[目的]研究外源施加棉子糖与植物成熟器官耐逆的相关性。[方法]将拟南芥离体叶片分别浸泡在Hoagland水溶液和0.1%棉子糖(W/V)的Hoagland水溶液中,黑暗中浸泡24 h,重复5次,探讨了含0.1%(W/V)棉子糖的Hoagland溶液对拟南芥离体叶片失水速率的影响。[结果]含0.1%棉子糖的Hoagland水溶液处理叶片的饱和鲜重为对照的104.46%。在正常温度光照条件下,含0.1%棉子糖的Hoagland处理的叶片失水速率与对照的基本保持一致,但同样时间点测量的鲜重较高。含0.1%棉子糖的Hoagland处理的叶片干重较对照的偏高,为对照的104.20%。[结论]外源施加棉子糖增加了饱和鲜重和干重,提高了拟南芥离体叶片的脱水胁迫耐性,同样时间点的生物量下降较少,失水速率基本不变。  相似文献

The meal value of Soybean for monogastric animals is determined partly by sucrose, raffinose and stachyose. Of these, sucrose is desirable, while raffinose and stachyose are indigestible, causing flatulence and abdominal discomfort. The objective of this study was to identify quantitative trait loci (QTL) controlling seed sucrose, raffinose, and stachyose in a set of 140 SoyNAM (Nested Association Mapping) recombinant inbred lines (RILs), developed from the cross between lines IA3023 and LD02‐4485. A total of 3,038 SNP markers from the Illumina SoyNAM BeadChip SNP were used to map the QTLs for sucrose and the RFOs, raffinose, and stachyose. Significant genotypic differences (p < .001) among RILs were observed for sucrose, raffinose and stachyose contents across years. A 3038 Illumina SoyNAM BeadChip SNPs identified three QTLs for sucrose, one on chromosome 1, explaining 10% variance and two on chromosome 3 each explaining 22%. Raffinose QTL was detected on chromosome 6, explaining 6% variance. The mapped QTLs were novel and spanned regions harbouring candidate genes with roles in plant growth including seed development.  相似文献

The identity and quantity of and effect ofprocessing on raffinose oligosaccharides in raw,mature seeds of lima beans (Phaseolus lunatus),pigeon peas (Cajanus cajan), African yam beans(Sphenostylis sternocarpa) and jackbeans (Canavalia ensiformis) were investigated. Sucrose,raffinose, stachyose and verbascose were identified byHPLC in all the legume seeds. The total-galactoside contents of the seeds in decreasing order were African yam beans3.84 mg/100 mg; white lima beans 3.62 mg/100 mg; creampigeon peas 3.51 mg/100 mg; red lima beans3.37 mg/100 mg; jackbeans 2.83 mg/100 mg and brownpigeon peas 2.34 mg/100 mg. The predominantoligosaccharide was verbascose in pigeon peas andstachyose in the other three legumes. Cooking unsoakedseeds brought about a greater reduction in the total-galactoside content than soaking for ninehours. The removal of oligosaccharides was higher inlegumes cooked in alkaline solution than in water.Germination quantitatively reduced raffinose,stachyose and verbascose while sucrose was increasedin all seeds except red lima beans and jackbeans.  相似文献

Thirty-three peanut cultivars were examined for their alpha-1,6 and beta-1,4 galactosidase activities and oligosaccharide contents along with proximate compositions. The average moisture, protein, fat, ash, and carbohydrate contents were: 4.9%, 26.6%, 43.1%, 2.3% and 23.1%, respectively. The corresponding coefficients of variation were: 5.2%, 10.1%, 7.2%, 7.8% and 15.7%, respectively. Raffinose and stachyose contents (%) ranged from 0.05 to 0.12 and 0.31 to 0.61, respectively. The specific activity (micromol product/min/mg protein) of crude preparation of alpha-galactosidase for the 33 cultivars ranged from 1.096 to 2.784 for the non-germinated seeds and from not being detected in some samples up to 2.432 for the germinated seeds; the mean values for non-germinated and germinated seeds were: 1.781 and 1.410, respectively. The specific activity of beta-galactosidase ranged from 0.101 to 1.727 in the non-germinated seeds and from not being detected in some samples up to 0.898 in the germinated seeds. Germination decreased the activity of both galactosidases significantly (p < or = 0.05).  相似文献

