AIM:To investigate endocrine differentiation and its mechanism in endometrial carcinoma.METHODS:Endocrine cells (EC) were identified by immunohistochemical staining of chromogranin A in 50 cases of endometrioid adenocarcinoma and 20 cases of normal endometrium. Double-label techniques were used for simultaneous demonstration of the CgA and cytokeratin.RESULTS:The positive rate of CgA in endometrioid adenocarcinoma was 44.0%, and was significant higher than that in normal endometrium (15.0%, P<0.05). Scanty EC was present in normal endometrium. The number and staining intensity of EC in endometrioid adenocarcinoma were greater than that in normal. Co-expression of CgA and cytokeratin were detected in endometrioid adenocarcinoma.CONCLUSION:The presence of endocrine cells in endometrioid adenocarcinoma showed heterogeneity of tumors. The occurrence of "Multidirectional differentiation cells" within the endometrioid adenocarcinoma may indicate that endocrine cells derive from malignant cells of multidirectional differentiation.  相似文献

Somatostatin receptors expressed by insulinomas in 5 dogs were imaged in vivo by means of indium in 111 pentetreotide (OctreoScan) scintigraphy. The diagnosis in each dog was supported by the presence of hypoglycemia (<60 mg/dL), hyperinsulinemia (>20 microU/mL), and histopathologic review of neoplastic tissue. All insulinomas expressed high-affinity somatostatin receptors of subtype sst2, as shown by receptor autoradiography in vitro using 125I-[tyrosine3]-octreotide and 125I-[leucine8, Dtryptophan22, tyrosine25]-somatostatin-28 with an sst2 subtype-selective analogue. Scintigrams were obtained at 1, 4, 12, and 24 hours after the i.v. administration of 74-222 MBq of OctreoScan to each patient. Abnormal foci of activity were 1st observed from 1 hour after administration of the radioligand in dog 3, to 24 hours after its administration in dog 4; in dogs 1 and 2, abnormal foci of activity were visible from 12 hours. Dog 5 showed a questionable abnormal focus of activity at 12 hours, but not at 24 hours. Scintigraphy enabled accurate prediction of the anatomical location of the primary tumor in 1 of 4 dogs, but was unable to differentiate a right- from a left-pancreatic lobe tumor, or vice versa, in 3 dogs; the 5th dog had equivocal results. 111In-pentetreotide scintigraphy is a useful diagnostic adjunct to the clinical evaluation of the insulinoma patient, but is unable to localize the tumor in some cases.  相似文献

以中华鲟(Acipenser sinensis)脑垂体总RNA为模板,采用RT-PCR和RACE方法,获得中华鲟神经内分泌多肽(7B2)基因的3个重叠片段,测序后拼接得到986 bp全长基因序列,其中包括5'端非翻译区(5′-UTR)14 bp、3′端非翻译区(3-′UTR)261 bp和开放阅读框711 bp。翻译编码236个氨基酸。其中前43个氨基酸为7B2的信号肽。经BLAST比对发现中华鲟7B2蛋白的同源性与斑马鱼(Danio rerio)的相似性最高为82%。系统发育分析表明,中华鲟与斑马鱼亲缘关系最近。半定量RT-PCR分析表明:在脑中7B2 mRNA表达量最高,心脏、性腺、胰等组织中表达次之,肠、肾、皮肤等组织中少量表达,肝和鳃几乎不表达。  相似文献

为挖掘我国名优鱼类长吻鮠 (Leiocassis longirostris)生长相关基因,本研究运用Illumina高通量测序技术比较分析了快速生长组[平均体质量为(534.02±53.68) g]和缓慢生长组[平均体质量为(108.41±4.96) g]各9尾长吻鮠的脑组织基因表达谱。测序共获得267 404 674个高质量测序片段(clean reads),通过2种不同生长速率长吻鮠脑组织转录组比较筛选出518个差异表达基因,其中,412 个基因表达量上调,106个基因表达量下调。对12个差异表达基因进行实时荧光定量PCR验证的结果与转录组测序结果一致。GO功能分类显示,大量差异表达基因富集到生长(growth)、生长因子活性(growth factor activity)和激素介导的信号通路(hormone-mediated signaling pathway) GO条目中。KEGG富集分析显示,一些差异表达基因在MAPK信号通路(MAPK signaling pathway)、转化生长因子β信号通路(TGF-beta signaling pathway)、钙离子信号通路(calcium signaling pathway)和神经活性配体–受体相互作用(neuroactive ligand-receptor interaction)等途径中富集。根据GO功能注释和KEGG富集分析,筛选出gnrh、thr、egr1、fgf18、sst、gipr、cart和crf等基因是调控长吻鮠生长发育的关键候选基因。本研究结果为后续深入研究长吻鮠生长调控机制提供了重要的参考资料。  相似文献

