Fusarium oxysporum f. sp. ciceris (Foc), the causal agent of fusarium wilt of chickpea, consists of two pathotypes (yellowing and wilting) and eight races (races 0, 1B/C, 1A and 2–6) of diverse geographical distribution. Six Foc isolates, one each of races 0, 1B/C, 1A, 4, 5 and 6, representing the two pathotypes and the geographical range of the pathogen, showed identical sequences in introns of the genes for translation elongation factor 1α ( EF1 α), β-tubulin, histone 3, actin and calmodulin. Eleven additional Foc isolates representative of all races, pathotypes and geographical range, and three isolates of F. oxysporum (Fo) nonpathogenic to chickpea were further analysed for sequence variation in the EF1 α gene. All isolates pathogenic to chickpeas shared an identical EF1 α gene sequence, which differed from that shared by the three Fo isolates nonpathogenic to chickpea. EF1 α gene sequences from the 17 Foc isolates and the three Fo isolates were compared with 24 EF1 α gene sequences in GenBank from isolates of 11 formae speciales of F. oxysporum by parsimony analysis. Foc isolates formed a grouping distinct from other formae speciales and nonpathogenic isolates. These results indicate that F. oxysporum f. sp. ciceris is monophyletic. 相似文献

参考GeneBank发表的马立克氏病病毒(MDV)国际标准强毒株GA的基因序列,设计合成一对引物,分别以RBIB,814,GD2(广东分离株),J-1-E(北京分离株),Md11,Md5,CV1988等不同毒株的MDV基因组DNA为模板,通过PCR扩增,获得了预期大小的PCR产物。该产物经pGEM-T-easy克隆后测序,将所得序列进行比较分析。结果发现:不同毒株间pp38基因的启动子和增强子序列间有缺失突变,序列的同源性大于95.9%,其中大多数的突变发生在MDV复制的原点附近。 相似文献

The objectives of the current study were to investigate the mitochondrial genome and molecular phylogeny of Lueyang black-bone chicken, and provide molecule base to preserve and explore the specific chicken strain.2. Based on sequencing and clustering, the complete mitochondrial DNA map and sequences of Lueyang black-bone chicken were revealed, and two phylogenetic trees of Lueyang black-bone chickens based on D-loop sequences and the mitochondrial genome were constructed.3. The results showed that the complete mitochondrial genome of Lueyang black-bone chickens is 16,784bp in size, consisting of 22 transfer RNA genes, two ribosomal RNA genes, 13 protein-coding genes, and one non-coding control region. The base composition of the complete mtDNA sequence is 30.28% for A, 23.78% for T, 32.42% for C, 13.52% for G. Additionally, 10 haplotypes of D-loop sequences in 32 Lueyang black-bone chickens were detected, which were distributed into 4 clades (A, B, C and E).4. Under Mediterranean conditions, drought affects cereals production principally through a limitation of grain filling. In this study, the respective role of post‐anthesis photosynthesis and carbon remobilization and the contribution of flag leaf, stem, chaff and awns to grain filling were evaluated under Mediterranean conditions in durum wheat (Triticum turgidum var. durum) cultivars. For the purpose, we examined the effects of shading and excision of different parts of the plant and compared carbon isotope discrimination (Δ) in dry matter of flag leaf, stem, chaff, awns and grain at maturity and in sap of stem, flag leaf, chaff and awns, this last measurement providing information on photosynthesis during a short period preceding sampling. Source–sink manipulations and isotopic imprints of different organs on final isotope composition of the grain confirmed the high contribution of both carbons assimilated by ears and remobilized from stems to grain filling, and the relatively low contribution of leaves to grain filling. Grain Δ was highly and significantly associated with grain yield across treatments, suggesting the utilization of this trait as an indicator of source–sink manipulations effects on grain yield. Chaff and awns Δ were better correlated with grain Δ than stem and leaf Δ, indicating that chaff were more involved in grain filling than other organs. Moreover, in chaff, sap Δ was highly significantly correlated with dry matter Δ. These results suggest the use of Δ for a rapid and non‐destructive estimation of the variation in the contribution of different organs to grain filling. 相似文献

