【目的】 类胡萝卜素裂解双脱氧酶基因(Carotenoid Cleavage Dioxygenases 4,CCD4)控制桃果肉颜色(白/黄),CCD4存在3种等位基因。本研究利用Indel、SSR荧光标记毛细管电泳及SNP鉴定等基因分型技术分析我国主要桃黄白肉品种(系)中CCD4等位基因的差异,为主要黄/白肉品种(系)的基因型鉴定、亲本选配和选择相应的分子标记对不同来源子代的果肉颜色进行鉴定奠定基础。【方法】利用已经报道的桃不同果肉颜色中CCD4等位基因3种突变类型,合成不同引物进行PCR扩增,LTR反转录转座子插入突变经1%的琼脂糖凝胶电泳检测,CT单元重复的PCR产物在ABI3730XL测序仪上进行SSR荧光标记毛细管电泳检测,SNP标记经Sanger测序后利用ContigExpress软件分析CCD4等位基因的碱基替换(A→T)。综合以上结果,统计每份材料中CCD4等位基因的突变类型与果肉颜色的一致性。【结果】通过对不同来源的122份桃品种(系)材料进行基因型分析,发现CCD4发生LTR反转录转座子插入突变材料的基因型共有31份,占总材料的25.4%,其中纯合插入突变材料的片段扩增长度为729 bp,共有8份,占总突变的25.8%;CCD4发生微卫星重复序列突变材料存在2 bp的插入,扩增片段长度为179 bp,该类型共有68份,占总材料的55.7%,其中纯合插入材料25份,占总突变的36.8%;CCD4发生A→T碱基替换突变的材料较少,仅有1份,占总材料的0.82%,实际应用中可以不考虑该种类型。CT和LTR插入的两种突变类型的黄肉品种(系)有7份,占总材料的5.7%。研究结果表明,LTR反转录转座子插入突变和微卫星序列重复突变是黄肉桃中CCD4等位基因的主要突变类型。其中CCD4发生一种纯合突变或两种杂合突变桃品种(系)为黄肉类型,分子标记鉴定结果与调查的122份桃品种(系)黄白肉表型性状完全一致,准确率为100%。【结论】采用分子标记明确了122个桃品种(系)黄/白肉性状的基因型,为不同亲本组合子代表型鉴定的标记类型选择提供了技术支撑,为建立桃种质材料黄/白性状的分子辅助育种体系和黄肉桃的选育奠定了基础。 相似文献

Campylorrhinus lateralis, also known as “wry nose,” is a congenital malformation that mainly affects Thoroughbreds. These horses have a unilateral deviation of the maxillae that may be to one side or the other side, and it causes airway obstruction and dental malocclusion. The choice of treatment is not necessarily operation; however, the treatment of choice, which aims to repair the maxillae deviation, is surgical to improve the horse’s respiratory condition and correct the dental occlusion. There are currently no reports describing the first surgical technique for such deformity described by Valdez et al. The present study describes the case of a 2-month-old foal of the Mangalarga Marchador breed that was diagnosed with wry nose. The foal presented with accentuated maxillae deviation to the right side, malocclusion of the incisor teeth, and respiratory noises. After diagnosis and physical examination, the foal was treated by surgical correction of the deviation. To correct the deviation, the bone distraction technique with unilateral osteotomy of the right maxillae and fixation of the external bone distractor was chosen. After 90 days, the bone distractor was removed; consequently, malocclusion of the incisors was greatly improved, and respiratory noises were eliminated. 相似文献

A challenge test against Aeromonas hydrophila was conducted using 2062 rohu carp (Labeo rohita) fingerlings obtained from 52 dams and 87 sires (87 full-sib families) of two year-classes (2003 and 2004). Attempts to establish a cohabitant challenge model were not successful. Therefore, fish were challenged by intraperitoneal injection with A. hydrophila in two replicate tanks per year-class, and dead fish were collected hourly. The mortality reached its peak at 16–22 h after challenge and had almost completely stopped after 58 h. The test was terminated after 382 h at which the average survival was 43.9 and 48.9% in the two 2003 year-class tanks, and 77.1 and 35.8% in the two 2004 year-class tanks. Heritability estimates for survival were obtained from sire and dam threshold models (THR) and sire and dam linear repeatability models (LINR). For both types of models the heritability estimates based on data from single tanks were not consistent. For both year-classes, data from one of the two challenged tanks demonstrated significant additive genetic variation in survival during the A. hydrophila infection, whereas the heritability estimates were not significantly different from zero for the other tank. Further, genetic correlation between survivals in the two replicate tanks in each year-class was not significantly different from zero. The differential results from the replicate tanks demonstrate that additional challenge test experiments are needed before firm conclusions can be drawn about the magnitude of additive genetic variation for survival to aeromonasis in rohu carp. A cohabitant challenge model that allows the testing of important defence mechanisms in the skin and mucous membranes of the fish might have been more appropriate. 在颗粒起动观测试验中,传统长直型水槽及环形水槽试验无法观测流体长期作用下的颗粒运动,依据锥-板转子流变仪原理,设计了一种锥-板环形水槽装置;采用无量纲拖曳力作为起动时的流体力学指标,结合CCD(电荷耦合组件,charge coupled device)成像技术及PIV(粒子图像测速,particle image velocimetry)技术,运用该装置对层流时无黏性均匀颗粒在流体长期作用下的运动形态及起动规律进行了观测,观测发现流体长期作用下颗粒的起动为连续的起动过程,存在2个不同的临界状态,分别为初始临界状态和饱和临界状态,初始临界状态下的起动拖曳力和经典Shields曲线的取值基本一致,饱和临界状态下的起动拖曳力明显高于Shields曲线的取值,表明锥-板环形水槽是一种合理有效的颗粒起动观测装置,特别适用于流体长期作用下的颗粒起动规律研究。 相似文献