本文综述了近10年来关于异黄酮类化合物(芒柄花素和大豆黄酮)对动物(大鼠、猪为主)一些生产性能的影响及神经内分泌参与的研究.建立了芒柄花素的放射免疫方法;经放免受体分析表明,异黄酮植物雌激素可与动物的乳腺、垂体、下丘脑的胞浆雌二醇受体竞争性结合,表现弱的雌激素样作用;诱发血中催乳素、生长激素和IGF-1水平升高,从而促进乳腺发育和泌乳量;使雄性动物血中辜酮水平升高,促使胎儿和产后生长加速;促进小鼠的体液免疫和细胞免疫功能,提高妊娠母猪的特异免疫反应和新生仔猪特异性抗体水平;对雌性动物抑制LH的释放;此外,对反刍动物研究表明,可直接影响瘤胃代谢.并对其神经内分泌机制进行了探讨.  相似文献

The aim of the present study was to investigate the presence and the distribution of cells containing orexin A and orexin type 2 receptor in the horse stomach and gut, by means of immunohistochemical techniques.Orexin A was identified in the stomach fundic and pyloric regions and in the duodenum. In the same stomach regions, a large subset of orexin A-positive cells also showed orexin type 2 receptor-like immunoreactivity. Moreover, in the duodenum, many of them, seemed to store serotonin.Characteristically, enteric neurons or ganglia also displayed orexin A and, sometimes, orexin type 2 receptor immunoreaction.Orexin A and orexin type 2 receptor immunoreactivity was also found in the nerve fibers in the enteric submucosal layer.Our results, together with data present in the literature, could contribute to the understanding of complex mechanisms regulating the horse gut functionality that are depending very likely on the consequence of the co-operation of both a central and a peripheral control.  相似文献

AIM: To investigate the effect of 8-week middle intensity voluntary wheeling exercise on the depressive-like behavior and the circadian rhythmic alterations of plasma hormone and peptide induced by chronic unpredictable mild stress (CUMS) in rats. METHODS: Male Sprague-Dawley rats (n=90) were randomly divided into model group, model+exercise group and control group. Rats in model+exercise group received 8-week voluntary wheel running exercise plus CUMS procedure during the last 3 weeks at the same time. Exploratory locomotor activity was assessed by open field test, the anxiety-like behavior was measured by elevated plus-maze test, and lack of pleasure was detected by sucrose preference test. Blood samples were collected at each of 6 time points (ZT1, 5, 9, 13, 17 and 21 on the 2nd day after behavior testing). Plasma concentrations of corticosterone (CORT), melatonin (MT) and vasoactive intestinal peptide (VIP) were detected by ELISA. The plasma concentration of adrenocorticotropic hormone (ACTH) was measured by radioimmunoassay. The circadian rhythm changes of serum CORT, MT, VIP and ACTH concentrations in each group were compared by cosinor analysis. RESULTS: Compared with control group, locomotor activity, weight gain and sucrose consumption in model group were significantly reduced (P<0.01). The values of the percentage of open-arm time (OT%) and open arm entries (OE%) were obviously lower in model group than those in control group (P<0.01). Eight-week voluntary wheel running exercise may improve the above depression behavior caused by CUMS. The rats in model group showed an obvious disorder in circadian rhythm of plasma ACTH and CORT, including phase advance and decrease in amplitude. There also showed a markedly blunted circadian rhythm and decreased level of plasma MT in model rats compared to control rats. VIP expression was significantly higher than that in control group with 24 h rhythm, but the amplitude was significantly lower than that in control group, peak phase also delayed for 6 h. Eight-week exercise significantly ameliorated the abnormal expression and the disturbance secretion rhythm of ACTH, CORT, MT and VIP in plasma. CONCLUSION: Eight-week voluntary wheeling exercise ameliorates CUMS-induced depressive-like behaviors probably by rescuing the disturbed circadian rhythms and abnormal secretion of these neuroendocrine factors.  相似文献
