Starch-gel electrophoresis was used to resolve nine polymorphic enzyme loci from leaf tissue collected from 20 Korean populations ofEurya japonica in order to determine differences in allele frequencies between male and female trees. In addition, 84 adults were sampled and mapped in a population located on Naenaro Island in Korea to examine spatial genetic structure using spatial autocorrelation analysis. Allele frequencies between males and females gave few contribution to the genetic structuring within populations. Only nine (5%) of 180 cases were significantly different from both sexes in allele frequencies. On the other hand, Moran'sI was significantly different from the expected value in 31 (23.5%) of 132 cases. In the shortest distance (0<5m),I was significantly positive in 10 (22.7%) of 44 cases. The results indicate that a significant small scale genetic structure was detected in the population and patch widths were inferred to be approximately 5–7 m. A nonrandom distribution of genotypes may be indicative of restricted gene flowvia seed and pollen dispersal, and patchy establishments of genetically distinct individuals. These factors are responsible for shaping population genetic structure ofE. japonica.  相似文献

Summary The aim of the trial was to determine whether growth regulators and injuries could affect sex expression in the cultivated kiwifruit. Growth substances (NAA, BAP, GA, ethephon, ABA, TIBA, and CCC) were applied at 1% concentration in lanolin (w/w) to shoots of 17-year old plants of the staminate clone Matua and the pistillate clone Hayward. Injuries consisted in shoot defoliation or removal of a ring of phloematic tissue from the laterals. Both the application of growth regulators and injuries were done about 5 weeks before anthesis. In the pistillate clone, no treatment resulted in overcoming pollen grain sterility; ABA and GA mereased the stigma-style number as well as the density of papillae on the stigma surface. A positive correlation was found between stigma-style number and pedicel length (r=0.660). In the staminate clone, pollen grain viability was significantly altered by treatment; the highest (79.2%) and lowest (10.3%) values were recorded for ABA and GA, respectively. A negative correlation was found between pollen grain viability and pedicel length (r=–0.648). As a general rule, treatments which depressed pollen grain viability allowed the development of a variable number of styles. About 20 styles were consistently produced when GA was applied; the gibberellin-caused feminization of kiwifruit was confirmed in a repeated trial. The data collected also suggest that sex expression can be slightly changed by injuries; contrariwise, rather good results can be achieved by using a growth regulator earlier in the season or before flower initiation.  相似文献

【目的】研究蒿柳(Salix viminalis)雌雄株幼苗在生长季不同阶段的生长与防御差异,为雌雄株差异研究及其在生产实践中的选择性应用提供指导。【方法】以蒿柳1年生雌雄扦插苗为研究对象,在大田中采用完全随机区组设计,于6-10月(6月5日、7月3日、8月4日、9月7日和10月5日)对雌雄株幼苗的生长、光合作用参数、光合色素含量、化学防御物质含量以及防御酶活性等测定后进行动态分析,比较二者差异。【结果】蒿柳雌雄幼苗株高在6月、地径在6月和7月无显著差异,其他时间株高和地径均为雄株显著高于雌株。6月雌雄株间的净光合速率(Pn)和蒸腾速率(Tr)无显著差异,7-10月均表现为雄株显著高于雌株;6-9月雌雄株间气孔导度(Gs)无显著差异,仅在10月雄株显著高于雌株;6-10月整个生长季雌雄株间胞间CO2浓度(Ci)差异均不显著。雌雄株间叶绿素a(Chl a)、叶绿素b(Chl b)和类胡萝卜素(Car)含量在6月差异均不显著,7-10月各光合色素含量均为雄株显著高于雌株。雌雄株间多酚和缩合单宁含量在生长季均无显著差异,但雄株的SOD(6月除外)、POD、PPO和PAL(6月除外)活性均显著高于雌株。【结论】在有胁迫因素存在的自然环境中,蒿柳雌雄株幼苗防御物质含量无显著差异,但雌株防御酶活性较低、生长相对缓慢,可能在繁殖与生长、防御间存在进化的权衡。蒿柳雄株生长相对较快,且可解决飞絮问题,在实际应用中可优先选用。  相似文献