为了研究棉子糖对草鱼生长和血清生化指标的影响,选取初均重(69.51±6.29)g的草鱼288尾,随机分为6组,每组3个重复,每个重复16尾鱼.对照组饲喂基础饲料,试验组分别饲喂含200、400、800、1200和2000 mg/kg棉子糖的试验饲料,试验期为70 d.结果显示:与对照组相比,1)添加棉子糖能够提高草鱼的特定生长率,降低饲料系数,其中1200 mg/kg组与其余各组差异显著(P<0.05);2)各试验组蛋白质沉积率均有所提高,但无显著差异(P>0.05);脂肪沉积率除400 mg/kg组外,其余各试验组均显著升高(P<0.05);尤以1200 mg/kg组的蛋白质沉积率和脂肪沉积率最高;3)各试验组血清甘油三酯含量显著升高(P<0.05),高密度脂蛋白、低密度脂蛋白含量均显著降低(P<0.05).由此得出:1)棉子糖能够提高草鱼对饲料的吸收利用,增强鱼体对脂肪的吸收并节约蛋白质,从而促进草鱼生长;2)棉子糖可以明显的起到调节草鱼脂质代谢的作用.  相似文献

棉子糖是人与动物重要的营养成分。为了研究生态因子对大豆子粒中棉子糖含量的影响,2001—2002年在河南省夏大豆主产区的3个试点,以豫豆25为材料分13期播种,将78个样本子粒的棉子糖含量与气象、土壤养分和海拔等37个生态因子进行统计分析。诸样本棉子糖含量的变异范围为0.22%~0.87%。用逐步回归法筛选出8个生态因子与大豆棉子糖含量极显著正相关,各因子对于棉子糖含量影响的最大、最小值分别为0.392%和0.156%,按各因子的贡献大小依次为鼓粒成熟期昼夜温差、分枝期日照、土壤pH值、花荚期日照、土壤锰含量、花荚期降水、分枝期降水、出苗期降水。该结果对高棉子糖含量品种选育和配套栽培技术研究有参考价值。  相似文献

The effects of storage at 5, 10 or 15 °C for 6 months on the concentrations of starch and soluble sugar in winter squash (Cucurbita maxima Duch.) cultivar ‘TC2A’ fruits were examined. Starch contents were significantly lower at 15 °C than at the other temperatures, although concentrations decreased throughout the storage period at all temperatures. Total soluble sugar contents increased during the first 3 months of storage regardless of temperature, and decreased at 5 °C or 15 °C, but not at 10 °C after 6 months. Myo-inositol and raffinose concentration patterns were more complex, and may reflect some role in regulating fruit metabolism during storage that may be important in maintaining overall squash fruit quality.  相似文献

本研究建立了一种同时测定大豆低聚糖粉中棉子糖和水苏糖含量的高效液相色谱分析方法(HPLC)。采用示差折光检测器(RID),色谱柱为Agilent Zorbax carbohydrate柱(4.6mm&;#215;150mm,5μm),流动相为乙腈-水(75:25,V:V),流速1.0mL/min,柱温30℃。实验结果表明:棉子糖和水苏糖分别在1~10mg/mL、1~10mg/mL范围内线性良好,相关系数分别为0.9925、0.9971;2种糖的相对标准偏差(RSD)分别为1.57%、1.81%(n=5),平均回收率分别为95.8%、96.0%。该分析方法适用于大豆低聚糖粉中棉子糖和水苏糖含量的检测。  相似文献

Fructans are prebiotics, with potentially beneficial effects on human health. This study aimed to examine genetic variation in wheat grain fructan content using a simplified analytical method. The method involves extracting fructans from wheat grain followed by enzymatic hydrolysis to break down fructans into monosaccharides that can then be quantitatively measured by anion-exchange liquid chromatography coupled with pulsed amperometric detection. The modified procedure is reliable and allows the handling of large numbers of flour samples at a low cost, and could therefore be useful for assessing large numbers of wheat breeding lines. Using this method, grain samples taken from 19 bread wheat cultivars and breeding lines grown in both glasshouse and the field were analysed for grain fructan content. In addition, grain samples of 29 international wheat landraces and 14 new wheat breeding lines from the International Maize and Wheat Improvement Center (CIMMYT) were surveyed for their fructan contents. There was significant genotypic variation among these materials, with grain fructan content ranging from 0.7 to 2.9% of grain dry weight. There was no evidence of strong genotype-by-environment interaction; the fructan contents of field-grown grain samples were positively correlated (r = 0.83) with those of glasshouse-grown samples of the same cultivars. It should therefore be possible to investigate the genetic control of variation for this trait using the simplified HPLC method and to select effectively for increased grain fructan content in wheat breeding.  相似文献
