaved caspase-3, GRP78, CHOP and caspase-12 were decreased (P<0.05), and the protein levels of p-PI3K and p-AKT were increased in NAR group (P<0.05). LY294002 attenuated the protective effect of naringenin to some extent. CONCLUSION Naringenin reduces myocardial I/R injury in rats possibly by activating PI3K/AKT signaling pathway and subsequently regulating endoplasmic reticulum stress and its related apoptotic pathways.  相似文献

AIM To study the effect of dihydroartemisinin (DHA) on the radiotherapy efficiency in hepatocellular carcinoma H22 cell tumor-bearing mice and the role of phosphatidylinositol 3-kinase (PI3K)/protein kinase B (AKT) signaling pathway in this process. METHODS A model of H22 cell tumor-bearing mice was established. The mice was divided into model group, single radiotherapy group, 5-fluorouracil (5-FU) group, and low-, medium- and high-dose DHA groups. The body weight and tumor volume in each group were measured every other day. At the end of administration, blood was collected from the tail of the mice and the animals were killed by neck removal immediately. The synergistic effect of DHA on radiotherapy was determined, and tumor growth inhibitory rate was calculated. The degree of lymphocyte transformation and natural killer (NK) cell activity were measured by MTT, the serum levels of interleukin-2 (IL-2) and IL-4 were measured by ELISA, and the protein levels of PI3K, AKT and p-AKT were determined by Western blot. RESULTS The H22 cell tumor-bearing mouse model was successfully constructed. Compared with model group, the TGT3 (tumor growth time to reach 3 times of volume) of single radiotherapy group was remarkably increased (P<0.05), while tumor weight, lymphocyte transformation degree, NK cell activity, IL-2 and IL-4 levels, PI3K protein level and AKT phosphorylation level were remarkably decreased (P<0.05). Compared with single radiotherapy group, TGT3, EF (enhancement factor), tumor inhibitory rate, lymphocyte transformation degree, NK cell activity, IL-2 level and IL-4 level were increased with the increase in DHA dose (P<0.05), and the PI3K protein level and AKT phosphorylation level were decreased (P<0.05). CONCLUSION DHA may enhance the immunity of tumor-bearing mice by inhibiting the activity of PI3K/AKT signaling pathway, thereby enhancing the efficacy of radiotherapy.  相似文献

AIM: To study the effect of fibroblast growth factor receptor 1 (FGFR1) expression knock-down on the viability, apoptosis, invasion and migration of infantile hemangioma endothelial cells (HemECs). METHODS: FGFR1 was down-regulated by FGFR1 small interfering RNA (si-FGFR1) transfection. The viability of the cells was measured by CCK-8 assay. The apoptotic rate was analyzed by flow cytometry and the invasion and migration abilities were determined by Transwell assay. The protein levels of phosphatidylinositol 3-kinase (PI3K), protein kinase B (AKT), and phosphorylated AKT (p-AKT) were examined by Western blot. RESULTS: Transfection of si-FGFR1 into HemECs had significant effects on inhibiting cell viability (P<0.05), promoting apoptosis (P<0.05), and decreasing cell invasion and migration abilities (P<0.05). The results of Western blot showed that knockdown of FGFR1 gene expression in the cells reduced the protein levels of PI3K and p-AKT (P<0.05), and had no significant effect on AKT protein level. CONCLUSION: Knock-down of FGFR1 expression changes the biological characteristics of endothelial cells in infantile hemangiomas by regulating PI3K/AKT signaling pathway.  相似文献

AIM: To investigate the autophagy of human ovarian cancer SKOV3 cells induced by cepharanthine and to explore its mechanism. METHODS: The effect of cepharanthine on the viability of ovarian cancer SKOV3 cells was measured by CCK-8 assay. The SKOV3 cells were treated with cepharanthine, and then the formation of autophagosome was observed with acridine orange staining under fluorescence microscope. The protein levels of LC3, AKT, p-AKT, mTOR, p-mTOR and GAPDH in the SKOV3 cells treated with cepharanthine were determined by Western blot.RESULTS: Cepharanthine significantly inhibited the viability of ovarian cancer SKOV3 cells in a dose-dependent manner (P<0.05). The number of the intracellular acidic autophagosomes with bright red fluorescence was significantly increased after cepharanthine treatment in the SKOV3 cells. The expression of LC3-Ⅱ in SKOV3 cells was significantly enhanced after cepharanthine treatment. Furthermore, treatment with cepharanthine in the SKOV3 cells also resulted in a significant down-regulation of phosphorylated form of AKT and mTOR (P<0.01), while the total protein level was not changed. Combination of cepharanthine and 3-methyladenine resulted in a substantial decrease in the cell viability compared with using cepharanthine alone.CONCLUSION: Cepharanthine significantly inhibits the growth of human ovarian cancer SKOV3 cells and induces the autophagy, which may be correlated with down-regulation of PI3K/AKT/mTOR signaling pathway.  相似文献

