将72只日龄为14d的“星布罗”肉用仔鸡随机分为实验和对照两组,每组36只。实验组接受强度为151x的1L:3D间歇性白炽灯光照,对照组为自然光照。两组的饲喂条件相同。结果:与对照组相比,实验组体重略高而料肉比略低,实验组鸡胰腺A、B细胞的内分泌颗粒减少,A颗粒粒芯周围出现大致均匀的晕轮,B颗粒粒芯呈现不典型的针状、棒状或环状,PP细胞的内分泌颗粒明显增大、增多,胞浆中的粗面内质网、高尔基复合体和线粒体等数量增多。两组鸡的D细胞无明显差异。34日龄的实验组鸡血浆胰岛素含量明显高于对照组,且与体重呈正相关。  相似文献

AIM: To observe the change of insulin receptor in rabbit kidney with acute ischemic-reperfusion injury. METHODS: 15 Japanese white rabbits were allocated randomly into control group, ischemic-reperfusion group(IR group). IR group received clamping for 1 h followed by 2 h or 48 h of reperfusion. At 2 h or 48 h after reperfusion, glucose and insulin in serum were determined. Insulin receptor in renal tissue was analyzed by radioligand binging assay(BAD). RESULTS: The level of serum glucose increased after 2 h reperfusion in 2 groups, but in IR group the value increased much more higher than those in control groups(P<0.05). Plasma insulin of IR group was significantly higher than that in control after 2 h reperfusion(P<0.05). Scatchard analysis of data resulted in curvilinear profiles, indicating that there are two classes of receptors with different affinity or the presence of a single class of receptors with a negative cooperative hormone-receptor interaction. Data analyzed for a two-site model showed that the values of Bmax1(high affinity site), Bmax2(low affinity site) and Kd1, Kd2 were significantly lower than that of control (P<0.05) after 2 h perfusion. 48 h after IR there was no difference of Bmax1, Bmax2, and Kd1 between 2 groups,but Kd2 of IR group was higher than that of control (P<0.05). CONCLUSIONS: The results indicate that the effect of intrinsic insulin decreases in the progress of the renal ischemic-reperfusion. The resulting high serum glucose may aggravate renal injury in the progress of ischemic-reperfusion.  相似文献

将55头21日龄断奶的雄性仔猪随机分为5组,各组仔猪饲喂赖氨酸含量为0.60%、0.80%、1.00%、1.20%和1.40%的试验日粮21d。42日龄结束饲养试验时进行葡萄糖灌注试验,观察赖氨酸采食量对早期断奶仔猪生长发育和胰腺内分泌的影响。结果表明:早期断奶仔猪体增重与赖氨酸采食量为强正相关,根椐体增重指标,21~42日龄阶段,仔猪的赖氨酸采食量不应低于3.16g/d,日粮赖氨酸含量不应低于1.40%。赖氨酸严重缺乏(采食量小于1.45g/d)时,胰岛素的分泌量下降,对高血糖应激的反应能力降低。  相似文献

Sodium‐glucose cotransporter 2 (SGLT2) inhibitors are used in the treatment of human diabetics. They increase glucose excretion and correct hyperglycemia. We examined the investigational SGLT2 inhibitor velagliflozin in two groups of six neutered adult obese cats (equal gender distribution). Placebo (Pl) or drug (D; 1 mg/kg) was administered for 35 days. Routine blood examinations, fructosamine, beta‐hydroxybutyrate (BHB), nonesterified fatty acids (NEFA), glucagon, adiponectin, and leptin were measured before and after treatment, also water intake, and urinary electrolytes, glucose, and volume. Indirect calorimetry, an intravenous glucose tolerance test (IVGTT; 0.8 g/kg) and insulin tolerance test (IVITT) were conducted. All cats tolerated treatment well. Significant changes with D included a decrease in the respiratory exchange ratio, an increase in cholesterol, a small increase in albumin, and a rise in BHB and NEFA. Glucose clearance was unaltered, although less insulin was secreted during the IVGTT (p = .056) suggesting improved insulin sensitivity. IVITT was unchanged. Treatment did not affect glucagon, leptin, or adiponectin. Water intake, urine output, urinary glucose excretion, and the glucose/creatinine ratio but not urinary electrolytes were significantly higher post‐D. We conclude that velagliflozin is a promising drug, which increases urinary glucose excretion in cats and could thereby be beneficial for the treatment of hyperglycemia.  相似文献

