In this study, primary and immortalized bovine intestinal epithelial cells (BIECs) were characterized for the expression of surface carbohydrate moieties. Primary BIEC-c4 cells showed staining greater than 90 % for 16 lectins but less than 50 % staining for four lectins. Immortalized BIECs showed significantly different lectin binding profile for few lectins compared to BIEC-c4 cells. BIEC-c4 cells were studied for infectivity to E. coli, Salmonella enterica, bovine rotavirus, bovine coronavirus, and bovine viral diarrhea virus. Bovine strain E. coli B41 adhered to BIEC-c4 cells and Salmonella strains S. Dublin and S. Mbandaka showed strong cell invasion. BIEC-c4 cells were susceptible to bovine rotavirus. LPS stimulation upregulated IL-10, IL-8, and IL-6 expression and Poly I:C upregulated TLR 8 and TLR 9 expression. This study provides important knowledge on the glycoconjugate expression profile of primary and immortalized BIECs and infectivity and immune responses of primary BIECs to bacterial and viral pathogens or ligands.  相似文献

The objective of this study was to determinate grain unique protein inherent molecular structure that are related physiochemical and nutrient profiles in CDC developed oat varieties [CDC Nasser (Feed Type) and CDC Seabiscuit (Milling Type)] grown in cool climate condition in western Canada in comparison with conventional barley variety of CDC Meredith as a control using advanced molecular spectroscopy. Multivariate analyses, including an agglomerative hierarchical cluster analysis (CLA) and principal component analysis (PCA), were performed to identify protein molecular structural differences among the grains. The results revealed that CDC Seabiscuit contained greater (P < 0.05) protein structural Amide I and II than CDC Nasser and CDC Meredith, while the greater (P < 0.005) structural Amide I to II area and height ratios was detected in CDC Meredith. New oat grains had greater (P < 0.05) β-sheet height than barley grains, however, there was no difference in α-helix to β-sheet ratio values among the varieties. In conclusion, CDC Nasser and CDC Meredith had no difference in protein molecular structural features, while CDC Seabiscuit contains different protein structural characteristics as compared to CDC Meredith grain. The molecular structure features are highly associated with physiochemical and nutrient profiles in grains, which indicate that it also affect nutrient utilization and availability.  相似文献

Agrobacteria were previously isolated from tumors developing on branches and aerial and hypogeous roots of weeping fig plants in Italy and in The Netherlands. A representative group of 48 strains was analyzed by PCR–RFLP of 16S and 16S + IGS ribosomal regions, PCR–RFLP of six Ti plasmid (pTi) regions and characterized for plasmid content. Two groups of agrobacteria were separated by cluster analysis of PCR–RFLP profiles of rrs gene: seventeen strains were similar to the new species Agrobacterium larrymoorei, while the remaining strains were included within the agrobacterium biovar 1 group. Sixteen different plasmid profiles from one to five plasmids were observed. In addition, 21 ribotypes and 20 pTi structures were arranged in many different combinations, showing that fig agrobacteria were characterized by a wide heterogeneity. A general lack of correlation between strain ribotypes and plasmid content was observed.  相似文献

Liver metabolism is affected by nutrients. The aim of this study was to explore the effects of low‐protein diets (17% crude protein, CP) supplemented with branched‐chain amino acids (BCAAs), including leucine (Leu), isoleucine (Ile) and valine (Val), on hepatic amino acid profile and lipid metabolism in growing pigs. The ratio of Leu : Ile : Val in all groups was 1 : 0.51 : 0.63 (20% crude protein, CP), 1 : 1 : 1 (17% CP), 1 : 0.75 : 0.75 (17% CP), 1 : 0.51 : 0.63 (17% CP) and 1 : 0.25 : 0.25 (17% CP) respectively. Results revealed that compared to the positive control group (1 : 0.51 : 0.63, 20% CP), the low‐protein diets significantly augmented the concentrations of most essential amino acids and non‐essential amino acids (< .05), with the greatest values observed in the 1 : 0.25 : 0.25 group. Moreover, relative to the control, the low‐protein diets with the Leu : Ile : Val ratio ranging from 1 : 0.75 : 0.75 to 1 : 0.25 : 0.25 markedly downregulated the mRNA abundance of acetyl‐CoA carboxylase (ACC), lipoprotein lipase (LPL) and fatty acid‐binding protein 4 (FABP‐4) (< .05), and upregulated the mRNA expression of hormone‐sensitive lipase (HSL), peroxisome proliferator‐activated receptor‐g coactivator‐1α (PGC‐1α), uncoupling protein 3 (UCP3) and liver carnitine palmitoyltransferase 1 (L‐CPT‐1) (< .05). Therefore, our data suggest that protein‐restricted diets supplemented with optimal BCAA ratio, that is, 1 : 0.75 : 0.75–1 : 0.25 : 0.25, induce a shift from fatty acid synthesis to fatty acid oxidation in the liver of growing pigs. These effects may be associated with increased mitochondrial biogenesis.  相似文献