试验以"LAMP+微流控"技术为基础,开发饲料动物源性成分检测芯片。针对牛、羊、猪、鸡的16srRNA靶序列,根据LAMP引物设计软件进行4种肉类LAMP引物设计。在LAMP优化试验、芯片优化测试、特异性验证试验和芯片应用测试中,进行最佳引物筛选测试和特异性测试。结果显示,筛选出的4种肉类最佳引物特异性强,最佳反应温度为65℃。芯片特异性验证试验的归一化荧光曲线显示,试验结果准确、特异性稳定。对饲料市场购买的16份样本中检测出的动物源性成分,采用中华人民共和国农业行业标准验证,符合率100%。微流控检测芯片的开发与应用为饲料中动物源性成分检测提供新方法,为饲料产品质量安全提供保障。 相似文献

马鹿是非常重要的物种资源,由于栖息地的流失和人为干扰进行近亲繁殖等导致野生马鹿数量急剧减少,而家养马鹿多经过改良,因此马鹿的纯种数量锐减。对马鹿进行分子遗传学研究不仅可以加深人们对马鹿起源和物种形成的认识,还能帮助开展遗传多样性保护研究。随着高通量测序技术、分子生物学和生物信息学的迅速发展,马鹿的起源进化研究已发展到全基因组水平,并取得了一定的成果。马鹿的起源进化研究从最初对体态外貌和染色体核型的研究逐渐发展到对DNA序列与生理指标的研究。文章回顾了近年来国内外对马鹿起源进化和遗传多样性方面的研究,从起源时间、起源地和迁徙路线等方面揭示了马鹿的演化历史,介绍了父系、母系和常染色体研究方面分析了马鹿遗传多样性选取的不同分子标记,为进一步揭示马鹿种群的遗传变异、分化情况、迁徙路线和系统发育关系等提供基础信息,同时为马鹿遗传资源的利用和保护以及马鹿产业的良性发展提供重要的借鉴与参考。 相似文献

人工驯养野生动物是获取野生动物产品的重要手段,但是非法盗猎的野生动物产品以人工饲养的名义进入市场,就会对野生动物的保护形成挑战。因此,准确鉴别市场上的动物产品是来自养殖场还是野外,是保护野外种群,维持养殖业正常秩序的关键。本研究以北美水貂(Neovison vison)为例,基于颧骨内侧空间(ZAS)、颧宽(ZAA和ZBA)、颅基长(BSL)、下颌长(MDL)、眶后收缩(POC)和最大面部宽度(GFB)等量度,建立了9个头骨形态计量学指标:POC/GFB、POC/BSL、POC/MDL、ZAS SQRT/BSL、ZA SSQRT/MDL、ZBA×BSL/2ZAS、ZBA×MDL/2ZAS、ZBB×BSL/2ZAS和ZBB×MDL/2ZAS。野生组(n=32,9雄23雌)和饲养组(n=45,35雄10雌)的比较表明,除POC/GFB外,所有指标对于在两组样品之间均有显著差异,整体判别正确率为73.2%—85.5%。在性别已知的情况下,雄性的整体正确率为82.4%—93.8%,雌性为41.7%—85.7%。为此,应针对不同性别、不同来源的动物建立相应的参考数据,利用似然比进行鉴别。本研究的结果不仅可用于北美水貂的鉴别,也为其他小型食肉动物的鉴别提供经验。 相似文献

Milk produced by the grazing system, referred to as “grazing milk” contains many components required for human health. The milk fatty acid (FA) profile is strongly associated with the diet on the farms. In the present study, based on the FA profile of farmer's bulk milk, we determined how to discriminate between milk produced on grazing and on a confinement system. A field survey was conducted four times (grazing and confinement season) in the Konsen (29 farms) and Okhotsk (25 farms) area in Hokkaido. Farmer's bulk milk samples and details of feeding management were collected and the FA profile of milk was measured. Milk produced during the grazing season contained less C16:0 and cis‐9 C16:0, and more C18:0, cis‐9 C18:1, trans‐11 C18:1, cis‐9,12 C18:2, cis‐9,trans‐11 C18:2 and cis‐9,12,15 C18:3 than milk produced during the confinement season. Discrimination analysis using 16 FA revealed that almost all milk samples were discriminated correctly (confinement season: 90% correct and 10% borderline, grazing season: 88% correct, 9% borderline and 3% incorrect). For farmers that were categorized incorrectly and were considered borderline in the grazing season, the dependency on pasture was low compared with that for farmers correctly discriminated.