The objective of this study was to observe how fat incorporated into an equine forage‐based diet through supplementation altered levels of plasma glucose, insulin and fatty acids. Five Shetland/Hackney cross pony mares were fed alfalfa pellet diets top dressed with commercially available vegetable oil (blend of soya bean, canola and corn oils) at 0%, 5%, 10% or 15% of diet. Ponies were randomly assigned one of four diets to start, with a 14‐day adjustment period between transitioning to another one of the four diets. Ponies were gradually adapted to the new diet within the 14‐day period before a five‐day trial period. Each pony received all four diets by the end of the study. Each trial was a five‐day period with a three‐day sample collection. Blood samples for each collection week were taken 0, 30, 60, 90, 120, 150, 180, 210, 240 and 270 min and at 5, 6, 7, 8, 9 and 10 hr post‐feeding. Excess fat did not impact plasma glucose (p > .1), nor did it affect blood plasma insulin concentration. While there was no time alteration found for plasma fatty acid concentration (p > .1), C14:0 increased when ponies were fed 0% fat and C18:2 decreased when ponies were fed 0% fat. Plasma fatty acids (% of total FA) were higher in C18:0, C18:1, C18:2 and C20:1 in the added fat diets (p < .1). These findings suggest the amounts reported in this study of fat supplementation on a forage‐based diet did influence the fatty acid analysis within the pony, but did not negatively impact blood glucose and insulin concentrations.  相似文献

探讨茶色素对胰岛素抵抗大鼠红细胞胰岛素受体的影响。将50只清洁级SD大鼠随机分成对照组、高脂组和低中高剂量茶色素干预组。高脂组和茶色素干预组以高脂饲料饲喂8周诱发肥胖致胰岛素抵抗模型,茶色素干预组再给予0.1、0.2g/(kgbw·d)和0.4g/(kgbw·d)剂量茶色素灌胃8周,实验结束后采血测定红细胞胰岛素受体水平,同时测定血糖、血胰岛素、血FFA及血痩素。与对照组相比,高脂组空腹胰岛素、痩素和FFA明显升高(P<0.05),而红细胞高亲和力受体数目明显降低(P<0.05),与高脂组相比,中高剂量茶色素干预组空腹胰岛素、痩素和FFA明显降低(P<0.05),而红细胞高亲和力胰岛素受体数目明显升高(P<0.05)。说明茶色素干预可提高胰岛素抵抗大鼠胰岛素高亲和力受体数目,其机制可能与降低痩素和FFA水平间接提高受体数目或直接作用于受体基因调节元件有关  相似文献

BackgroundFor the treatment of diabetes mellitus (DM) in dogs, novel insulins with decreased injection frequency while maintaining safety and efficacy are desirable. Insulin fused with immunoglobulin‐fragment‐crystallizable (Fc) has an ultra‐long plasma half‐life because it recycles through cells, protected from proteolysis.HypothesisGlycemic control can be achieved in diabetic dogs with a recombinant fusion protein of a synthetic insulin and canine Fc (AKS‐218d) administered subcutaneously once‐weekly.AnimalsFive client‐owned dogs with naturally occurring DM.MethodsProspective clinical trial in dogs with DM that were recruited from the UC Davis Veterinary Teaching Hospital and local veterinary clinics. Dogs previously controlled using intermediate‐acting insulin q12h were transitioned to once‐weekly injections of a preliminary construct identified as AKS‐218d. The dose of AKS‐218d was titrated weekly for 8 weeks based on clinical response and continuous interstitial glucose monitoring. Clinical signs, body weight, serum fructosamine concentrations, and mean interstitial glucose concentrations (IG) over the preceding week were compared between baseline (before AKS‐218d) and during the last week of treatment. Data were compared using nonparametric paired tests.ResultsOnce‐weekly AKS‐218d, compared to baseline twice‐daily insulin therapy, resulted in no significant changes in clinical signs, median (range) body weight (+0.4 kg [−0.5‐1.1]; P = .6), fructosamine concentration (−75 mmol/L [−215 to +126]; P = .4), or mean IG (+81 mg/dL [−282 to +144]; P = .8). No adverse reactions were reported.ConclusionControl of clinical signs, body weight, and maintenance of glycemia was achieved with this once‐weekly novel insulin construct in 4 of 5 dogs.  相似文献

目的观察口服和静脉注射重组hGLP-1类似物对糖尿病大鼠血糖、胰岛素及胰岛组织病变的影响。方法用链脲佐菌素(STZ)建立SD大鼠糖尿病模型,随机分为模型组、灌胃组和注射组,每组10只,分别给予生理盐水灌胃、重组hGLP-1类似物多肽灌胃、hGLP-1类似物基因重组载体尾静脉注射,观察4周内各组大鼠血糖、胰岛素、胰岛组织病理变化。结果与模型组比较,灌胃组和注射组的血糖水平在给药后1~4周明显下降,而血清胰岛素水平显著升高(P〈0.01)。灌胃组在各时间点的血糖、胰岛素水平与注射组相比,差异无统计学意义(P〉0.05)。HE染色发现,灌胃组和注射组大鼠的胰岛组织病变有所改善。结论重组hGLP-1类似物可降低糖尿病大鼠的血糖、提高血清胰岛素水平。  相似文献
