We analyzed the nuclear ribosomal internal transcribed spacer (ITS) 1 and ITS2 sequences for Bangladesh isolates of Ascaridia galli, and we determined that the sequences were unreliable as molecular markers for distinguishing A. galli from other Ascaridia species, because the sequences showed high identity with that of A. columbae. However, the ITS1 sequences were available for designing PCR primers distinguishable between Ascaridia galli and Heterakis spp. Bangladesh isolates of A. galli constituted a monophyletic clade along with other geographical isolates in the cytochrome c oxidase subunit I (COI) phylogenetic tree, however, we could not clarify the phylogenetic relationships between A. galli and other Ascaridia spp., because their available sequences in GenBank were very few. The developed PCR method using DNA from A. galli and Heterakis spp. eggs would enable differential diagnosis of the individual infections in the future.  相似文献

利用RT-PCR技术检测VHA-c基因对高盐、水和ABA胁迫的响应,结果表明所有的VHA-c基因都参与了V-ATPase的抗盐胁迫反应;在响应水胁迫的反应中VHA-c2、VHA-c4和VHA-c5基因的表达量增加;在ABA胁迫反应中VHA-c1,VHA-c2,VHA-c3和VHA-c4基因的表达水平均提高,但是VHA-c1和VHA-c3基因只在早期的ABA胁迫中增加其表达量。  相似文献

为了探究盐酸小檗碱(berberine hydrochloride)对草鱼(Ctenopharyngodon idellus)补体c3的作用机理,用含0、0.06%、0.12%、0.24%盐酸小檗碱饲料投喂草鱼,在第7 d、14 d检测血清中补体c3含量,在第14 d检测肝脏中补体c3 mRNA表达量及急性感染嗜水气单胞菌(Aeromonas hydrophila)后存活情况。并进一步对盐酸小檗碱体内药代动力学和补体消耗试验进行了研究。结果显示;摄食盐酸小檗碱的草鱼血清中补体c3含量和肝脏补体c3 mRNA表达量均显著性增加(P0.05/P0.05),草鱼的存活率与对照组相比也有显著性提高(P0.01)。补体消耗试验结果显示,当盐酸小檗碱在血清中浓度小于5 mg/L时与对照组相比补体消耗不显著,当盐酸小檗碱浓度大于5 mg/L时,补体消耗达到显著性差异(P0.05),表明此时盐酸小檗碱与补体c3直接结合,激活补体。药代动力学实验发现,灌服盐酸小檗碱后,出现双峰现象,两峰值分别是0.243 mg/L和0.117 mg/L,此浓度远远低于与补体c3直接结合量最低量(5 mg/L)。结果表明,盐酸小檗碱对草鱼补体c3的作用机理可能是通过上调补体c3mRNA的表达增加补体c3数量,而非通过直接结合补体c3来改变其活性。  相似文献

An 8‐week feeding trial was conducted to evaluate the effects of ascorbic acid (AsA), in the form of l ‐ascorbyl‐2‐polyphosphate (LAPP) on growth performance, body composition, antioxidative capacity and salinity stress tolerance of juvenile Pacific white shrimp, Litopenaeus vannamei. Five practical diets (46% crude protein and 7.6% lipid) supplemented with graded levels of AsA (14.64, 48.55, 84.98, 308.36 and 639.27 mg kg?1 diet) were fed to five replicate groups of L. vannamei (mean initial wet weight 0.57 g). No significant differences were found on growth performance among all treatments. However, whole body lipid content significantly decreased with dietary AsA levels increasing. Activities of total antioxidant capacity, glutathione reductase and glutathione peroxidase were significantly affected by dietary AsA levels. Shrimp fed LAPP‐free diet had higher malondialdehyde content than those fed the diets supplemented with LAPP. Dietary AsA levels higher than 308.36 mg kg?1 diet increased the survival of shrimps after 1, 2 and 3 h of acute salinity change. Broken‐line regression analysis on survival after 3 h of salinity stress and second‐degree polynomial regression analysis on glutathione reductase data indicated that the optimal dietary AsA requirement of L. vannamei was estimated to be 306.39, 319.75 mg kg?1 diet respectively.  相似文献

