Two viruses, detected frequently in the Netherlands in pelargonium, were identified by serology and test plant reactions. Antisera were prepared and an ELISA procedure was developed to detect the viruses in pelargonium.One of the viruses, PFBV-N, proved to be pelargonium flower-break virus. With the antiserum to PFBV-N, it could be detected reliably throughout the year inPelargonium zonale Springtime Irene.The other virus, PLPV-N, was serologically closely related to pelargonium line pattern virus (PLPV) and to pelargonium ring pattern virus (PRPV), as were an old virus isolate from Saturnus, collected in the Netherlands in 1971 (L128), and PLPV isolates from Yugoslavia (PLPV-Y) and Denmark (PLPV-D). There were only minor differences in host-plant reactions between the virus isolates. Based on these tests, PLPV and PRPV are considered as isolates of the same virus, for which, for practical reasons, the name pelargonium line pattern virus is proposed.PLPV could be reliably detected by ELISA inP. zonale Springtime Irene and Amanda throughout the year with only a few exceptions. InPelargonium peltatum Tavira, however, reslts were erratic due to uneven distribution of virus in the plant. Best results were obtained when petioles of fully expanded leaves were tested.  相似文献

An outbreak ofDichelia cedricola (Diakonoff) (Lep.: Tortricidae), the cedar shoot moth (CSM), began in spring 1998 and lasted 3 years. This was the first monitored outbreak of the CSM in Isparta, Turkey. Tree crowns recovered to near normal condition by the middle of each growing season (in early June) during the outbreak. Tree volume and volume element increments were examined throughout the outbreak cycle from 1954 to 2001. In the past, CSM activity in stands of Lebanon cedar (Cedrus libani A. Rich.) was assessed through radial increment analyses. Cedar tree ring chronologies were analyzed for evidence of the CSM. Tree-ring chronologies from nonhost cedar (nondefoliated sample trees) were used to estimate potential growth in the host cedar (defoliated sample trees) during current and past outbreaks; all trees selected were the same subspecies and varieties. Regional outbreaks of the CSM were identified by synchronous and sustained growth periods of the trees. In 2001, increment cores were collected from 17 host and 16 nonhost dominant or codominant trees and annual radial growth indices from 1954–2001 were calculated for each of two host and two nonhost sample plots. Growth functions were defined as the cumulative sum of radial, height, and volume increment, and were graphically compared between CSM host cedar and nonhost cedar trees. Tree ring evidence suggests that a large-scale outbreak occurred in 1955 (from 1955 to 1966) and a small outbreak occurred in 1985 (1985–1990) and in 1998 (1998-continued) in the study area. The average diameter growth reductions around 1955, 1985 and 1998 were 40%, 46% and 7% of potential, respectively. 结果母枝和主枝环切处理导致糯米糍荔枝的果实大小显著减小,裂果率显著增加,环切圈以上叶片中可溶性糖和淀粉的含量显著增加,结果母枝中的可溶性糖含量增加,果实中的总糖与蔗糖含量则显著地降低。据此认为,近几年来,糯米糍的果个比20世纪80年代明显减小,裂果增加,很可能与生产中多年连续施行主枝环切(或环剥)技术有关,而碳水化合物运入果实量的减少可能是环切导致果实变小的原因之一。  相似文献

Sertoli cells are the only somatic cells in the seminiferous epithelium which directly contact with germ cells. Sertoli cells exhibit polarized alignment at the basal membrane of seminiferous tubules to maintain the microenvironment for growth and development of germ cells, and therefore play a crucial role in spermatogenesis. Androgens exert their action through androgen receptor (AR) and AR signalling in the testis is essential for maintenance of spermatogonial numbers, blood–testis barrier integrity, completion of meiosis, adhesion of spermatids and spermiation. In the present study, we demonstrated that AR gene could promote the proliferation of immature porcine Sertoli cells (ST cells) and the cell cycle procession, and accelerate the transition from G1 phase into S phase in ST cells. Meanwhile, miR-124a could affect the proliferation and cell cycle procession of ST cells by targeting 3′-UTR of AR gene. Furthermore, AR bound to the RNF4 via AR DNA-binding domain (DBD) and we verified that RNF4 was necessary for AR to regulate the growth of ST cells. Above all, this study suggests that AR regulates ST cell growth via binding to RNF4 and miR-124a, which may help us to further understand the function of AR in spermatogenesis.  相似文献

为了明确苹果轮纹病菌Botryosphaeria dothidea对吡唑醚菌酯的敏感性和吡唑醚菌酯的靶标基因序列, 采用菌丝生长速率法测定了水杨肟酸对B. dothidea 菌丝生长的作用, 探讨了添加或不添加水杨肟酸的情况下病原菌对吡唑醚菌酯敏感性的变化; 并测定了不同产区的80株B. dothidea 对吡唑醚菌酯的敏感性以及440株B. dothidea 对吡唑醚菌酯的抗性; 然后, 扩增并分析了具有不同敏感性的菌株的细胞色素b基因 (Cytb) 序列?结果表明:不同浓度水杨肟酸对菌丝生长的抑制作用不同?添加40 μg/mL水杨肟酸不影响吡唑醚菌酯的EC50?菌株的敏感性频率符合近似正态分布, 各产区菌株对吡唑醚菌酯的敏感性没有显著差异, 吡唑醚菌酯平均EC50为(2.95±2.11) μg/mL, 没有检测到抗性菌株?靶标基因序列分析表明, Cytb基因在F129?G137和G143位密码子上没有产生点突变, 首次发现在143位密码子后有内含子插入?  相似文献

由葡萄座腔菌Botryosphaeria dothidea引起的梨轮纹病是梨生产中重要的真菌性病害,主要造成树势早衰和果实腐烂,严重影响梨的产量和品质。本研究选取市售的6个品种的梨果实,用分离自梨枝干的轮纹病菌进行了离体果实接种,测定不同品种果实对梨轮纹病的抗性差异。接种后6 d,按照病斑直径从大到小的顺序,品种排序依次为‘丰水梨’>‘南果梨’>‘库尔勒香梨’>‘皇冠梨’>‘雪花梨’>‘砀山酥梨’,随病斑减小抗性依次增强。测定了4种杀菌剂对梨轮纹病菌菌丝生长的抑制作用,结果表明,咯菌腈、氟啶胺和咪鲜胺对梨轮纹病菌的菌丝生长有很强的抑制作用,EC 50分别为0.010 3、0.022 4μg/mL和0.034 3μg/mL;代森锰锌的EC 50为3.860 7μg/mL,也有较好的抑菌效果。本试验结果为了解不同梨品种对轮纹病的抗性及杀菌剂的选用提供了依据,具有一定的指导意义。  相似文献