This study aimed to isolate and characterize pepsin-solubilized collagen (PSC) from marine and freshwater fish swim bladders. The physicochemical properties, protein pattern, amino acid composition, structure, thermal denaturation temperature, and antioxidant activity of PSC from four different swim bladder sources were investigated and compared. The results demonstrated that the four types of collagen extracted were all type I collagen. The yield of PSC extracted from grass carp (GCSB-PSC), bighead carp (BCSB-PSC), grouper (GSB-PSC), and monkfish swim bladders (MSB-PSC) were 38.98, 27.97, 18.16, and 10.35%, respectively. Compared to the other three PSCs, BCSB-PSC has the highest thermal denaturation temperature (38.60 °C). Based on FTIR spectroscopy and circular dichroism (CD) analysis, the extracted PSCs retained the triple helix and secondary structure well. Antioxidant studies showed that in the swim bladders of four species the swim bladder PSC could scavenge DPPH and ABTS radicals. Overall, swim bladders from marine and freshwater fish can be utilized as raw materials for collagen extraction, and the extracted collagen has potential commercial applications.  相似文献

为明确Hsp70蛋白在绿豆象Callosobruchus chinensis高温耐受性中的作用,该研究筛选并克隆β-actin、α-tubulin、β-tubulin、EF1-α、GAPDH、Hsc70、AK和RPL40八个候选内参基因,采用实时荧光定量PCR(quantitative real-time PCR,qPCR)技术测定其在绿豆象雌雄成虫体内的表达量,通过geNorm、BestKeeperr和NormFinde软件对这8个候选内参基因的表达稳定性进行评价,选择最适宜的内参基因对不同高温胁迫下绿豆象体内Hsp70基因的表达特性进行测定。结果表明,8个候选基因引物均有良好的特异性和引物扩增效率,引物扩增效率介于91.6%~108.1%之间。高温胁迫下,β-tubulin基因较其他7个候选基因有较小的平均变异度、稳定值和标准差,为最适宜的内参基因。39、42和45℃高温处理1 h后绿豆象雌雄成虫体内Hsp70表达水平较对照显著上调,当处理时间延长至3 h后,Hsp70基因的相对表达量较对照也有不同程度上调,但差异不显著,表明绿豆象Hsp70基因可以响应高温胁迫,Hsp70蛋白在绿...  相似文献

一维马尔可夫链模拟黑河中游流域土壤质地垂向变异   总被引:1,自引:1,他引:0  
黑河中游地区土壤剖面砂、黏层次相间排列的特点及部分区域剖面中出现的不透水层对土壤中的水分运动和溶质迁移具有重要影响。该研究调查了黑河中游100 km2区域内土壤剖面的质地分层情况,运用一维嵌入马尔可夫链模型模拟该地区的土壤质地剖面。研究表明,研究区土壤剖面共出现砂土、壤质砂土、砂质壤土、壤土、黏质壤土、粉黏壤土和粉黏土7种质地类型,层次厚度呈对数正态分布。粉黏土未在表层土壤出现,而砂土在表层出现的概率明显高于其余质地类型,剖面某一质地层之下多出现细粒含量比其高的下一质地类型。剖面上相邻两质地层之间的转移具有明显的马尔可夫链特征(简称马氏性),且马氏链是平稳的。一维嵌入马尔可夫链模型能够较好地描述研究区土壤质地层次的垂向变化,剖面主要的质地层次组合为:砂—壤、壤—砂、壤—黏、黏—壤。对土壤质地剖面的定量模拟,可以为开展该区域土壤水循环、转化和溶质迁移等相关研究提供参考。  相似文献

以磴口华莱士蜜瓜为试材,研究不同贮藏温度对其采后果实质构特性、理化指标、感官品质变化及相关性分析。选用成熟度一致的果实随机分为2组,放置于常温(28℃)和低温(6℃)条件,每隔1 d进行感官评定,常规理化指标检测、质地多面分析(TPA)及整果穿刺试验。结果显示,低温贮藏能有效减缓果实可溶性固形物和有机酸含量的下降,有效延缓果皮硬度、果肉硬度和咀嚼性等的下降,并抑制果实的软化,较常温贮藏延长贮藏期12 d。相关性分析表明,质构参数之间、质构参数与感官品质指标之间存在不同程度的相关性。综上所述,磴口华莱士蜜瓜果皮硬度、咀嚼性和内聚性等均能较好地反映果实品质的变化,可作为其品质评价指标;该结果为开发磴口华莱士蜜瓜的贮藏保鲜技术提供了科学依据。  相似文献

 【目的】分离授粉早期玉米花丝的总蛋白质,为从蛋白质组角度揭示玉米花粉与花丝早期的相互作用奠定基础。【方法】以玉米(Zea mays L.)自交系Ga25为试材,采用三氯乙酸-丙酮法提取总蛋白质,运用双向电泳、质谱分析与检索技术,比较自交授粉后1 h和2 h的花丝蛋白质组的差异。【结果】与授粉1 h的蛋白质组相比,在授粉2 h后的蛋白质组中出现28个差异蛋白点,其中,6个蛋白质点特异表达,19个蛋白质点上调表达,3个蛋白质点下调表达;通过MALDI-TOF-MS质谱测序和MASCOT序列分析,注释了23个蛋白质点,其余5个为未知蛋白;功能分类表明,差异蛋白质分别参与细胞壁合成(21.4%)、防御/抗胁迫(17.9%)、细胞组织、蛋白命运、蛋白合成、转录等细胞过程。【结论】在玉米花粉与花丝早期的相互作用过程中蛋白质组有显著的变化,与授粉后1 h相比,授粉后2 h的蛋白质组中大部分差异蛋白呈上调趋势,并有特异蛋白质出现;分泌型过氧化物酶、膨胀素、果胶甲基化酶抑制蛋白、谷胱甘肽转移酶与未知蛋白4可能与玉米花粉和花丝的早期互作密切相关。  相似文献
