The effects of the severity and timing of leaf removal(LR) on the amino acids of Sauvignon Blanc grapes and wines were studied during the 2017 growing season. High-performance liquid chromatography(HPLC) was used to analyze the amino acids profiles of grape berries and wines. The basal leaves were removed at three time points(40, 56 and 72 days after flowering, named LR40, LR56 and LR72, respectively) at two severity levels(one at which the first, third, and fifth basal leaves of each shoot were removed(50% level); and another at which the first six basal leaves were removed(100% level)). The results showed that leaf removal had little impact on total soluble solids(°Brix), titratable acidity, pH or berry weight. The LR72-50% treated grapes had higher berry weight, titratable acidity and °Brix than those of the other treatments. The highest concentrations of total amino acids and of total amino acids except proline were detected in LR72-50% treated grapes(2 952.58 and 2 764.36 mg L~(-1), respectively); the lowest were detected in LR72-100% treated grapes(2 172.82 and 2 038.71 mg L~(-1), respectively). LR72-50% treatment significantly promoted the synthesis of aspartic acid, serine, arginine, alanine, aminobutyric acid and proline at both severity levels for grapes, the concentrations of all of these amino acids were increased relative to the control concentrations. The LR72-50%, LR40-100% and LR72-100% treated wines had higher total amino acids concentrations and higher concentrations of some individual amino acids, such as arginine, alanine and serine, than did the control wines. Of all the amino acids studied, glycine, tyrosine, cysteine, methionine and lysine were not significantly influenced by the timing or severity basal defoliation in grapes and wines. The present study reveals the effects of the timing and severity of leaf removal on the amino acids profiles of grapes and wines.  相似文献

冀谷19是采用杂交方法从"矮88×青丰谷"中选育而成,表现优质、高产和水分利用效率高.2002年和2003年国家夏谷新品种区域试验,平均单产5 296.5 kg/hm2,较对照豫谷5号增产14.4%.2004年通过全国谷子品种鉴定委员会鉴定.2004年推广面积58 000 hm2,最高单产达8 430 kg/hm2,成为河北省旱地无公害优质小米开发的骨干品种.  相似文献

Th IPK2 is an inositol polyphosphate kinase gene cloned from Thellungiella halophila that participates in diverse cellular processes. Drought is a major limiting factor in wheat(Triticum aestivum L.) production. The present study investigated whether the application of the Th IPK2 gene could increase the drought resistance of transgenic wheat. The codon-optimized Th IPK2 gene was transferred into common wheat through Agrobacterium-mediated transformation driven by either a constitutive maize ubiquitin promoter or a stress-inducible rd29 A promoter from Arabidopsis. Molecular characterization confirmed the presence of the foreign gene in the transformed plants. The transgenic expression of Th IPK2 in wheat led to significantly improve drought tolerance compared to that observed in control plants. Compared to the wild type(WT) plants, the transgenic plants showed higher seed germination rates, better developed root systems, a higher relative water content(RWC) and total soluble sugar content, and less cell membrane damage under drought stress conditions. The expression profiles showed different expression patterns with the use of different promoters. The codon-optimized Th IPK2 gene is a candidate gene to enhance wheat drought stress tolerance by genetic engineering.  相似文献

利用ANSYS合理建立模型及划分网格,将直接影响计算结果的精度,也是充分发挥计算软件数值模拟能力解决实际问题的关键。通过对ANSYS模型建立和网格划分模块的系统分析,结合巷道支护经验,找出了对U29型钢可缩性支架进行模拟计算的方法。为精确计算截面比较复杂、结构比较庞大的可缩性金属支架在各种载荷作用下的结构反应奠定了基础,并指出ANSYS建模和网格划分的一些应用技巧。  相似文献

以含有RD29A目的基因和NPTⅡ筛选基因的根癌农杆菌菌株LBA4404、GV3101和EHA105侵染饲用型四倍体刺槐(Robinia pseudoacacia)、国槐(Sophora japonica L.)和红叶石楠(Davidson Photinia)的组培苗叶片,研究3个树种对根癌农杆菌菌株和不同抗生素(替门汀和头孢霉素)的敏感性、以及根癌农杆菌菌株对3个树种的侵染能力。结果表明,不同树种、不同根癌农杆菌株和不同抗生素种类均对转化效果产生明显影响,红叶石楠是最佳受体材料,3个菌株侵染后的平均抗性愈伤率为37.0%,其次为国槐,四倍体刺槐最低;根癌农杆菌菌株LBA4404侵染能力最强,3个树种的平均抗性愈伤率达到33.3%、平均分化率达到17.6%;替门汀对农杆菌侵染后的红叶石楠组培叶片抑菌效果最好。  相似文献

AIM: To investigate the role of microRNA-29b (miR-29b)-mediated TGF-β/Smad signaling pathway in the activation of hepatic stellate cells (HSC) and its effect on the progression of hepatic fibrosis in rats.METHODS: Hepatic liver fibrosis rat model was established, and its HSC were isolated. Normal rat HSC were also obtained and identified in vitro. RT-qPCR and Western blot were used to detect the alterations of miR-29b, TGF-β/Smad signaling pathway-related proteins and liver fibrosis marker proteins in the acquired cells. Finally, the direct targeting binding of miR-29b to TGF-β1 was identified by dual-luciferase reporter assay system.RESULTS: With the activation of HSC, the expression of miR-29b gradually decreased (P<0.01), while the expression of collagen type I and α-smooth muscle actin gradually increased (P<0.01). At the same time, the expression of Smad2/3/4 was significantly increased, and the expression of Smad7 was significantly decreased (P<0.01). Dual-luciferase reporter assay showed that miR-29b bound directly to "UCUCUCCGU" in the 3'UTR of TGF-β1, indicating that TGF-β1 was a downstream target gene of miR-29b.CONCLUSION: miR-29b may be involved in the inhibition of HSC activation and migration, thereby inhibiting the process of liver fibrosis. The biological function of miR-29b may be through the direct targeting of TGF-β1, thus regulating and inhibiting the TGF-β/Smad signaling pathway.  相似文献

依据本实验室前期狂犬病病毒(RABV)感染小鼠原代神经元microRNA(miRNA)表达谱数据,选取表达显著上调的miR-29a和下调miR-124为研究对象,采用实时荧光定量PCR技术检测目的 miRNA在RABV感染前后的小鼠原代神经元和小鼠海马组织内的表达变化。运用TargetScan数据库预测目的 miRNA潜在的靶基因,并对靶基因进行了GO(Gene Ontology)注释描述、GO注释富集分析和信号通路富集分析,富集结果呈现出数个与神经功能相关的GO注释和信号通路。本研究结果提示miR-29a和miR-124可能参与了RABV感染所致的宿主神经功能异常。  相似文献
