应用3种肿瘤细胞模型对未破壁灵芝孢子、机械破壁灵芝孢子和全灵芝剥壁孢子进行抗肿瘤试验研究。结果表明,未破壁及不同破壁方式的灵芝孢子对人恶性乳腺癌细胞(MT-1)、人恶性淋巴癌细胞株(Jurkat)、人恶性脑肿瘤细胞(U343)均有不同程度的抑制作用,抑制效果强弱为全灵芝剥壁孢子机械破壁灵芝孢子未破壁灵芝孢子,表明破壁的确能提高灵芝孢子对肿瘤细胞的抑制作用,而且选择破壁的方法也很重要,生物酶法破壁能使灵芝孢子的生物活性成分得到充分释放,所以大大提高灵芝孢子的功效。 相似文献

Choroid plexus papilloma (CPP) is a rare, benign, intracranial neoplasm. The present report described a choroid plexus papilloma of the fourth ventricle with unusual proliferative reaction of ependymal/subependymal cells in a stallion. A single, soft neoformation, red-gray in color, with an irregular surface, resembling a cauliflower, expanding from the fourth ventricle to the cerebellar peduncles was observed. Microscopically, the neoformation was composed of a single layer of simple cuboidal to columnar epithelium with a well-vascularized papillary-arboriform pattern. Multifocal subependymal aggregates forming rosettes were observed near the mesencephalic aqueduct (MA). Glial-fibrillary-acidic-protein (GFAP) was markedly positive only in the neuropil adjacent to the proliferative areas of the MA. Ependymal/subependymal reactions secondary to CPP have never been described in equine or human neuropathology. The capabilities of ependymal cells to regenerate and eventually proliferate are controversial and unclear and, thus, require additional investigation to understand the adaptive mechanisms of this rare pathogenic condition. 相似文献

A non-radiometric method for measuring serum thymidine kinase activity in malignant lymphoma in dogs
The aim of this study was to evaluate an enzyme-linked immunosorbent assay (ELISA), for determination of serum thymidine kinase 1 (sTK1) activity in dogs with malignant lymphoma (ML) and compare it with a thymidine kinase (TK) radioenzymatic assay (TK-REA). The TK-REA has recently been shown to be useful in determining the clinical stage and prognosis in canine ML. In addition, serum lactate dehydrogenase (LDH) was measured. Forty-five dogs were included in the study. Sixty serum samples from these dogs, stored in a tumour serum sample bank (stored at -20 degrees C), were analysed. Apart from 37 dogs with ML, four normal dogs as well as two dogs with mammary carcinomas, one dog with bladder carcinoma, and one dog with malignant fibrous histiocytoma were included. Staging of ML was based on the modified World Health Organization (WHO) staging system for canine ML. The diagnosis of all tumours was verified by histopathology. The TK activity (units per litre [U/L]) ranged from 1.0 to 607.9 in the TK-REA analysis and from 1.1 to 510 in the TK-ELISA (normal reference value <7U/L). The range for LDH was between 12 and 1194 U/L (normal reference value <228 U/L). There was a significant correlation between the TK-REA and the TK-ELISA. The correlation coefficient (CC) was 0.97 and the standard error of the estimate (SEE) was 3.7 U/L. There was no correlation between LDH and either the TK-REA or the TK-ELISA (CC=0.53 for both assays; SEE=26.7 and 12.7 U/L, respectively). Most of the variation in LDH was still within the normal reference range. The mean LDH in dogs with high-stage (stage IV+V) disease was 201.9 U/L. The corresponding values for the TK-REA and TK-ELISA were 109 and 109.9 U/L, respectively. The significant relation between the TK-REA and the TK-ELISA was confirmed by Bland-Altman analysis. The TK-ELISA assay, because of its relative simplicity, will permit measurement of TK in cases of ML in dogs to become a routine procedure. 相似文献

The angiopoietin (Ang) family of proteins are central to the regulation of angiogenesis. The purposes of this study were to determine cDNA sequences of canine Ang-1 and Ang-2 and investigate their expressions in normal tissues and spontaneous tumours. The cDNA sequences of canine Ang-1 and Ang-2 were 1,494 and 1,488 bp, and the deduced amino acid sequences were 497 and 495 residues, respectively. The cDNA sequences of canine Ang-1 and Ang-2 showed high homology with those of the other mammalian species. Canine Ang-1 and Ang-2 mRNA were detectable in all 22 normal tissues and spontaneous tumours. Higher mRNA expression level of canine Ang-2 was demonstrated in mammary simple carcinomas, haemangiosarcoma and hepatocellular carcinoma in comparison with normal tissues. 相似文献