We evaluated the effects of enriched rotifers on growth, survival and on the lipid composition of haddock larvae. The treatments tested were (1) AlgaMac 2000®, (2) AquaGrow® Advantage and (3) Pavlova sp. paste and AlgaMac 2000®. The treatments did not influence larval growth rate throughout the experimental period (P = 0.70). Larvae from all treatments grew approximately 8% of their dry weight per day between 1 and 29 days post hatch (dph). Treatment 3 resulted in the best survival, estimated to be 3 on a scale from 0 to 5, whereas for the two other groups the survival estimates were 0 and 2. Rotifers from treatment 1 had low sterol concentrations, high eicosapentaenoic acid/arachidonic acid ratio and their feeding resulted in high larval mortality. Rotifers enriched with Pavlova sp. had the lowest proportions of the sum of saturated fatty acids, docosahexaenoic acid and sum of ω3 and the highest proportions of the sum of monounsaturated fatty acids (ΣMUFA). This was partially reflected in larvae from treatment 3 in that they had the highest proportions of ΣMUFA and the lowest proportions of Σω3 (P < 0.0001 for both analyses). In addition, these larvae had the highest and lowest ΣC20 and ΣC22 polyunsaturated fatty acids (PUFA) respectively (P < 0.0001 for both analyses). We suggest that more research with ω3 and ω6 PUFA can lead to improvements in the rearing of haddock larvae produced in hatcheries.  相似文献

Effects of dietary l ‐carnitine were studied in juvenile black sea bream (Sparus macrocephalus). The semipurified basal diet [crude protein 450 g kg?1 dry matter (DM); crude lipid 126 g kg?1 DM] was formulated to choose white fishmeal as the protein source and fish oil plus corn oil (1 : 1) as the lipid source. Six diets (control + diets 1–5) containing 0.1, 0.12, 0.16, 0.24, 0.39 and 1.1 g of l ‐carnitine kg?1 diet were fed to triplicate groups of black sea bream (initial weight 13.10 ± 0.05 g) for 8 weeks. At the end of the feeding trial, growth performance, body composition and antioxidant status were determined. The results showed that relative growth rate (RGR) was significantly improved by the elevation of dietary l ‐carnitine level from 0.1 to 0.24 g kg?1, but decreased with further increment (P < 0.05). Lipid content decreased significantly (P < 0.05) in the dorsal muscle whereas increased (P < 0.05) in the liver with the addition of dietary l ‐carnitine. Dietary l ‐carnitine supplements elevated enzymatic antioxidants (superoxide dismutase, SOD; catalase, CAT; glutathione‐S‐transferase, GST) activities (P < 0.05) yet decreased the content of non‐enzymatic factor, total sulphydryl groups (TSH) (P < 0.05). In summary, the optimum dietary l ‐carnitine level was 0.284 g kg?1 diet by second‐polynomial regression analysis based on RGR (y = ?647.4x2 +367.97x + 234.55; R2 = 0.977, x = dietary l ‐carnitine levels, y = RGR), and dietary l ‐carnitine addition within the levels adopted in our study could depress lipid peroxidation in tissues of juvenile black sea bream.  相似文献

Abstract

The protein contents of the grain of 50 interspecific progenies developed from the cross between WAB56-104, an Oryza sativa variety, and CG 14, an Oryza glaberrima line, were investigated. In contrast to the higher protein content of O. glaberrima than O. sativa on the average, the protein content of CG 14 was always lower than that of WAB56-104. However, judging from the average of three seasons, 72% of the interspecific progenies had a higher protein content than the mid-parent and 50% of them had a higher protein content than WAB56-104. Although the actual values of protein content of the interspecific progenies were significantly different among the seasons, a highly significant correlation was always observed in protein content between any two of the three seasons. Protein content therefore was considered character of each interspecific progeny though it was also affected by environment. A significant correlation was not observed between paddy yield and protein content in any season; several interspecific progenies showed higher protein content and paddy yield than the mid-parents. A low paddy yield is likely to be associated with high protein content through physiological regulation without a genetic linkage between the two traits. However, the results suggest that the transgressive segregation of protein content observed in the interspecific progenies is attributed not to this physiological regulation but to a certain mechanism to concentrate protein in grains with a genetic background.  相似文献