植物细胞中的K^+通道蛋白AKT1(Arabidopsis K^+transpoter 1)主要负责吸收K^+。本研究基于木薯基因组数据库信息,利用PCR技术从木薯中克隆一个MeAKT1基因,该基因全长2634 bp,编码877个氨基酸,生物信息学分析表明MeAKT1含有10个外显子和9个内含子,编码的氨基酸分子量为99.02 kD,等电点为8.43,属于稳定蛋白。MeAKT1包含有5个跨膜区域,N端含有1个Ion_trans结构域,C端有1个KHA结构域,其二级结构以α-螺旋和无规则卷曲为主,进化树分析发现MeAKT1与蓖麻RcAKT1的亲缘关系最近。通过对MeAKT1蛋白的生物信息学分析有助于下一步深入研究MeAKT1在木薯耐贫瘠过程中的功能。  相似文献

细胞自噬是哺乳动物细胞物质代谢的一个重要机制,与细胞凋亡共同参与卵巢卵泡的发育和闭锁,并发挥重要的作用。近年研究发现,磷脂酰肌醇3-激酶/蛋白激酶B(phosphatidylinositol 3-kinase/protein kinase B,PI3K/AKT)信号通路参与卵巢疾病的发生。PI3K和AKT的过度激活可使原始卵泡过早发育以及卵泡过快凋亡,卵巢颗粒细胞作为卵泡发育重要的支持细胞,其功能的减退或凋亡很可能引发一系列女性内分泌方面的疾病。FOXO3a转录因子是PI3K/AKT信号通路下游的重要靶蛋白之一,参与抗增殖和凋亡。本文就关于卵巢颗粒细胞自噬与PI3K/AKT/FOXO3a信号通路的相关进展加以综述。  相似文献

The lack of advanced animal models of human cancers is considered a barrier to developing effective therapeutics. Canine and human melanomas are histologically disparate but show similar disease progression and response to therapies. The purpose of these studies was to compare human and canine melanoma tumours and cell lines regarding MAPK and PI3K/AKT signalling dysregulation, and response to select molecularly targeted agents. Pathway activation was investigated via microarray and mutational analysis. Growth inhibition and cell cycle effects were assessed for pathway inhibitors AZD6244 (MAPK) and rapamycin (PI3K/AKT) in human and canine melanoma cells. Human and canine melanoma share similar differential gene expression patterns within the MAPK and PI3K/AKT pathways. Constitutive pathway activation and similar sensitivity to AZD6244 and rapamycin was observed in human and canine cells. These results show that human and canine melanoma share activation and sensitivity to inhibition of cancer‐related signalling pathways despite differences in activating mutations.  相似文献

AIM: To investigate the effects of high mobility group A2(HMGA2) gene knockdown on the cell viability, apoptosis, collagen synthesis and oxidative stress of human embryonic lung fibroblast (HELF) induced by transforming growth factor-β1 (TGF-β1). METHODS: The HELF were divided into blank group, TGF-β1 group,negative control (NC) group and HMGA2 siRNA(si-HMGA2) group. The protein levels of HMGA2, AKT and p-AKT were determined by Western blot. The cell viability and apoptotic rate was analyzed by MTT assay and flow cytometry,respectively. The mRNA expression of collagen I (COL-Ⅰ) and COL-Ⅲ was detected by RT-qPCR. DCFH-DA was used to detect the content of reactive oxygen species (ROS). RESULTS: Compared with blank group, the protein levels of HMGA2 and p-AKT, the cell viability, the mRNA expression of COL-Ⅰ and COL-Ⅲ in TGF-β1 group were significantly increased, but the apoptotic rate and ROS level were significantly decreased (P<0.05). Compared with TGF-β1 group, the protein levels of HMGA2 and p-AKT, the cell viability, the mRNA expression of COL-Ⅰ and COL-Ⅲ in si-HMGA2 group were significantly decreased, but the apoptotic rate and ROS level were significantly increased (P<0.05). CONCLUSION: Knockdown of HMGA2 gene expression decreases the viability and collagen synthesis, and promotes apoptosis and ROS production of human embryonic lung fibroblasts induced by TGF-β1. The mechanism may be related to down-regulation of PI3K/AKT signaling pathway.  相似文献
