【目的】克隆淡色库蚊氯菊酯抗性相关XND-P450基因,并对其进行生物信息学分析,研究其在敏感品系和抗性品系淡色库蚊中表达量的差异,为阐明XND-P450的功能和抗性机制奠定基础。【方法】依据淡色库蚊抗性品系与敏感品系差异表达的EST片段设计PCR引物,采用RACE技术克隆淡色库蚊氯菊酯抗性相关基因XND-P450全长cDNA序列,并对其进行生物信息学分析;采用半定量PCR技术,对XND-P450基因在敏感品系和抗性品系淡色库蚊中的表达量差异进行检测。【结果】经过克隆获得长度为1 679bp的淡色库蚊氯菊酯抗性相关基因XND-P450全长cDNA,编码523个氨基酸。生物信息学分析表明,该基因与致倦库蚊、埃及伊蚊CYP450基因的同源性在70%以上,其编码蛋白相对分子质量为33 244.9u,理论等电点为8.10,属于不稳定的亲水蛋白,氨基酸序列与致倦库蚊及冈比亚按蚊CYP450蛋白的同源性在40%以上。半定量PCR电泳分析显示,抗性品系淡色库蚊中的XND-P450基因表达量较敏感品系高。【结论】克隆了淡色库蚊氯菊酯抗性相关XND-P450基因,该基因在抗性品系淡色库蚊中的表达量高于敏感品系,推测其与氯菊酯抗性相关。  相似文献

The effectiveness of a stocking, constructed of nylon and cotton netting, in protecting containerized conifer seedlings against pine weevil attack was evaluated in field tests on 37 clear‐cut reforestation areas in southern and central Sweden. The stockings significantly reduced pine weevil feeding on treated seedlings as well as seedling mortality. The protective effect of the stockings was similar to that of an insecticide (permethrin) treatment. In most experiments the survival of stocking‐enclosed seedlings was satisfactory from a practical point of view, whereas untreated control seedlings suffered heavy mortality.  相似文献

A retrospective analysis of all adverse experience reports received by the Australian Pesticides and Veterinary Medicines Authority's Adverse Experience Reporting Program for veterinary medicines since 1995, showed that permethrin toxicity in cats usually occurred after the owner applied a canine permethrin-containing product, typically a spot-on. Cats are also at risk from grooming or being in direct contact with recently treated dogs. This paper reviews permethrin toxicosis and its treatment in cats, incorporating information from the Australian and selected overseas veterinary pharmacovigilance programs.  相似文献

OBJECTIVE: Permethrin is a synthetic pyrethroid widely used in flea control products for small animals. Accidental toxicity can occur with off-label usage, and cats are particularly susceptible. METHODS: Retrospective study of 20 cases of permethrin toxicity in cats treated at an emergency clinic in Brisbane, Queensland from October 2004 to June 2005. RESULTS: The diagnosis of permethrin toxicity was made on the basis of a history of exposure and characteristic clinical signs, including seizures, muscle fasciculations, and tremors. Decontamination and appropriate seizure or muscle fasciculation control were the basis of treatment. The outcome was good after rapid intervention and 19 of the 20 cats were successfully treated, with the only death occurring in a kitten for which treatment was delayed for 24 h. No long-term complications were reported by the cats' owners at 4-month follow-up after discharge from hospital. CONCLUSIONS: Owner education, together with more appropriate product labelling, may help eliminate this problem in the future.  相似文献

Abstract

Trials were carried out in Upper Volta to assess equipment for the application of insecticides as aerosol from helicopters for the control of riverine tsetse flies (G. tachinoides). Flying techniques were developed to direct aerosols in the downwash of the helicopter to tsetse breeding sites close to the water's edge. Good penetration of droplets below the riverine canopy was achieved during the late afternoon and early morning when weather conditions were very stable.  相似文献

The stability to heart of cis-permethrin and beta-cypermethrin in the solid phase was studied and the decomposition products identified. Samples heated at 210 degrees C in an oven in the dark showed that, in the absence of potassium chlorate (the salt present in smoke-generating formulations of these pyrethroids), cis-permethrin was not isomerized, although in the presence of that salt, decomposition was greater and thermal isomerization occurred. Other salts of the type KXO3 or NaXO3, with X being halogen or nitrogen, also led to a considerable thermal isomerization. Heating the insecticides in solution in the presence of potassium chlorate did not produce isomerization in any of the solvents assayed. Salt-catalysed thermal cis-trans isomerization was also found for other pyrethroids derived from permethrinic or deltamethrinic acid but not for those derived from chrysanthemic acid. The main thermal degradation processes of cis-permethrin and beta-cypermethrin decomposition when potassium chlorate was present were cyclopropane isomerization, ester cleavage and subsequent oxidation of the resulting products. Permethrinic acid, 3-phenoxybenzyle chloride, alcohol, aldehyde and acid were identified in both cases, as well as 3-phenoxybenzyl cyanide from beta-cypermethrin. A similar decomposition pattern occurred after combustion of pyrethroid fumigant formulations.  相似文献

The synthetic pyrethroid insecticides cis‐ and transpermethrin are widely used, particularly in sheep‐dips and for mothproofing within the textile industry. This study aims to establish the concentrations and mobility of permethrin within rivers in the Humber catchment, which contain some of the highest densities of wool‐scouring activity in the world. Our approach was to utilise three different surveys: (a) weekly and storm‐responsive sampling of ‘whole waters’ in the rivers Aire, Ouse, Don, Trent and Calder; (b) intensive sampling of ‘whole waters’ and suspended sediments in the rivers Aire (0.5 h) and Calder (1 h); (c) a bed‐sediment survey of the River Calder. Sediments were extracted by supercritical fluid extraction and all samples were analysed by gas chromatography with mass spectrometry. Results demonstrated the presence of permethrin in ‘whole waters’ and sediments, particularly in the rivers Aire and Calder, and at concentrations in the bed‐sediment likely to cause ecotoxicological effects to benthic macro‐invertebrates. Mass‐balance calculations indicated some loss of permethrin from the water column during transport, with the greatest losses at low river‐discharge. Isomer ratios (trans:cis) give retention times of permethrin in different components of the system as ‘whole water’ < suspended sediments < bed‐sediments, with estimates of 4–26 days for suspended sediments and a maximum of 103–125 days for surface bed‐sediments. © 2000 Society of Chemical Industry  相似文献

BACKGROUND

Stable flies [Stomoxys calcitrans (L.)] are economically important pests of cattle and other livestock. As an alternative to conventional insecticides, we tested a push–pull management strategy using a coconut oil fatty acid repellent formulation and an attractant-added stable fly trap.

RESULTS

In our field trials we found that weekly applications of a push–pull strategy can reduce stable fly populations on cattle as well as a standard insecticide (permethrin). We also found that the efficacy periods of the push–pull and permethrin treatments following on-animal application were equivalent. Traps with an attractant lure used as the pull component of the push–pull strategy captured sufficient numbers of stable flies to reduce on-animal numbers by an estimated 17–21%.

CONCLUSIONS

This is the first proof-of-concept field trial demonstrating the effectiveness of a push–pull strategy using a coconut oil fatty acid-based repellent formulation and traps with an attractant lure to manage stable flies on pasture cattle. Also notable is that the push–pull strategy had an efficacy period equivalent to that of a standard, conventional insecticide under field conditions. © 2023 The Authors.