DNA from female and male hemp (Cannabis sativa L.) plants belonging to nine different varieties were screened with180 RAPD primers in a search for sex-associated DNA markers. About 1500bands were produced in total, nine primers were found yielding one or two DNA bands amplified in all nine male DNA bulks and absent in all female DNA bulks. These putatively male-associated markers were then scored in three different F1progenies, deriving from a cross between a common male parent and three different female plants. The sex of the progeny was accurately scored on the basis of the floral phenotype, and the presence of the nine male-associated markers was verified by RAPD analysis. In all three progenies examined, all the male plants showed the DNA markers previously identified by bulk segregant analysis (BSA) on the hemp varieties, while all the female plants lacked them. The fact that the association between these markers and the staminate phenotype is found when examining male plants of distantly related varieties, and that such linkage is never broken when different progenies are examined, strongly supports the hypothesis that the markers found are physically located on the Y chromosome, in a region excluded from recombination during meiosis. Another marker was shown to be present in the male parent, in all the male plants of each progeny, and in 50% of the female progenies, while it was absent in the female parent; the possible occurrence of markers deriving from multiple amplification sites of the genome is discussed. 【目的】以雌雄异株山鸡椒为研究材料,分析雌雄植株在生殖生长过程中水分及氮素利用策略的性别特异性动态变化规律,以期为雌雄异株植物资源分配动态变化机制提供理论依据。【方法】测定雌雄植株在开花后105~165天共5个时期叶片养分含量(碳含量、氮含量、碳氮比)、稳定碳同位素组成(δ13C)、稳定氮同位素组成(δ15N),并分析其动态变化规律。【结果】1)雌雄株叶片δ13C平均值分别为-29. 38‰和-28. 08‰。性别和发育时期对雌雄株叶片δ13C值影响均显著,5个时期雌株δ13C值均显著低于雄株;且随发育进程雌雄植株δ13C值均不断下降。2)雌雄株叶片δ15N平均值分别为1. 90‰和2. 95‰。性别和发育时期对雌雄株叶片δ15N值均有极显著影响,开花后105~150天(即雌株果实中精油及柠檬醛含量快速积累期)雌株δ15N值显著低于雄株;随着发育进程,雌雄株叶片δ15N值均表现出双峰变化趋势,双峰出现在开花后105天和135天,雌株叶片δ15N在开花后120天达到最低值,雄株叶片在开花后165天时达到最低值。3)雌雄株叶片碳含量平均值分别为49. 44%和49. 28%。开花后105天和120天,雌株叶片C含量高于雄株叶片C含量,开花后135~165天(果实精油快速积累期到稳定期),雌株叶片C含量低于雄株。雄株叶片C含量随发育进程显著升高,而雌株叶片C含量在不同时期无显著差异。4)雌雄株叶片N含量平均值分别为1. 71%和1. 51%。性别对N含量影响显著,雄株叶片N含量均低于雌株(除开花后135天)。5)雌雄株叶片C/N平均值分别为29. 15和33. 72。性别对叶片碳氮比影响显著,雌株C/N值在不同发育时期均小于雄株,但发育时期对其无显著影响;随着发育时期推进,雄株C/N值下降程度高于雌株。6)雌雄植株叶片δ13C与N含量以及δ13C与δ15N之间均无显著相关性。【结论】山鸡椒雌雄植株在养分含量、水分及氮素利用策略存在差异,且在开花后105~165天表现出动态变化规律。雌株水分利用效率低于雄株,从果实精油及柠檬醛含量快速积累期到稳定期,雌株水分利用效率不断下降;雌株氮利用低于雄株,且随着植株发育,雌雄株叶片氮素利用效率均表现出双峰变化趋势;雄株叶片氮含量低于雌株,分配更多的氮素至花芽以保证花粉形成;果实精油快速积累期到稳定期,雌株叶片碳含量低于雄株,为果实及种子的形成提供更多的碳元素。  相似文献

在栝楼生长发育各阶段,采用高效液相色谱技术分别对雌雄植株叶片内源激素及多胺含量进行分析。结果表明,吲哚乙酸(IAA)含量在所测定的生长发育期,雌株始终高于雄株,而脱落酸(ABA)含量雌株始终低于雄株;玉米素核苷(ZR)和赤霉素(GA1/3)含量在营养生长期雄株高于雌株,雄株进入生殖生长阶段后迅速下降,而雌株继续保持上升趋势,直至雌株进入生殖生长期后才迅速下降。雌株叶片中亚精胺(Spd)含量极显著高于雄株;精胺(Spm)含量在营养生长期雄株显著高于雌株,而在雌株进入生殖生长后雌雄株间差异不显著;腐胺(Put)含量在营养生长期雄株显著高于雌株,而在雌株进入生殖生长后雌株显著高于雄株。试验结果表明:高含量的IAA和Spd有利于栝楼雌花发育,而高含量的Put和Spm有利于雄花发育。当雌雄花盛开后,多胺均呈下降趋势,Put和Spm在雌雄株间含量差异越来越小。  相似文献

Summary Cannabis sativa L. is a dioecious species with sexual dimorphism occurring in a late stage of plant development. Sex is determined by heteromorphic chromosomes (X and Y): male is the heterogametic sex (XY) and female is the homogametic one (XX). The sexual phenotype of Cannabis often shows some flexibility leading to the differentiation of hermaphrodite flowers or bisexual inflorescences (monoecious phenotype). Sex is considered an important trait for hemp genetic improvement; therefore, the study of the mechanism of sexual differentiation is of paramount interest in hemp research. A morphological and molecular study of Cannabis sativa sexual differentiation has been carried out in the Italian dioecious cultivar Fibranova.Microscopic analysis of male and female apices revealed that their reproductive commitment may occur as soon as the leaves of the fourth node emerge; the genetic expression of male and female apices at this stage has been compared by cDNA-AFLP. A rapid method for the early sex discrimination has been developed, based on the PCR amplification of a male-specific SCAR marker directly from a tissue fragment.Five of the several cDNA-AFLP polymorphic fragments identified have been confirmed to be differentially expressed in male and female apices at the fourth node. Cloning and sequencing revealed that they belong to nine different mRNAs that were all induced in the female apices at this stage. Four out of them showed a high degree of similarity with known sequences: a putative permease, a SMT3-like protein, a putative kinesin and a RAC-GTP binding protein.  相似文献