Background
Seminomas in the dog have traditionally been assumed to resemble human spermatocytic seminomas, based on their low malignancy and high occurrence in old individuals. However, recently published studies indicate that canine seminomas can be classified as classical and spermatocytic seminomas in a similar way as in man, and that classical seminomas comprise a substantial proportion of seminomas in the dog. These two factors both contribute to increasing the potential of canine seminoma as a relevant model for human testicular cancer. The aim of the present study was to characterise seminoma in Norwegian dogs using morphology and immunohistochemistry, and determine whether these tumours are comparable with human classical seminoma.Methods
By applying diagnostic criteria from human pathology, 45 seminomas from the Norwegian Canine Cancer Register were examined histologically with hematoxylin and eosin (HE) and periodic acid-Schiff (PAS) stains. All sections were stained immunohistochemically with antibodies against human placental alkaline phosphatase (PLAP) and the transmembrane receptor c-KIT.Results
Although two of the seminomas showed immunohistochemical staining characteristics indicative of classical seminoma (PLAP+/c-KIT+), all 45 examined seminomas were morphologically consistent with spermatocytic seminoma.Conclusions
The value of canine seminoma as a model for SE in man remains unclear. Among the 45 investigated tumours from Norwegian dogs, none were classified as classical seminoma based on morphological criteria consistent with human seminomas. Regional or breed differences in the occurrence of classical seminoma in the dog, as well as the lack of uniform diagnostic criteria, might explain the discrepancy between the findings in the current study and the results presented by other authors. 相似文献7.

Summary Bovine blood containing piroplasms of Theileria parva, as well as non‐injected blood, was lysed and subjected to iso‐electric focussing. Staining for 13 different enzymes revealed parasite‐associated bands of glucose phosphate isomerase (GPI) activity, not of any of the other enzymes. There were no variations between individual donor animals in the host cell GPI bands and these bands did not interfere with the recognition of the parasite‐associated bands, so that purification of the piroplasms was unnecessary. Blood from cattle infected with T. mutans also gave parasite‐associated bands of GPI, but no such bands were seen in zymograms of blood from cattle infected with a Theileria sp. from Japan. Depending on the level of parasitaemia, up to four parasite‐associated bands were found in one strain of T. parva and up to three in two other strains. Among the disadvantages of using piroplasm material for the study of isoenzymes of T. parva is the fact that animals often die before their parasitaemia is sufficiently high, and that some strains never give rise to a high parasitaemia. 相似文献

Serpin peptidase inhibitor clade E member 2 (SerpinE2), a member of serine peptidase inhibitor super family, regulates multiple serine proteases. SerpinE2 regulates the protein of extracellular matrix by serine protease system and matrix metalloproteinase (MMP) system. 实验性脾虚证大鼠免疫细胞因子变化及四君子汤对其影响 总被引:9,自引:1,他引:9
本文分别通过大鼠肌肉注射利血平和口服小承气汤两种方法复制脾虚证动物模型,比较研究了脾虚证大鼠细胞因子白细胞介素-2(IL-2)、白细胞介素-6(IL-6)及肿瘤坏死因子(TNF)的含量变化,并用四君子汤治疗以反证.结果表明,不论利血平法复制脾虚证动物模型,还是小承气汤复制脾虚证动物模型,均表现为外周血IL-2、IL-6和TNF含量明显低于正常对照组与四君子汤治疗组(P<0.01),对照组与治疗组之间除利血平所致脾虚证动物模型用四君子汤治疗后TNF含量明显高于对照组(P<0.05)以外,以上其它指标均无明显差异(P>0.05).说明脾虚大鼠产生IL-2、IL-6和TNF等细胞因子能力下降,使免疫功能处于低下状态,四君子汤治疗能够提高脾虚证大鼠产生细胞因子的能力,从而达到健脾益气调整机体的免疫功能. 相似文献
